    Data-driven approach for modeling Reynolds stress tensor with invariance preservation

    The present study represents a data-driven turbulent model with Galilean invariance preservation based on machine learning algorithm. The fully connected neural network (FCNN) and tensor basis neural network (TBNN) [Ling et al. (2016)] are established. The models are trained based on five kinds of flow cases with Reynolds Averaged Navier-Stokes (RANS) and high-fidelity data. The mappings between two invariant sets, mean strain rate tensor and mean rotation rate tensor as well as additional consideration of invariants of turbulent kinetic energy gradients, and the Reynolds stress anisotropy tensor are trained. The prediction of the Reynolds stress anisotropy tensor is treated as user's defined RANS turbulent model with a modified turbulent kinetic energy transport equation. The results show that both FCNN and TBNN models can provide more accurate predictions of the anisotropy tensor and turbulent state in square duct flow and periodic flow cases compared to the RANS model. The machine learning based turbulent model with turbulent kinetic energy gradient related invariants can improve the prediction precision compared with only mean strain rate tensor and mean rotation rate tensor based models. The TBNN model is able to predict a better flow velocity profile compared with FCNN model due to a prior physical knowledge.Comment: 23 page

    Variations of Mid-Pacific Trough and Their Relations to the Asian-Pacific-North American Climate: Roles of Tropical Sea Surface Temperature and Arctic Sea Ice

    The mid-Pacific trough (MPT), occurring in the upper troposphere during boreal summer, acts as an atmospheric bridge connecting the climate variations over Asia, the Pacific, and North America. The first (second) mode of empirical orthogonal function analysis of the MPT, which accounts for 20.3 (13.4) percent of the total variance, reflects a change in its intensity on the southwestern (northeastern) portion of the trough. Both modes are significantly correlated with the variability of tropical Pacific sea surface temperature (SST). Moreover, the first mode is affected by Atlantic SST via planetary waves that originate from the North Atlantic and propagate eastward across the Eurasian continent, and the second mode is influenced by the Arctic sea ice near the Bering Strait by triggering an equatorward wave train over the Northeast Pacific. A stronger MPT shown in the first mode is significantly linked to drier and warmer conditions in the Yangtze Basin, southern Japan, and northern U.S. and a wetter condition in South Asia and northern China, while a stronger MPT shown in the second mode is associated with drier and warmer southwestern U.S. In addition, an intensified MPT (no matter in the southwestern or the northeastern portion) corresponds to more tropical cyclones (TCs) over the western North Pacific (WNP) and less TCs over the eastern Pacific (EP) in summer, which is associated with MPT-induced ascending and descending motions over the WNP and the EP, respectively

    Two-Period Inventory Control with Manufacturing and Remanufacturing under Return Compensation Policy

    As an effective way of decreasing production cost, remanufacturing has attracted more and more attention from firms. However, it also brings many difficulties to firms, especial when firms remanufacture products which they produce. A primary problem for the case is how to acquire the used product sold by the firm itself. In this paper, we consider a return compensation policy for acquiring used product from customers. Under this policy, the return quantity of used product is a proportion of demand. We study an inventory replenishment and production planning problem for a two-period inventory system with dependent return and demand. We formulate the problem into a three-stage stochastic programming problem, where the firm needs to make decisions on the replenishment quantity of new raw material inventory in each period and the production quantities of manufacturing and remanufacturing ways. We give the optimal production policy of manufacturing and remanufacturing ways for the realized demand and prove the objective function for each stage to be concave in the inventory replenishment quantity. Moreover, we prove that the basic inventory policy is still optimal for each period and give the analytical conditions of the optimal inventory levels which are unrelated to acquisition price.     Based on the model of emotion as social information, this study explores the effects of facial emotions and trustworthy behavior on trust decision-making in trust game through two experiments. The present study used trust game explores the impact of players’ facial emotion, arousal and trustworthy behavior on individual trust decision-making through two experiments. The results can be summed up as follows: (1) in the repeated interaction with four players, individuals invest more in trustworthy players than untrustworthy players; (2) individuals invested more in trustworthy players with happy facial emotions, while untrustworthy players with angry facial emotions received less investment. High-arousal facial emotion results in a more extreme investment by the individual; (3) when the players’ facial emotion and behavior are inconsistent, the individual will judge according to the player’s current behavior, rather than the facial emotion or past behavior

    Enhanced oral bioavailability of cyclosporine A by liposomes containing a bile salt

    The main purpose of this study was to evaluate liposomes containing a bile salt, sodium deoxycholate (SDC), as oral drug delivery systems to enhance the oral bioavailability of the poorly water-soluble and poorly permeable drug, cyclosporine A (CyA). Liposomes composed of soybean phosphatidylcholine (SPC) and SDC were prepared by a thin-film dispersion method followed by homogenization. Several properties of the liposomes including particle size, polydispersity index, and entrapment efficiency were characterized. The in vitro release of CyA from these liposomes was less than 5% at 12 hours as measured by a dynamic dialysis method. The pharmacokinetic results in rats showed improved absorption of CyA in SPC/SDC liposomes, compared with CyA-loaded conventional SPC/cholesterol (Chol) liposomes and microemulsion-based Sandimmune Neoral®. The relative oral bioavailability of CyA-loaded SPC/SDC and SPC/Chol liposomes was 120.3% and 98.6%, respectively, with Sandimmun Neoral as the reference. The enhanced bioavailability of CyA was probably due to facilitated absorption by the liposomes containing SDC rather than improved release rate

    Grouping, spectrum-effect relationship and antioxidant compounds of Chinese propolis from different regions using multivariate analyses and off-line anti-DPPH assay

    49 samples of propolis from different regions in China were collected and analyzed for their chemical compositions, contents of total flavonoids (TFC), total phenolic acid (TPC) and antioxidant activity. High-performance liquid chromatography (HPLC) analysis identified 15 common components, including key marker compounds pinocembrin, 3-O-acetylpinobanksin, galangin, chrysin, benzyl p-coumarate, pinobanksin and caffeic acid phenethyl ester (CAPE). Cluster analysis (CA) and correlation coefficients (CC) analysis showed that these propolis could be divided into three distinct groups. Principal component analysis (PCA) and multiple linear regression analysis (MLRA) revealed that the contents of isoferulic acid, caffeic acid, CAPE, 3,4-dimethoxycinnamic acid, chrysin and apigenin are closely related to the antioxidant properties of propolis. In addition, eight peak areas decreased after reacting with 1,1-Diphenyl-2-picrylhydrazyl (DPPH) radicals, indicating that these compounds have antioxidant activity. The results indicate that the grouping and spectrum-effect relationship of Chinese propolis are related to their chemical compositions, and several compounds may serve as a better marker for the antioxidant activity of Chinese propolis than TFC and TPC. The findings may help to develop better methods to evaluate the quality of propolis from different geographic origins

    Up-regulation of MiR-205 under hypoxia promotes epithelial-mesenchymal transition by targeting ASPP2

    The epithelial–mesenchymal transition (EMT) is one of the crucial procedures for cancer invasion and distal metastasis. Despite undergoing intensive studies, the mechanisms underlying EMT remain to be completely elucidated. Here, we identified that apoptosis-stimulating protein of p53-2 (ASPP2) is a novel target of MiR-205 in various cancers. Interestingly, the binding site of MiR-205 at the 3′-untranslated region of ASPP2 was highly conserved among different species. An inverse correlation between MiR-205 and ASPP2 was further observed in vivo in cervical cancers, suggesting MiR-205 may be an important physiological inhibitor of ASPP2. Hypoxia is a hallmark of solid tumor microenvironment and one of such conditions to induce EMT. Notably, MiR-205 was remarkably induced by hypoxia in cervical and lung cancer cells. A marked suppression of ASPP2 was observed simultaneously. Further studies confirmed that hypoxia-induced ASPP2 suppression was mainly attributed to the elevated MiR-205. Interestingly, the alteration of MiR-205/ASPP2 under hypoxia was accompanied with the decreased epithelial marker E-cadherin and increased mesenchymal marker Vimentin, as well as a morphological transition from the typical cobblestone-like appearance to the mesenchymal-like structure. More importantly, MiR-205 mimics or ASPP2 silencing similarly promoted EMT process. By contrast, ASPP2 recovery or MiR-205 inhibitor reversed MiR-205-dependent EMT. Further studies demonstrated that the newly revealed MiR-205/ASPP2 axis promoted cell migration and also increased cell proliferation both in vivo and in vitro. These data together implicated a critical impact of MiR-205/ASPP2 on promoting EMT. MiR-205/ASPP2 may be potential diagnostic and therapeutic biomarkers in cervical and lung cancers

    A novel epigenetic AML1-ETO/THAP10/miR-383 mini-circuitry contributes to t(8;21) leukaemogenesis

    DNA methylation patterns are frequently deregulated in t(8;21) acute myeloid leukaemia (AML), but little is known of the mechanisms by which specific gene sets become aberrantly methylated. Here, we found that the promoter DNA methylation signature of t(8;21)(+) AML blasts differs from that of t(8;21)(-) AMLs. This study demonstrated that a novel hypermethylated zinc finger-containing protein, THAP10, is a target gene and can be epigenetically suppressed by AML1-ETO at the transcriptional level in t(8;21) AML. Our findings also show that THAP10 is a bona fide target of miR-383 that can be epigenetically activated by the AML1-ETO recruiting co-activator p300. In this study, we demonstrated that epigenetic suppression of THAP10 is the mechanistic link between AML1-ETO fusion proteins and tyrosine kinase cascades. In addition, we showed that THAP10 is a nuclear protein that inhibits myeloid proliferation and promotes differentiation both in vitro and in vivo Altogether, our results revealed an unexpected and important epigenetic mini-circuit of AML1-ETO/THAP10/miR-383 in t(8;21) AML, in which epigenetic suppression of THAP10 predicts a poor clinical outcome and represents a novel therapeutic target

    The effects of (+)-Gossypol on 11β-HSD and the concentration of corticosterone and dehydrocorticosterone in mice serum and tissues

    11β-hydroxysteroid dehydrogenase (11β-HSD) plays an important part in mediating glucocorticoid action, catalyzing the interconversion of corticosterone (B) and dehydrocorticosterone (A) in rodents. The aim of our study is to investigate the effects of (+)-gossypol (G+) on 11β-HSD. Adult ICR mice were given B and B + (G+) by intraperitoneal injection. The activity of 11β-HSD was evaluated by measuring the ratio of A and B, meanwhile the effects of (+)-gossypol on the conversion rate of B to A was determined with HPLC. Serum A/B levels of the B+(G+) group decreased by 2.42, 7.32, 17.85, 31.39, and 40.02 % compared to the B group at each measured time interval. A/B levels at 1 h for the B + (G+) group decreased by 43.78, 21.29 and 34.47% in liver, kidney and adrenal glands, respectively, in comparison to the B group. However, (+)-gossypol had no effect on brain and testis. (+)-Gossypol was an inhibitor of 11β-HSD.Colegio de Farmacéuticos de la Provincia de Buenos Aire
