    Novi VP2/VP3 rekombinantni senekavirus A izoliran u sjevernoj Kini

    Senecavirus A (SVA), previously called the Seneca Valley virus, is the only member of the genus Senecavirus within the family Picornaviridae. This virus was discovered as a serendipitous finding in 2002 and named Seneca Valley virus 001 (SVV-001). SVA is an emerging pathogen that can cause vesicular lesions and epidemic transient neonatal a sharp decline in swine. In this study, an SVA strain was isolated from a pig herd in Shandong Province in China and identified as SVA-CH-SDFX-2022. The full-length genome was 7282 nucleotides (nt) in length and contained a single open reading frame (ORF), excluding the poly (A) tails of the SVA isolates. Phylogenetic analysis showed that the isolate shares its genomic organization, resembling and sharing high nucleotide identities of 90.5% to 99.6%, with other previously reported SVA isolates. The strain was proved by in vitro characterization and the results demonstrate that the virus has robust growth ability in vitro. The recombination event of the SVA-CH-SDFX-2022 isolate was found and occurred between nts 1836 and 2710, which included the region of the VP2 (partial), and VP3 (partial) genes. It shows the importance of faster vaccine development and a better understanding of virus infection and spread because of increased infection rates and huge economic losses. This novel incursion has substantial implications for the regional control of vesicular transboundary diseases, and will be available for further study of the epidemiology of porcine SVA. U ovom je istraživanju soj SVA izoliran u populaciji svinja iz provincije Shandong u Kini i identificiran kao SVA-CHSDFX-2022. Kompletni genom izolata SVA imao je 7282 nukleotida (nt) u dužini i sadržavao je jedan otvoreni okvir za očitavanje (ORF), bez poli-A repova. Filogenetska je analiza pokazala da izolat u velikoj mjeri sadržava genomsku organizaciju i nukleotidne identitete, od 90,5 % do 99,6 %, s drugim poznatim SVA izolatima. Karakterizacija virusa je pokazala da ima veliku sposobnost rasta in vitro. Pronađena je rekombinacija izolata SVA-CH-SDFX-između nukleotida 1836 i 2710 što je uključilo regiju gena VP2 (parcijalno) i gena VP3 (parcijalno). Zbog visoke stope infektivnosti i golemih ekonomskih gubitaka važan je brži razvoj cjepiva i bolje razumijevanje zaraze. Rezultati ovog istraživanja pružaju korisne podatke za proučavanje SVA virusa, posebno s obzirom na njegovu epidemiologiju u svinja i regionalnu prekograničnu kontrolu vezikularnih bolesti

    Analiza genskih varijacija rekombinantnog soja dobivenog iz triju linija virusa-2 reproduktivnog i respiratornog sindroma svinja

    Since the rise of the porcine reproductive and respiratory syndrome virus (PRRSV) in China, gene mutations have frequently occurred. To understand the current prevalence and evolution of PRRSV in Shandong Province, 1,528 samples suspected of PRRSV were collected from local pig farms of different sizes. The complete genome sequence of the PRRSV strain SDLY-27 was determined by next-generation sequencing (NGS) technology. The genomic sequence of SDLY-27 was 15,363 nucleotides (nt) in length, comparative analysis of the whole genome sequence suggested that the homology between SDLY 27 and 81 PRRSV strains from China and other countries in genbank was 61.9 ~ 96.4%. This study is the first to detect recombinants from multiple recombination events among the Lineage 8 (JXA1-like strains), Lineage 5 (RespPRRSV-MLV and VR2332 strains) and Sublineage 1.5 (NADC34-like strains) in Shandong, China, and provides new data for the epidemiological study of PRRSV in China. This study enriches the epidemiological data on PRRSV in Shandong Province, China. It provides an important reference for the development of new vaccines and for the prevention and control of PRRSV in China.Usporedno sa širenjem virusa reproduktivnog i respiratornog sindroma svinja (PRRSV) u Kini, sve su češće bile i njegove genske mutacije. Kako bi se ustanovila trenutačna prevalencija i evolucija PRRSV-a u pokrajini Shandong, s lokalnih farmi prikupljeno je 1528 uzoraka svinja različitih kategorija za koje je postojala sumnja na zarazu PRRSVom. Kompletan genomski slijed soja SDLY-27 PRRSV-a određen je tehnologijom sekvenciranja sljedeće generacije (NGS). Slijed je imao dužinu od 15 363 nukleotida (nt), a komparativna analiza cijeloga genomskog slijeda uputila je na to da je homolognost između sojeva SDLY 27 i 81 PRRSV-a iz Kine i uzoraka u banci gena iz drugih zemalja 61,9~96,4%. Ovo je prvo istraživanje koje je otkrilo rekombinantne sojeve iz višestrukih rekombinacija među linijama 8 (sojevi nalik na JXA1), 5 (sojevi RespPRRSV-MLV i VR2332) i podlinije 1,5 (sojevi nalik na NADC34) u Shandongu, Kina.Kao takvo, istraživanje pruža nove podatke o epidemiologiji PRRSV-a u Kini, posebno u pokrajini Shandong, a ujedno predstavlja i važnu referenciju za razvoj novih cjepiva te prevenciju i kontrolu bolesti uzrokovane navedenim virusom

    Corn straw-saccharification fiber improved the reproductive performance of sows in the late gestation and lactation via lipid metabolism

    With the development of animal husbandry, the shortage of animal feedstuffs has become serious. Dietary fiber plays a crucial role in regulating animal health and production performance. The aim of this study was to investigate the effects of three kinds of corn straw-saccharification fibers (CSSF) such as high-fiber and low-saccharification (HFLS), medium-fiber and medium-saccharification (MFMS), low-fiber and high-saccharification (LFHS) CSSF on the reproductive performance of sows. Thirty-two primiparous Yorkshire sows were randomly assigned to 4 groups, 8 sows for each group. Group A was the basal diet as the control group; groups B – D were added with 6% HFLSCSSF, 6% MFMSCSSF and 6% LFHSCSSF to replace some parts of corn meal and wheat bran in the basal diet, respectively. The experimental period was from day 85 of gestation to the end of lactation (day 25 post-farrowing). The results showed that 6% LFHSCSSF addition significantly increased number of total born (alive) piglets, litter weight at birth (p < 0.05), whereas three kinds of CSSF significantly decreased backfat thickness of sows during gestation (p < 0.001), compared with the control group. Furthermore, CSSF improved the digestibility of crude protein, ether extract and fiber for sows. In addition, the levels of total cholesterol, total triglycerides, and high-density lipoprotein cholesterol in serum of sows were decreased by different kinds of CSSF. Further analysis revealed that CSSF regulated lipid metabolism through adjusting the serum metabolites such as 4-pyridoxic acid, phosphatidyl cholines and L-tyrosine. In summary, CSSF addition to the diets of sows during late gestation and lactation regulated lipid metabolism and improved reproductive performance of sows. This study provided a theoretical basis for the application of corn straw in sow diets

    Heterogeneous network embedding enabling accurate disease association predictions.

    BackgroundIt is significant to identificate complex biological mechanisms of various diseases in biomedical research. Recently, the growing generation of tremendous amount of data in genomics, epigenomics, metagenomics, proteomics, metabolomics, nutriomics, etc., has resulted in the rise of systematic biological means of exploring complex diseases. However, the disparity between the production of the multiple data and our capability of analyzing data has been broaden gradually. Furthermore, we observe that networks can represent many of the above-mentioned data, and founded on the vector representations learned by network embedding methods, entities which are in close proximity but at present do not actually possess direct links are very likely to be related, therefore they are promising candidate subjects for biological investigation.ResultsWe incorporate six public biological databases to construct a heterogeneous biological network containing three categories of entities (i.e., genes, diseases, miRNAs) and multiple types of edges (i.e., the known relationships). To tackle the inherent heterogeneity, we develop a heterogeneous network embedding model for mapping the network into a low dimensional vector space in which the relationships between entities are preserved well. And in order to assess the effectiveness of our method, we conduct gene-disease as well as miRNA-disease associations predictions, results of which show the superiority of our novel method over several state-of-the-arts. Furthermore, many associations predicted by our method are verified in the latest real-world dataset.ConclusionsWe propose a novel heterogeneous network embedding method which can adequately take advantage of the abundant contextual information and structures of heterogeneous network. Moreover, we illustrate the performance of the proposed method on directing studies in biology, which can assist in identifying new hypotheses in biological investigation

However, the morphology of these ETLs is easily altered by heating due to the relatively low glass transition temperature (Tg) of BPhen and this change often reduces the performance of OLEDs. Here, an enhancement in the thermal stability of OLEDs when doping their BPhen‐based ETLs with cesium (Cs) is reported. To investigate the role of the Cs dopant in the BPhen matrix, the crystallization features of Cs‐doped BPhen films with different doping concentrations are examined. Next, the electrical and optical properties of blue fluorescent and red phosphorescent OLEDs containing Cs‐doped BPhen ETLs are characterized after annealing the OLEDs at temperatures up to 100 °C. Cs plays a critical role in inhibiting the undesired crystallization of BPhen films, which enhances the thermal stability of OLEDs beyond the Tg of neat BPhen. Finally, highly stable BPhen‐based OLEDs encapsulated via atomic layer deposition at 80 °C are demonstrated. This work may lead to a new strategy for enhancing the intrinsic thermal durability of organic devices and their compatibility with thermally demanding processes.PostprintPeer reviewe

    Genomic analysis of oesophageal squamous-cell carcinoma identifies alcohol drinking-related mutation signature and genomic alterations

    Approximately half of the world's 500,000 new oesophageal squamous-cell carcinoma (ESCC) cases each year occur in China. Here, we show whole-genome sequencing of DNA and RNA in 94 Chinese individuals with ESCC. We identify six mutational signatures (E1–E6), and Signature E4 is unique in ESCC linked to alcohol intake and genetic variants in alcohol-metabolizing enzymes. We discover significantly recurrent mutations in 20 protein-coding genes, 4 long non-coding RNAs and 10 untranslational regions. Functional analyses show six genes that have recurrent copy-number variants in three squamous-cell carcinomas (oesophageal, head and neck and lung) significantly promote cancer cell proliferation, migration and invasion. The most frequently affected genes by structural variation are LRP1B and TTC28. The aberrant cell cycle and PI3K-AKT pathways seem critical in ESCC. These results establish a comprehensive genomic landscape of ESCC and provide potential targets for precision treatment and prevention of the cancer

    Adiponectin protects against myocardial ischemia–reperfusion injury: a systematic review and meta-analysis of preclinical animal studies

    Abstract Background Myocardial ischemia–reperfusion injury (MIRI) is widespread in the treatment of ischemic heart disease, and its treatment options are currently limited. Adiponectin (APN) is an adipocytokine with cardioprotective properties; however, the mechanisms of APN in MIRI are unclear. Therefore, based on preclinical (animal model) evidence, the cardioprotective effects of APN and the underlying mechanisms were explored. Methods The literature was searched for the protective effect of APN on MIRI in six databases until 16 November 2023, and data were extracted according to selection criteria. The outcomes were the size of the myocardial necrosis area and hemodynamics. Markers of oxidation, apoptosis, and inflammation were secondary outcome indicators. The quality evaluation was performed using the animal study evaluation scale recommended by the Systematic Review Center for Laboratory animal Experimentation statement. Stata/MP 14.0 software was used for the summary analysis. Results In total, 20 papers with 426 animals were included in this study. The pooled analysis revealed that APN significantly reduced myocardial infarct size [weighted mean difference (WMD) = 16.67 (95% confidence interval (CI) = 13.18 to 20.16, P < 0.001)] and improved hemodynamics compared to the MIRI group [Left ventricular end-diastolic pressure: WMD = 5.96 (95% CI = 4.23 to 7.70, P < 0.001); + dP/dtmax: WMD = 1393.59 (95% CI = 972.57 to 1814.60, P < 0.001); -dP/dtmax: WMD = 850.06 (95% CI = 541.22 to 1158.90, P < 0.001); Left ventricular ejection fraction: WMD = 9.96 (95% CI = 7.29 to 12.63, P < 0.001)]. Apoptosis indicators [caspase-3: standardized mean difference (SMD) = 3.86 (95% CI = 2.97 to 4.76, P < 0.001); TUNEL-positive cells: WMD = 13.10 (95% CI = 8.15 to 18.05, P < 0.001)], inflammatory factor levels [TNF-α: SMD = 4.23 (95% CI = 2.48 to 5.98, P < 0.001)], oxidative stress indicators [Superoxide production: SMD = 4.53 (95% CI = 2.39 to 6.67, P < 0.001)], and lactate dehydrogenase levels [SMD = 2.82 (95% CI = 1.60 to 4.04, P < 0.001)] were significantly reduced. However, the superoxide dismutase content was significantly increased [SMD = 1.91 (95% CI = 1.17 to 2.65, P < 0.001)]. Conclusion APN protects against MIRI via anti-inflammatory, antiapoptotic, and antioxidant effects, and this effect is achieved by activating different signaling pathways

    Integrated Transcriptomic and Metabolomic Analysis of the Mechanism of Foliar Application of Hormone-Type Growth Regulator in the Improvement of Grape (<i>Vitis vinifera</i> L.) Coloration in Saline-Alkaline Soil

    (1) Background: To solve the problems of incomplete coloration and quality decline caused by unreasonable use of regulators in grapes, this study clarified the differences in the effects of a hormone-type growth regulator (AUT) and two commercial regulators on grape coloration and quality through field experiments. (2) Methods: The color indexes (brightness (L*), red/green color difference (a*), yellow/blue color difference (b*), and color index for red grapes (CIRG)) of grape fruit were measured using a CR-400 handheld color difference meter. The titratable acid content, total phenol content, and total sugar content were measured using anthrone colorimetry, folinol colorimetry, and NaOH titration, respectively, and the chalcone isomerase activity, phenylalanine ammoniase activity, dihydroflavol reductase activity, and anthocyanin content were measured using a UV spectrophotometer. (3) Results: The a*, total sugar and total phenol contents, and chalcone isomerase (CHI) and phenylalanine ammoniase (PAL) activities of grape fruit in the AUT treatment significantly increased, while the titratable acid content significantly decreased, compared to those in the CK treatment. The expressions of the differentially expressed genes (DEGs) trpB and argJ in AUT treatment were significantly up-regulated. The expressions of the differentially expressed metabolites (DEMs) phenylalanine and 4-oxoproline were significantly up-regulated, while those of 3,4-dihydroxybenzaldehyde and N-acetyl glutamate were significantly down-regulated. The CIRG significantly increased by 36.4% compared to that in the CK, indicating improved fruit coloration. (4) Conclusion: The AUT could shorten the color conversion period of grape fruit and improve the coloration, taste, and tolerance to saline and alkaline stresses

    Effects of saccharified corn straw on growth performance, nutrient metabolic rates, gastrointestinal tract development and serum antioxidant index of broilers

    This study was conducted to investigate effects of saccharified corn straw (SCS) on broiler growth. 250 one-day-old female Arbor Acres (AA) broilers were distributed into 5 groups. The broilers in groups 1-5 were fed with diets containing 0 (basal diet), 4%, 8%, 12% SCS and 8% SCS (high oil), respectively. The experiment lasted 42 d including early and later stages. In early stage, average daily gain (ADG) and feed conversion ratio (FCR) in groups 2 and 5 were almost the same as control group; higher levels of SCS additions would decrease ADG and increase FCR (PThe accepted manuscript in pdf format is listed with the files at the bottom of this page.     Decreased integration of the frontoparietal network during a working memory task in major depressive disorder

    Background: Working memory deficits are a common feature in major depressive disorder and are associated with poor functional outcomes. Intact working memory performance requires the recruitment of large-scale brain networks. However, it is unknown how the disrupted recruitment of distributed regions belonging to these large-scale networks at the whole-brain level brings about working memory impairment seen in major depressive disorder. Methods: We used graph theory to examine the functional connectomic metrics (local and global efficiency) at the whole-brain and large-scale network levels in 38 patients with major depressive disorder and 41 healthy controls during a working memory task. Altered connectomic metrics were studied in a moderation model relating to clinical symptoms and working memory accuracy in patients, and a machine learning method was employed to assess whether these metrics carry enough illness-specific information to discriminate patients from controls. Results: Global efficiency of the frontoparietal network was reduced in major depressive disorder (false discovery rate corrected, p = 0.014); this reduction predicted worse working memory performance in patients with less severe illness burden indexed by Brief Psychiatric Rating Scale (β =–0.43, p = 0.035, t =–2.2, 95% confidence interval = [–0.043,–0.002]). We achieved a classification accuracy and area under the curve of 73.42% and 0.734, respectively, to discriminate patients from controls based on connectomic metrics, and the global efficiency of the frontoparietal network contributed most to the diagnostic classification. Conclusions: We report a putative mechanistic link between the global efficiency of the frontoparietal network and impaired n-back performance in major depressive disorder. This relationship is more pronounced at lower levels of symptom burden, indicating the possibility of multiple pathways to cognitive deficits in severe major depressive disorder
