    Atorvastatin reduces calcification in valve interstitial cells <i>via</i> the NF-ÎşB signalling pathway by promoting Atg5-mediated autophagy

    Aortic valve calcification (AVC) is a common cardiovascular disease and a risk factor for sudden death. However, the potential mechanisms and effective therapeutic drugs need to be explored. Atorvastatin is a statin that can effectively prevent cardiovascular events by lowering cholesterol levels. However, whether atorvastatin can inhibit AVC by reducing low-density lipoprotein (LDL) and its possible mechanism of action require further exploration. In the current study, we constructed an in vitro AVC model by inducing calcification of the valve interstitial cells. We found that atorvastatin significantly inhibited osteogenic differentiation, reduced the deposition of calcium nodules in valve interstitial cells, and enhanced autophagy in calcified valve interstitial cells, manifested by increased expression levels of the autophagy proteins Atg5 and LC3B-II/I and the formation of smooth autophagic flow. Atorvastatin inhibited the NF-ÎşB signalling pathway and the expression of inflammatory factors mediated by NF-ÎşB in calcified valve interstitial cells. The activation of the NF-ÎşB signalling pathway led to the reversal of atorvastatin's effect on enhancing autophagy and alleviating valve interstitial cell calcification. In conclusion, atorvastatin inhibited the NF-ÎşB signalling pathway by upregulating autophagy, thereby alleviating valve interstitial cell calcification, which was conducive to improving AVC

    Use of Shared-Mobility Services to Accomplish Emergency Evacuation in Urban Areas via Reduction in Intermediate Trips—Case Study in Xi’an, China

    Under no-notice evacuation scenarios with limited time horizons, the effectiveness of evacuation can be negatively impacted by intermediate trips that are made by family members and the identification of vulnerable populations. The emergence of shared-mobility companies, such as Uber and DiDi, can be considered as a potential means to address above-mentioned concerns. The proposed study explores the utility of shared-mobility services under emergency-evacuation scenarios and makes recommendations to relevant bodies that are based on the obtained and they are discussed herein. The study investigates attitudes of the public, experts, and drivers towards the use of shared-mobility resources during emergency evacuations based on a stated preference survey. Results of questionnaires, driver interviews, and face-to-face expert interviews have been analyzed to validate the feasibility and identify potential problems of leveraging shared-mobility services during evacuation response, especially in metropolitan areas wherein such services are already ubiquitous. Numerical simulations have been performed to quantify potential improvements in the total trip distance and number of evacuees after incorporating the use of shared mobility into emergency-response operations. However, despite the observed improvement in emergency efficiency, certain realistic roadblocks must be overcome. Realization of the proposed objective heavily depends on actionable policy recommendations, provided herein as a reference for the government, emergency management agencies, and shared-mobility companies.     Effect and Action Mechanism of Magnetic Field Treatment on the Fermentation Characteristics of Dough

    Magnetic fields can improve the quality of foods by influencing the arrangement of water molecules, thus altering the distribution of water in foods. In this study, dough was fermented at magnetic field strengths of 0, 0.5, 1.0, 1.5, and 2.0 mT for 1 h. The effect of magnetic field treatment on the fermentation characteristics, moisture distribution, and protein molecular structure of dough was investigated by low-field nuclear magnetic resonance (NMR) and in situ Raman spectroscopy, with a view to providing a theoretical basis for the development of high-quality Chinese dough-based foods. The results showed that the fermentability of dough tended to increase first and then decrease as the magnetic field strength increased, and the most significant effect on the fermentability was observed at a magnetic field strength of 1.5 mT. The specific volume of steamed bread initially increased and then decreased. When the magnetic field strength was 1.0 mT, the specific volume was 2.24 mL/g. The hardness, stickiness, and chewiness showed a trend of initially decreasing and then increasing, but there was no obvious change in the elasticity, cohesiveness or resilience. The contents of strongly bound water and free water initially decreased and then increased, whereas the opposite trend was observed for weakly bound water. The effect of 1.0 mT magnetic field strength on the water distribution was most significant. The relative content of α-helix increased first and then declined, while the reverse trend was observed for random coil. At a magnetic field strength of 0.5 mT, the relative content of α-helix was the highest (54%) and the relative content of random coil was lowest (21%). Proper magnetic field treatment could increase the fermentability of dough, the specific volume and textural properties of steamed bread, accelerate the water distribution in dough and improve the stability of the gluten network. In conclusion, magnetic field treatment could improve the fermentability of dough and the textural properties of steamed bread by improving the distribution of water, increasing the moisture uniformity, and changing the molecular structure of gluten proteins in dough

    Group membership and adolescents’ third-party punishment: a moderated chain mediation model

    Third-party punishment (TPP) reflects people’s social preference for fairness norms and is fundamental to maintaining fairness norms on a large scale. Several empirical studies have shown that the offender’s group membership impacts TPP, but the detailed mechanisms have yet to be fully elucidated. The current study used the third-party punishment game task to explore the relationship between group membership, perceived unfairness, anger, and adolescents’ TPP. A total of 306 teenagers aged 12 to 15 were chosen as subjects through cluster sampling. The results showed that group membership (classmate vs. stranger) and gender can affect adolescents’ TPP together, which manifests as adolescents enacting significantly harsher punishments on strangers than on classmates, especially for boys. Group membership indirectly affects TPP through the mediating effects of perceived unfairness, anger and through a chain mediation of perceived unfairness and anger. Moreover, gender positively moderate the relationship between group membership and perceived unfairness. Specifically, group membership significantly affects boys’ perceived unfairness, but cannot predict girls’ perceived unfairness. The above results can be used to guide adolescents toward appropriate justice concepts and moral awareness, thus enhancing TPP

    Dose health education on dementia prevention have more effects on community residents when a community physician/nurse leads it? A cross-sectional study

    BackgroundDementia is a growing public health concern worldwide. Community residents still have limited knowledge about dementia prevention, although many sources are accessible for individuals to acquire knowledge.MethodsA questionnaire-based survey was conducted in five communities in Chongqing, China, between March 2021 and February 2022. Participants were divided into three groups according to the dementia-related education they received: physician/nurse-led, mass media, and no relevant education. Covariance analysis was performed to determine the differences among the three groups in knowledge, motivation, and lifestyle, with the covariate of MoCA scores (education-adjusted).ResultsOf the 221 participants, 18 (8.1%) received physician/nurse-led education, 101 (45.7%) received only mass media education, and 102 (46.2%) did not receive any relevant education regarding dementia prevention. Participants who only received mass media education had a higher level of education (t = 5.567, p = 0.004) and cognitive function (t = 13.978, p &lt; 0.001). The analysis of covariance showed that compared with participants who received no relevant education, those who received physician/nurse-led education had higher levels of knowledge, perceived benefits, and better lifestyle, and those who received mass media education had lower perceived barriers; however, higher levels of cues to action, general health motivation, self-efficacy, and lifestyle (all p &lt; 0.05).ConclusionThe popularization of dementia-related education was not ideal for communities. Physician/nurse-led education plays a vital role in providing knowledge and promoting lifestyles for dementia prevention, but may not motivate community residents. Mass media education may help encourage residents and promote their lifestyles

    Increased both cortical activation and functional connectivity after transcranial direct current stimulation in patients with post-stroke: A functional near-infrared spectroscopy study

    BackgroundPrevious studies have shown that cognitive impairment is common after stroke. Transcranial direct current stimulation (tDCS) is a promising tool for rehabilitating cognitive impairment. This study aimed to investigate the effects of tDCS on the rehabilitation of cognitive impairment in patients with stroke.MethodsTwenty-two mild–moderate post-stroke patients with cognitive impairments were treated with 14 tDCS sessions. A total of 14 healthy individuals were included in the control group. Cognitive function was assessed using the Mini-Mental State Examination (MMSE) and the Montreal Cognitive Assessment (MoCA). Cortical activation was assessed using functional near-infrared spectroscopy (fNIRS) during the verbal fluency task (VFT).ResultsThe cognitive function of patients with stroke, as assessed by the MMSE and MoCA scores, was lower than that of healthy individuals but improved after tDCS. The cortical activation of patients with stroke was lower than that of healthy individuals in the left superior temporal cortex (lSTC), right superior temporal cortex (rSTC), right dorsolateral prefrontal cortex (rDLPFC), right ventrolateral prefrontal cortex (rVLPFC), and left ventrolateral prefrontal cortex (lVLPFC) cortical regions. Cortical activation increased in the lSTC cortex after tDCS. The functional connectivity (FC) between the cerebral hemispheres of patients with stroke was lower than that of healthy individuals but increased after tDCS.ConclusionThe cognitive and brain functions of patients with mild-to-moderate stroke were damaged but recovered to a degree after tDCS. Increased cortical activation and increased FC between the bilateral cerebral hemispheres measured by fNIRS are promising biomarkers to assess the effectiveness of tDCS in stroke

    Plumbagin inhibits fungal growth, HMGB1/LOX-1 pathway and inflammatory factors in A. fumigatus keratitis

    To investigate the anti-inflammatory and antifungal effects of plumbagin (PL) in Aspergillus fumigatus (A. fumigatus) keratitis, the minimum inhibitory concentration (MIC), time-killing curve, spore adhesion, crystal violet staining, calcium fluoride white staining, and Propidium Iodide (PI) staining were employed to assess the antifungal activity of PL in vitro against A. fumigatus. The cytotoxicity of PL was assessed using the Cell Counting Kit-8 (CCK8). The impact of PL on the expression of HMGB1, LOX-1, TNF-α, IL-1β, IL-6, IL-10 and ROS in A. fumigatus keratitis was investigated using RT-PCR, ELISA, Western blot, and Reactive oxygen species (ROS) assay. The therapeutic efficacy of PL against A. fumigatus keratitis was assessed through clinical scoring, plate counting, Immunofluorescence and Hematoxylin-Eosin (HE) staining. Finally, we found that PL inhibited the growth, spore adhesion, and biofilm formation of A. fumigatus and disrupted the integrity of its cell membrane and cell wall. PL decreased IL-6, TNF-α, and IL-1β levels while increasing IL-10 expression in fungi-infected mice corneas and peritoneal macrophages. Additionally, PL significantly attenuated the HMGB1/LOX-1 pathway while reversing the promoting effect of Boxb (an HMGB1 agonist) on HMGB1/LOX-1. Moreover, PL decreased the level of ROS. In vivo, clinical scores, neutrophil recruitment, and fungal burden were all significantly reduced in infected corneas treated with PL. In summary, the inflammatory process can be inhibited by PL through the regulation of the HMGB-1/LOX-1 pathway. Simultaneously, PL can exert antifungal effects by limiting fungal spore adhesion and biofilm formation, as well as causing destruction of cell membranes and walls
