    Cold hardiness of Phauda flammans (Lepidoptera: Zygaenidae) larvae

    This study aimed to determine the cold hardiness of Phauda flammans (Lepidoptera: Zygaenidae) larvae. Supercooling points of the 1st–6th instar larvae of P. flammans ranged from –7.7 to –13.0 °C. The lethal temperatures were –8 °C for 1st, –5 °C for 2nd, and –7 °C for 3rd–6th instars. Lethal times at the instar-specific lethal temperatures were 12 h for 1st, 14 h for 2nd, 15 h for 3rd, 17 h for 4th, and 18 h for 5th–6th instars. The times required for all larvae to die in an incubator at 5 °C were 30 d for 1st, 3rd, 4th, and 5th instars, and 25 d for 2nd and 6th instars. The findings suggest that P. flammans is a chill-intolerant species, and larvae will die if the air temperature decreases to –5 to –8 °C for 12–18 h or to 5 °C for 25–30 d. Such conditions are, however, unlikely to occur in southern China

    Effects of high glucose on expression of OPG and RANKL in rat aortic vascular smooth muscle cells

    AbstractObjectiveTo explore effect of high glucose on expression of osteoprotegerin (OPG) and receptor activator of NF– κ B ligand (RANKE) in rat aortic vascular smooth muscle cells.MethodsSD rats were intraperitoneally injected with streptozotocin, OPG and RANKL expression in rat thoracic aortas were detected by immunohistochemical staining. In cultured vascular smooth muscle cells (VSMCs) (A7r5), qRT–PCR and Western blot analysis were used to examine the mRNA and protein levels of OPG and RANKL.ResultsOur results demonstrated that OPG expression was increased in hyperglycemic rat aortic VSMCs, while RANKL expression was decreased. Besides, in vitro experiments high glucose induced OPG expression, but depressed RANKL expression by dose– and time–dependent manner in cultured A7r5.ConclusionsOur findings suggested that high glucose could promote the expression of OPG, and inhibit the expression of RANKL in VSMCs, which may be partly be the molecular mechanism of diabetic vascular calcification

    The Effect of Direction on Cursor Moving Kinematics

    There have been only few studies to substantiate the kinematic characteristics of cursor movement. In this study, a quantitative experimental research method was used to explore the effect of moving direction on the kinematics of cursor movement in 24 typical young persons using our previously developed computerized measuring program. The results of multiple one way repeated measures ANOVAs and post hoc LSD tests demonstrated that the moving direction had effects on average velocity, movement time, movement unit and peak velocity. Moving leftward showed better efficiency than moving rightward, upward and downward from the kinematic evidences such as velocity, movement unit and time. Moreover, the unique pattern of the power spectral density (PSD) of velocity (strategy for power application) explained why the smoothness was still maintained while moving leftward even under an unstable situation with larger momentum. Moreover, the information from this cursor moving study can guide us to relocate the toolbars and icons in the window interface, especially for individuals with physical disabilities whose performances are easily interrupted while controlling the cursor in specific directions

    Programmed Cell Death and Aerenchyma Formation in Water-Logged Sunflower Stems and Its Promotion by Ethylene and ROS

    Previous studies have shown that waterlogging/ hypoxic conditions induce aerenchyma formation to facilitate gas exchange. Ethylene (ET) and reactive oxygen species (ROS), as regulatory signals, might also be involved in these adaptive responses. However, the interrelationships between these signals have seldom been reported. Herein, we showed that programmed cell death (PCD) was involved in aerenchyma formation in the stem of Helianthus annuus. Lysigenous aerenchyma formation in the stem was induced through waterlogging (WA), ethylene and ROS. Pre-treatment with the NADPH oxidase inhibitor diphenyleneiodonium (DPI) partially suppressed aerenchyma formation in the seedlings after treatment with WA, ET and 3-amino-1, 2, 4-triazole (AT, catalase inhibitor). In addition, pre-treatment with the ethylene perception inhibitor 1-methylcyclopropene (1-MCP) partially suppressed aerenchyma formation induced through WA and ET in the seedlings, but barely inhibited aerenchyma formation induced through ROS. These results revealed that ethylene-mediated ROS signaling plays a role in aerenchyma formation, and there is a causal and interdependent relationship during WA, ET and ROS in PCD, which regulates signal networks in the stem of H. annuus

    Learning from Rich Semantics and Coarse Locations for Long-tailed Object Detection

    Long-tailed object detection (LTOD) aims to handle the extreme data imbalance in real-world datasets, where many tail classes have scarce instances. One popular strategy is to explore extra data with image-level labels, yet it produces limited results due to (1) semantic ambiguity -- an image-level label only captures a salient part of the image, ignoring the remaining rich semantics within the image; and (2) location sensitivity -- the label highly depends on the locations and crops of the original image, which may change after data transformations like random cropping. To remedy this, we propose RichSem, a simple but effective method, which is robust to learn rich semantics from coarse locations without the need of accurate bounding boxes. RichSem leverages rich semantics from images, which are then served as additional soft supervision for training detectors. Specifically, we add a semantic branch to our detector to learn these soft semantics and enhance feature representations for long-tailed object detection. The semantic branch is only used for training and is removed during inference. RichSem achieves consistent improvements on both overall and rare-category of LVIS under different backbones and detectors. Our method achieves state-of-the-art performance without requiring complex training and testing procedures. Moreover, we show the effectiveness of our method on other long-tailed datasets with additional experiments.     Clinical features and major histocompatibility complex genes as potential susceptibility factors in pediatric immune thrombocytopenia

    Background/PurposeImmune thrombocytopenia (ITP) is a heterogeneous autoimmune disorder with diverse response rates to treatments that include corticosteroids, intravenous immunoglobulins (IVIG), and splenectomy. The predisposing causes of this autoimmune disorder, one of which is immunogenetic susceptibility, have not been fully determined. We investigated whether clinical features and human leukocyte antigen (HLA) genotypes influence the occurrence, treatment response, and disease duration of childhood ITP in Taiwan.MethodsWe performed HLA genotyping of 70 Taiwanese children with ITP and of 70 healthy controls and compared the data. Demographic data were also collected and evaluated.ResultsThe frequencies of heterozygous HLA-A11 and the HLA-Cw1 allele were both significantly decreased in the ITP group (p = 0.0160 and p = 0.0089, respectively), whereas the frequency of heterozygous HLA-DQ5 was significantly increased in the ITP group (p = 0.0057). Patients with HLA-DRB1*11 or -DRB1*15 were more likely to respond poorly to corticosteroids than IVIG (p = 0.0446 and p = 0.0008, respectively). In addition, we observed a positive association between HLA-A11 homozygosity and the development of persistent or chronic ITP [odds ratio (OR) = 6.3165, p = 0.0479]. The presence of HLA-DRB1*08 was, however, negatively correlated with the development of persistent or chronic ITP (OR = 0.1729, p = 0.0657). Children with antecedent of preceding illness (API) and with a younger age of onset were more likely to experience a better treatment response and shorter course of ITP.ConclusionWe suggest that API, age of onset, and particular HLA class I and class II alleles, may be involved in and influence the occurrence and disease duration of childhood ITP, as well as responses to different therapeutic approaches

    Suppression of the RAC1/MLK3/p38 Signaling Pathway by β-Elemene Alleviates Sepsis-Associated Encephalopathy in Mice

    It is still difficult to treat sepsis-associated encephalopathy (SAE) which is a diffuse brain dysfunction caused by sepsis, with excessive activation of microglia as one of the main mechanisms. Ras-related C3 botulinum toxin substrate 1 (RAC1) is proven to be a key molecule in the inflammatory signaling network. By using microglial cell line BV-2 and a mouse model of cecal ligation puncture (CLP), we herein evaluated the effects of β-elemene, an extract of Curcuma zedoaria Rosc., on RAC1 signaling in microglia. β-Elemene decreased the expressions of pro-inflammatory cytokines [tumor necrosis factor-α (TNF-α), interleukin-1β (IL-1β), and IL-6] and attenuated translocation of nuclear factor-κB (NF-κB) p65 from the cytosol to the nucleus in BV-2 cells after lipopolysaccharide (LPS) treatment. It also inhibited the activation of RAC1, mixed-lineage protein kinase 3 (MLK3) and p38 mitogen-activated protein kinase (MAPK). The phosphorylation of the RAC1 Ser71 site was increased by β-elemene. Moreover, the learning and memory abilities of CLP mice in the water maze test and fear conditioning test were improved after β-elemene treatment. It reduced the expression of the microglial marker IBA1, significantly increased RAC1 Ser71 phosphorylation, and suppressed the RAC1/MLK3/p38 signaling activation and inflammatory response in the hippocampus. In conclusion, β-elemene effectively alleviated SAE in mice and inhibited the RAC1/MLK3/p38 signaling pathway in microglia, and might be an eligible potential candidate for SAE treatment

    Increasing Area and Decreasing Depth: Climate Change Influence on Snow Variations in the Qilian Mountains

    The Qilian Mountains serve as a critical water source for the Yellow River and various inland rivers, playing a pivotal role in regulating the regional climate. Given their significance as one of the foremost water resources in the area, the spatial and temporal dynamics of the snow are crucial for understanding potential impacts on regional hydrology and ecology. This study examines the characteristics of spatial and temporal variations in snow-covered extent (SCE), snow depth (SD), snow-covered days (SCD), snow onset date (SOD), and snow end date (SED) within the Qilian Mountains region. We investigate the hydrological and ecological implications utilizing snow area and phenology data, alongside SD data. The findings indicate that: (1) the distribution of snow across the Qilian Mountains mainly splits between the central and western areas, with the central region showing deeper snow than both the eastern and western parts; (2) the area covered by snow in the Qilian Mountains is growing, but the depth of the snow is on a decline, especially in the central area; (3) in terms of snow phenology, most of the region is witnessing an earlier start of SOD, a longer SCD, and an earlier SED. An overall increase in precipitation is identified as the key factor behind the expanded SCE in the Qilian Mountains, while rising temperatures are pinpointed as the primary cause for the reduction in SD. As global climate change intensifies, the observed alterations in the snow of the Qilian Mountains present emerging challenges for regional water security and ecological equilibrium
