    Behavioral and Psychological Factors Associated with 12-Month Weight Change in a Physical Activity Trial

    Examining behavioral and psychological factors relating to weight stability over a 1-year period is of public health importance. We conducted a physical activity (PA) intervention trial for women (N = 247; mean age = 47.5 ± 10.7; mean BMI = 28.6 ± 5.3) in which participants were assigned to one of three groups (two PA and one contact-control). By Month 12, participants achieved 140.4 ± 14.82 min of PA/week, with no group differences. Weight status change from baseline to Month 12 was categorized: no change (N = 154; 62.4%); increase (N = 34; 13.8%); decrease (N = 59; 23.9%). Discriminant function analyses indentified two statistically significant dimensions associated with weight change. Dimension 1 was positively weighted by mood (0.73) and self-efficacy (0.79); dimension 2 was positively weighted to change in physical activity (0.58) and fat consumption (0.55). Results provide further evidence for the importance of behavior in long-term weight maintenance, particularly physical activity and dietary fat. These findings also provide evidence for the importance of addressing psychosocial variables, in particular depressed mood and self-efficacy

    Population status, habitat dependence, and reproductive ecology of Bahama Orioles: a critically endangered synanthropic species.

    Recent elevation of critically endangered Bahama Orioles (Icterus northropi) to species status promptedustoevaluate theirpopulation status,habitatuse,andbreedingecology.Fromsurveys,weestimatedthat at least 141 to 254 individuals remain globally, with 90 to 162, 24 to 44, and 27 to 48 individuals remaining on North Andros Island, Mangrove Cay, and South Andros Island, The Bahamas, respectively. Orioles were observed nesting exclusively in anthropogenic habitat (residential and agricultural land), but home ranges also included nearby pine forest and coppice (dry broadleaf forest). Most nests (40 of 46, or 87%) were in nonnative coconut palm (Cocos nucifera), with native Sabal palmetto and Thrinax morrisii, and an introduced Brassaia actinophylla also used. Trees selected by orioles for nesting were significantly taller, less likely to have shrubs underneath, further from cover, and had more palm trees nearby than randomly selected palm trees. Three of eight nests with known contents were parasitized by Shiny Cowbirds (Molothrus bonariensis). Lethal yellowing disease recently devastated coconutpalmsandreducedthenumberoforiolesonNorthAndros,butpalmsonMangroveCayandSouthAndros remain healthy. The juxtaposition of anthropogenic habitat to suitable native habitats may be more important than any single factor for Bahama Orioles, especially for breeding adults and fledged young. Conservation of coppice habitat, at high risk for agricultural and residential development, is crucial for survival of this critically endangered synanthropic species

    Wildland Fire Reburning Trends Across the US West Suggest Only Short-Term Negative Feedback and Differing Climatic Effects

    Wildfires are a significant agent of disturbance in forests and highly sensitive to climate change. Short-interval fires and high severity (mortality-causing) fires in particular, may catalyze rapid and substantial ecosystem shifts by eliminating woody species and triggering conversions from forest to shrub or grassland ecosystems. Modeling and fine-scale observations suggest negative feedbacks between fire and fuels should limit reburn prevalence as overall fire frequency rises. However, while we have good information on reburning patterns for individual fires or small regions, the validity of scaling these conclusions to broad regions like the US West remains unknown. Both the prevalence of reburning and the strength of feedbacks on likelihood of reburning over differing timescales have not been documented at the regional scale. Here we show that while there is a strong negative feedback for very short reburning intervals throughout wildland forests of the Western US, that feedback weakens after 10–20 years. The relationship between reburning intervals and drought diverges depending on location, with coastal systems reburning quicker (e.g. shorter interval between fires) in wetter conditions and interior forests in drier. This supports the idea that vegetation productivity—primarily fine fuels that accumulate rapidly (years)—is of primary importance in determining reburn intervals. Our results demonstrate that while over short time intervals increasing fires inhibits reburning at broad scales, that breaks down after a decade. This provides important insights about patterns at very broad scales and agrees with finer scale work, suggesting that lessons from those scales apply across the entire western US

    Cafeteria diet-induced obesity causes oxidative damage in white adipose

    Obesity continues to be one of the most prominent public health dilemmas in the world. The complex interaction among the varied causes of obesity makes it a particularly challenging problem to address. While typical high-fat purified diets successfully induce weight gain in rodents, we have described a more robust model of diet-induced obesity based on feeding rats a diet consisting of highly palatable, energy-dense human junk foods – the “cafeteria” diet (CAF, 45-53% kcal from fat). We previously reported that CAF-fed rats became hyperphagic, gained more weight, and developed more severe hyperinsulinemia, hyperglycemia, and glucose intolerance compared to the lard-based 45% kcal from fat high fat diet–fed group. In addition, the CAF diet-fed group displayed a higher degree of inflammation in adipose and liver, mitochondrial dysfunction, and an increased concentration of lipid-derived, pro-inflammatory mediators. Building upon our previous findings, we aimed to determine mechanisms that underlie physiologic findings in the CAF diet. We investigated the effect of CAF diet-induced obesity on adipose tissue specifically using expression arrays and immunohistochemistry. Genomic evidence indicated the CAF diet induced alterations in the white adipose gene transcriptome, with notable suppression of glutathione-related genes and pathways involved in mitigating oxidative stress. Immunohistochemical analysis indicated a doubling in adipose lipid peroxidation marker 4-HNE levels compared to rats that remained lean on control standard chow diet. Our data indicates that the CAF diet drives an increase in oxidative damage in white adipose tissue that may affect tissue homeostasis. Oxidative stress drives activation of inflammatory kinases that can perturb insulin signaling leading to glucose intolerance and diabetes

    Prevalence and predictors of post‐traumatic stress symptoms in adolescent and young adult cancer survivors: a 1‐year follow‐up study

    Objectives Post‐traumatic stress symptoms (PTSS) have been identified as a meaningful indicator of distress in cancer survivors. Distinct from young adult survivors of childhood cancer, young people diagnosed with cancer as adolescents and young adults (AYAs) face unique psychosocial issues; however, there is little published research of PTSS in the AYA population. This study examines prevalence and predictors of PTSS among AYAs with cancer. Methods As part of a longitudinal study of AYAs with cancer, 151 patients aged 15–39 years completed mailed surveys at 6 and 12 months post‐diagnosis. Severity of PTSS was estimated at 6 and 12 months post‐diagnosis. Multiple regression analyses were conducted to investigate the predictive effects of socio‐demographic and clinical characteristics on changes in PTSS over time. Results At 6 and 12 months, respectively, 39% and 44% of participants reported moderate to severe levels of PTSS; 29% had PTSS levels suggestive of post‐traumatic stress disorder. No significant differences in severity of PTSS between 6 and 12 months were observed. Regression analyses suggested that a greater number of side effects were associated with higher levels of PTSS at 6 months. Currently receiving treatment, having surgical treatment, diagnosis of a cancer type with a 90–100% survival rate, remaining unemployed/not in school, and greater PTSS at 6 months were associated with higher levels of PTSS at 12 months. Conclusions Post‐traumatic stress symptoms were observed as early as 6 months following diagnosis and remained stable at 12‐month follow‐up. The development of early interventions for reducing distress among AYA patients in treatment is recommended.     Rapidly Characterizing the Fast Dynamics of RNA Genetic Circuitry with Cell-Free Transcription Translation (TX-TL) Systems

    RNA regulators are emerging as powerful tools to engineer synthetic genetic networks or rewire existing ones. A potential strength of RNA networks is that they may be able to propagate signals on time scales that are set by the fast degradation rates of RNAs. However, a current bottleneck to verifying this potential is the slow design-build-test cycle of evaluating these networks in vivo. Here, we adapt an Escherichia coli-based cell-free transcription-translation (TX-TL) system for rapidly prototyping RNA networks. We used this system to measure the response time of an RNA transcription cascade to be approximately five minutes per step of the cascade. We also show that this response time can be adjusted with temperature and regulator threshold tuning. Finally, we use TX-TL to prototype a new RNA network, an RNA single input module, and show that this network temporally stages the expression of two genes in vivo

    Dissolved and particulate barium distributions along the US GEOTRACES North Atlantic and East Pacific zonal transects (GA03 and GP16): global implications for the marine barium cycle

The definitive version was published in Global Biogeochemical Cycles 36(6), (2022): e2022GB007330, https://doi.org/10.1029/2022gb007330.Processes controlling dissolved barium (dBa) were investigated along the GEOTRACES GA03 North Atlantic and GP16 Eastern Tropical Pacific transects, which traversed similar physical and biogeochemical provinces. Dissolved Ba concentrations are lowest in surface waters (∼35–50 nmol kg−1) and increase to 70–80 and 140–150 nmol kg−1 in deep waters of the Atlantic and Pacific transects, respectively. Using water mass mixing models, we estimate conservative mixing that accounts for most of dBa variability in both transects. To examine nonconservative processes, particulate excess Ba (pBaxs) formation and dissolution rates were tracked by normalizing particulate excess 230Th activities. Th-normalized pBaxs fluxes, with barite as the likely phase, have subsurface maxima in the top 1,000 m (∼100–200 μmol m−2 year−1 average) in both basins. Barite precipitation depletes dBa within oxygen minimum zones from concentrations predicted by water mass mixing, whereas inputs from continental margins, particle dissolution in the water column, and benthic diffusive flux raise dBa above predications. Average pBaxs burial efficiencies along GA03 and GP16 are ∼37% and 17%–100%, respectively, and do not seem to be predicated on barite saturation indices in the overlying water column. Using published values, we reevaluate the global freshwater dBa river input as 6.6 ± 3.9 Gmol year−1. Estuarine mixing processes may add another 3–13 Gmol year−1. Dissolved Ba inputs from broad shallow continental margins, previously unaccounted for in global marine summaries, are substantial (∼17 Gmol year−1), exceeding terrestrial freshwater inputs.
