    Quad Dominant 2-Manifold Mesh Modeling

    In this dissertation, I present a modeling framework that provides modeling of 2D smooth meshes in arbitrary topology without any need for subdivision. In the framework, each edge of a quad face is represented by a smooth spline curve, which can be manipulated using edge vertices and additional tangential points. The overall smoothness is achieved by interpolating all four edges of any given quad across the quad surface. The framework consists of simple quad preserving operations that manipulate the principal curves of the smooth model. These operations are all variants of a generic “Curve Split" and its inverse, “Region Collapse". By only using these sets of simple operations, it is possibly to model any desired shape conveniently. I also provide implementation guidelines for these operations. In the results of this dissertation, I present three main applications for this modeling framework. The major application is modeling Mock3D shapes; shapes with well defined interior normals by interpolating the normals at the boundaries of the shape across its surface which can serve as a mock 3D model to mimic a 3D CGI look. As a second application, the framework can be used in origami modeling by allowing assignment of crease patterns across the surface of 2D shapes modelled. Finally, vectorization of reference photos via modeling figures by following their contours is presented as a third application

    A Prevention and a Traction System for Ransomware Attacks

    Over the past three years, especially following WannaCry malware, ransomware has become one of the biggest concerns for private businesses, state, and local government agencies. According to Homeland Security statistics, 1.5 million ransomware attacks have occurred per year since 2016. Cybercriminals often use creative methods to inject their malware into the target machines and use sophisticated cryptographic techniques to hold hostage victims' files and programs unless a certain amount of equivalent Bitcoin is paid. The return to the cybercriminals is so high (estimated \$1 billion in 2019) without any cost because of the advanced anonymity provided by cryptocurrencies, especially Bitcoin \cite{Paquet-Clouston2019}. Given this context, this study first discusses the current state of ransomware, detection, and prevention systems. Second, we propose a global ransomware center to better manage our concerted efforts against cybercriminals. The policy implications of the proposed study are discussed in the conclusion section

    Doxycycline down-regulates matrix metalloproteinase expression and inhibits NF-κB signaling in LPS-induced PC3 cells

    Introduction. Matrix metalloproteinase enzymes (MMPs) play important role in inflammation, malignant cell proliferation, invasion and angiogenesis by mediating extracellular matrix degradation. Doxycycline, a synthetic tetracycline, behaves as a MMP inhibitor at a subantimicrobial dose and inhibits tumor cell proliferation, invasion and angiogenesis. The aberrant activity of nuclear factor kappa B (NF-κB) causes activation of MMPs and thereby proliferation and invasion of cancer cells. The aim of this study was to investigate the effects of doxycycline on the expression of MMPs in lipopolysaccharide (LPS)-induced PC3 human prostate cancer cells and the possible role of NF-κB signaling. Material and methods. PC3 cells were incubated with LPS (0.5 μg/mL) for 24 h in the presence or absence of doxycycline (5 μg/mL). The effects of LPS and doxycycline on the expressions of MMP-2, MMP-8, MMP-9, MMP-10, NF-κB/p65, IκB-α, p-IκB-α, IKK-β were examined by Western blotting and immunohistochemistry in PC3 cells. Furthermore, relative proteinase activities of MMP-2 and MMP-9 were determined by gelatin zymography. Results. LPS increased expression and activity of MMP-9 and expression of MMP-8, MMP-10, NF-κB /p65, p-IκB-α, IKK-β and doxycycline down-regulated its effects with the exception of MMP-10 expression. The expression of MMP-2 and IκB-α was affected by neither LPS nor doxycycline. Conclusions. Our findings indicate that doxycycline inhibits the expression of various MMPs and NF-κB signaling may play a role in the regulation of MMPs expression in LPS-induced PC3 human prostate cancer cells

    Combining Data Sources Nonlinearly for Cell Nucleus Classification of Renal Cell Carcinoma

    In kernel-based machine learning algorithms, we can learn a combination of different kernel functions in order to obtain a similarity measure that better matches the underlying problem instead of using a single fixed kernel function. This approach is called multiple kernel learning (MKL). In this paper, we formulate a nonlinear MKL variant and apply it for nuclei classification in tissue microarray images of renal cell carcinoma (RCC). The proposed variant is tested on several feature representations extracted from the automatically segmented nuclei. We compare our results with single-kernel support vector machines trained on each feature representation separately and three linear MKL algorithms from the literature. We demonstrate that our variant obtains more accurate classifiers than competing algorithms for RCC detection by combining information from different feature representations nonlinearly

    The effect of Kinesio taping on back pain in patients with Lenke Type 1 adolescent idiopathic scoliosis: A randomized controlled trial

    Purpose: This study investigated the short-term effects of KT on back pain (BP) in patients with Lenke Type 1 adolescent idiopathic scoliosis (AIS). Methods: We chosen Lenke Type 1 scoliosis who have had only back pain (the localization of the pain: the only in the apical convex edge). Forty patients suffering from BP with Lenke Type 1 AIS were randomly separated into two groups, Group 1 (20 patients) and Group 2 (20 patients). Group 1 was given KT with tension and home exercises and Group 2 was given KT without tension and home exercises. KT and home exercises was applied to the thoracic area of the patients in both groups for four weeks. Pain intensity was measured using a visual analog scale (VAS) and SRS-22 (subtotal SRS-20) before and after treatment. Results: Mean age of both groups was 16.1 years. Mean Cobb angle of the thoracic scoliosis was 31.8° (range: 17°–44°) in Group 1 and 32.8° (range: 19°–43°) in Group 2 before the treatment. The decrease in VAS score of Group 1 after taping was higher than that of Group 2. The difference between the pre- and post-treatment VAS scores of both groups was statistically significant (p < 0.05). The increase in mean SRS-20 score of Group 1 following taping application was significantly higher than the increase in the control group (p < 0.05). Conclusion: Results demonstrated that KT application with tension effectively leads to back pain relief shortly after application. In addition, KT has a positive impact on quality of life. Thus, KT may be a suitable intervention in treating back pain of patients with AIS.     Low-dose doxycycline inhibits hydrogen peroxide-induced oxidative stress, MMP-2 up-regulation and contractile dysfunction in human saphenous vein grafts

    WOS: 000469125200001PubMed ID: 31213768Background: Cardiopulmonary bypass (CPB) applied during coronary artery bypass grafting (CABG), promotes inflammation, generation of reactive oxygen species (ROS) and up-regulation of matrix metalloproteinases (MMPs). All these complications may lead to contractile dysfunction, restenosis and early graft failure, restricting long-term efficacy of bypass grafts. Low-dose doxycycline is a potent MMP inhibitor and ROS scavenger. In this study, we aimed to investigate the effects of doxycycline on ROS generation, MMP regulation and contractile dysfunction induced by H2O2 in human saphenous vein (HSV) grafts. Methods: HSV grafts (n=7) were divided into four groups after removing endothelial layer by mechanical scratching and incubated with 10 mu M H2O2 and/or 10 mu M doxycycline for 16 hrs. Untreated segments served as control. Concentration-response curves to noradrenaline (NA), potassium chloride (KCl), serotonin (5-HT) and papaverine were performed. Superoxide anion and other ROS levels were determined by using lucigenin- and luminol-enhanced chemiluminescence assays, respectively. Expression/activity of gelatinases (MMP-2/MMP-9) was examined by gelatin zymography. MMP-13 expression was evaluated by immunostaining/immunoscoring. Results: H2O2 incubation increased superoxide anion and other ROS levels. Doxycycline prevented these increments. H2O2 suppressed contractile responses to NA, KCl and 5-HT. Doxycycline ameliorated contractions to NA and KCl but not to 5-HT. H2O2 or doxycycline did not altered relaxation to papaverine. MMP-2 and MMP-13 expression increased with H2O2, but doxycycline inhibited MMP-2 up-regulation/activation.
