    Design and Characterisation of Soft Biomaterials and Interfaces for Bio-Adhesives and Cell Culture

    PhDThis thesis consists of three chapters which can be split into studies of (1) the impact of length scale, surface adhesion and heterogeneity on the mechanical charac-terisation of soft biomaterials, (2) the effect of non-covalent cross-linking chemistry on the mechanics of CMC hydrogels and (3) analytical characterisation of the interfa-cial mechanics of liquid-liquid interfaces. The mechanical properties of soft materials used in the biomedical field play an important role on their performance. In the field of tissue engineering, it is known that cells sense the mechanical properties of their environment, however some mate-rials, such as Sylard 184 PDMS (poly(dimethylsiloxane)), have failed to elicit such response. It was proposed that differences in the mechanical properties of such soft materials, at different scales, could account for these discrepancies. Indeed, the varia-tion in the elastic moduli obtained for soft materials characterised at different scales can span several orders of magnitude. This called for a side-by-side comparison of the mechanical behaviour of soft materials at different scales. Here we use indentation, rheology and atomic force microscopy nanoidentation (using different tip geometries) to characterise the mechanical properties of PDMS, poly(acrylamide) (PAAm) and carboxymethyl cellulose (CMC) hydrogels at different length scales. Our results high-light the importance of surface adhesion and the resulting changes in contact area, and sample microstructural heterogeneity, in particular for the mechanical characterisation of ultra-soft substrates at the nano- to micro-scale. Next the impact of inorganic divalent cationic and organic cationic polyelec-trolyte cross-linkers on the rheological and adhesive properties of CMC hydrogels was studied. The inorganic cations used in this study were Sr2+ and Ca2+ resulted in no significant change in the bulk rheological properties and apparent chain collapse, but an increase in the adhesive strength. The introduction of organic polyelectrolytes caused complex coacervation with the CMC polymer chains. Some CMC chain bridg-ing occurred, but the interactions between the polyelectrolytes and CMC chains caused complexes to form, resulting in polymer rich complexes to form within a more dilute matrix. The introduction of organic polyelectrolyte cross-links failed to increase the bulk mechanical properties studied by rheology but did increase the adhesive strength. Finally the impact of protein adsorption on interfacial mechanics was studied. The interfacial mechanics was studied by interfacial shear rheology and interfacial nanoindentation by AFM, and PLL, BSA and lysozyme were the proteins chosen for this study. Proteins adsorbed at the interface resulted in a significant increase in the interfacial shear moduli, however this was not seen by nanoindentation. By modelling the interfacial mechanics as the superposition of the film forces and the interfacial surface tension forces we showed that when testing by nanoindentation changes in surface tension and surface potential have a significant impact on the interfacial me-chanics. The addition of protein films was shown to have a surfactant effect on inter-faces essentially softening them, this results in the films essentially having soft sub-strates and is proposed as the main cause of discrepancies between interfacial rheology and nanoindentation by AFM

    This paper is inspired by the sentiments of Winston Churchill, “The further back you can look, the farther forward you are likely to see.” Notwithstanding all the work and intellectual efforts by current scholars and practitioners on the topic of good leadership and good character, there is much to be learned by drawing from the ancient philosophical tradition, notably Socrates, Plato and Aristotle. Simply said, it is critically important that we learn from and apply the wisdom of the ancients to the current thoughts and practices as to how character and leadership are acquired and developed. The timeless wisdom of the ancients is even more relevant today given the ever-increasing pace, complexity and uncertainty faced by individuals, organizations and societies

    Extreme Reversal in Mechanical Anisotropy in Liquid-Liquid Interfaces Reinforced with Self-Assembled Protein Nanosheets

    The structuring of liquid–liquid and liquid–air interfaces may play an important role in novel microfabrication platforms and biotechnologies, from the spontaneous formation of microfilaments from liquid droplets and the 3D printing of liquids, to the culture of stem cells on emulsions. Understanding the mechanical anisotropy of associated liquid interfaces is essential for the development of such systems. Models of AFM indentation at liquid interfaces, based on the Young-Laplace model, currently do not allow the quantification of interfacial mechanical properties of associated molecular films. This report presents such a model and compares its predictions to interfacial mechanical properties characterised via interfacial shear rheology. An extreme reversal of mechanical anisotropy of liquid–liquid interfaces is observed, upon self-assembly of protein nanosheets, by 5 orders of magnitude. Results indicate that, although interfacial rheology is more sensitive than AFM indentation to the mechanics of molecular films in the low range of interfacial mechanics, AFM indentation allows the quantification of mechanical properties of stiffer molecular films, and remains better adapted to the characterisation of small samples and enables the characterisation of local heterogeneity

    The culture of HaCaT cells on liquid substrates is mediated by a mechanically strong liquid-liquid interface

    The mechanical properties of naturally-derived matrices and biomaterials are thought to play an important role in directing cell adhesion, spreading, motility, proliferation and differentiation. However, recent reports have indicated that cells may respond to local nanoscale physical cues, rather than bulk mechanical properties. We had previously reported that primary keratinocytes and mesenchymal stem cells did not seem to respond to the bulk mechanical properties of poly(dimethyl siloxane) (PDMS) substrates. In this study, we examine the mechanical properties of weakly crosslinked PDMS substrates and observe a liquid-like behaviour, with complete stress relaxation. We then report the observation that HaCaT cells, an epidermal cell line, proliferate readily at the surface of uncrosslinked liquid PDMS, as well as on low viscosity (0.77 cSt) fluorinated oil. These results are surprising, considering current views in the field of mechanotransduction on the importance of bulk mechanical properties, but we find that strong mechanical interfaces, presumably resulting from protein assembly, are formed at liquid–liquid interfaces for which cell adhesion and proliferation are observed. Hence our results suggest that cells sense the nanoscale mechanical properties of liquid–liquid interfaces and that such physical cues are sufficient to sustain the proliferation of adherent cells

    Programmed Self-Assembly of DNA Nanosheets with Discrete Single-Molecule Thickness and Interfacial Mechanics: Design, Simulation, and Characterization.

    DNA is programmed to hierarchically self-assemble into superstructures spanning from nanometer to micrometer scales. Here, we demonstrate DNA nanosheets assembled out of a rationally designed flexible DNA unit (F-unit), whose shape resembles a Feynman diagram. F-units were designed to self-assemble in two dimensions and to display a high DNA density of hydrophobic moieties. oxDNA simulations confirmed the planarity of the F-unit. DNA nanosheets with a thickness of a single DNA duplex layer and with large coverage (at least 30 ÎĽm Ă— 30 ÎĽm) were assembled from the liquid phase at the solid/liquid interface, as unambiguously evidenced by atomic force microscopy imaging. Interestingly, single-layer nanodiscs formed in solution at low DNA concentrations. DNA nanosheet superstructures were further assembled at liquid/liquid interfaces, as demonstrated by the fluorescence of a double-stranded DNA intercalator. Moreover, the interfacial mechanical properties of the nanosheet superstructures were measured as a response to temperature changes, demonstrating the control of interfacial shear mechanics based on DNA nanostructure engineering. The rational design of the F-unit, along with the presented results, provide an avenue toward the controlled assembly of reconfigurable/responsive nanosheets and membranes at liquid/liquid interfaces, to be potentially used in the characterization of biomechanical processes and materials transport

    Protein Nanosheet Mechanics Controls Cell Adhesion and Expansion on Low-Viscosity Liquids

    Adherent cell culture typically requires cell spreading at the surface of solid substrates to sustain the formation of stable focal adhesions and assembly of a contractile cytoskeleton. However, a few reports have demonstrated that cell culture is possible on liquid substrates such as silicone and fluorinated oils, even displaying very low viscosities (0.77 cSt).     Improving quality of care and outcome at very preterm birth: the Preterm Birth research programme, including the Cord pilot RCT

    BACKGROUND: Being born very premature (i.e. before 32 weeks’ gestation) has an impact on survival and quality of life. Improving care at birth may improve outcomes and parents’ experiences. OBJECTIVES: To improve the quality of care and outcomes following very preterm birth. DESIGN: We used mixed methods, including a James Lind Alliance prioritisation, a systematic review, a framework synthesis, a comparative review, qualitative studies, development of a questionnaire tool and a medical device (a neonatal resuscitation trolley), a survey of practice, a randomised trial and a protocol for a prospective meta-analysis using individual participant data. SETTING: For the prioritisation, this included people affected by preterm birth and health-care practitioners in the UK relevant to preterm birth. The qualitative work on preterm birth and the development of the questionnaire involved parents of infants born at three maternity hospitals in southern England. The medical device was developed at Liverpool Women’s Hospital. The survey of practice involved UK neonatal units. The randomised trial was conducted at eight UK tertiary maternity hospitals. PARTICIPANTS: For prioritisation, 26 organisations and 386 individuals; for the interviews and questionnaire tool, 32 mothers and seven fathers who had a baby born before 32 weeks’ gestation for interviews evaluating the trolley, 30 people who had experienced it being used at the birth of their baby (19 mothers, 10 partners and 1 grandmother) and 20 clinicians who were present when it was being used; for the trial, 261 women expected to have a live birth before 32 weeks’ gestation, and their 276 babies. INTERVENTIONS: Providing neonatal care at very preterm birth beside the mother, and with the umbilical cord intact; timing of cord clamping at very preterm birth. MAIN OUTCOMES MEASURES: Research priorities for preterm birth; feasibility and acceptability of the trolley; feasibility of a randomised trial, death and intraventricular haemorrhage. REVIEW METHODS: Systematic review of Cochrane reviews (umbrella review); framework synthesis of ethics aspects of consent, with conceptual framework to inform selection criteria for empirical and analytical studies. The comparative review included studies using a questionnaire to assess satisfaction with care during childbirth, and provided psychometric information. RESULTS: Our prioritisation identified 104 research topics for preterm birth, with the top 30 ranked. An ethnographic analysis of decision-making during this process suggested ways that it might be improved. Qualitative interviews with parents about their experiences of very preterm birth identified two differences with term births: the importance of the staff appearing calm and of staff taking control. Following a comparative review, this led to the development of a questionnaire to assess parents’ views of care during very preterm birth. A systematic overview summarised evidence for delivery room neonatal care and revealed significant evidence gaps. The framework synthesis explored ethics issues in consent for trials involving sick or preterm infants, concluding that no existing process is ideal and identifying three important gaps. This led to the development of a two-stage consent pathway (oral assent followed by written consent), subsequently evaluated in our randomised trial. Our survey of practice for care at the time of birth showed variation in approaches to cord clamping, and that no hospitals were providing neonatal care with the cord intact. We showed that neonatal care could be provided beside the mother using either the mobile neonatal resuscitation trolley we developed or existing equipment. Qualitative interviews suggested that neonatal care beside the mother is valued by parents and acceptable to clinicians. Our pilot randomised trial compared cord clamping after 2 minutes and initial neonatal care, if needed, with the cord intact, with clamping within 20 seconds and initial neonatal care after clamping. This study demonstrated feasibility of a large UK randomised trial. Of 135 infants allocated to cord clamping ≥ 2 minutes, 7 (5.2%) died and, of 135 allocated to cord clamping ≤ 20 seconds, 15 (11.1%) died (risk difference –5.9%, 95% confidence interval –12.4% to 0.6%). Of live births, 43 out of 134 (32%) allocated to cord clamping ≥ 2 minutes had intraventricular haemorrhage compared with 47 out of 132 (36%) allocated to cord clamping ≤ 20 seconds (risk difference –3.5%, 95% CI –14.9% to 7.8%). LIMITATIONS: Small sample for the qualitative interviews about preterm birth, single-centre evaluation of neonatal care beside the mother, and a pilot trial. CONCLUSIONS: Our programme of research has improved understanding of parent experiences of very preterm birth, and informed clinical guidelines and the research agenda. Our two-stage consent pathway is recommended for intrapartum clinical research trials. Our pilot trial will contribute to the individual participant data meta-analysis, results of which will guide design of future trials. FUTURE WORK: Research in preterm birth should take account of the top priorities. Further evaluation of neonatal care beside the mother is merited, and future trial of alternative policies for management of cord clamping should take account of the meta-analysis.     An argument for the use of Aristotelian method in bioethics

    The main claim of this paper is that the method outlined and used in Aristotle's Ethics is an appropriate and credible one to use in bioethics. Here “appropriate” means that the method is capable of establishing claims and developing concepts in bioethics and “credible” that the method has some plausibility, it is not open to obvious and immediate objection. It begins by suggesting why this claim matters and then gives a brief outline of Aristotle's method. The main argument is made in three stages. First, it is argued that Aristotelian method is credible because it compares favourably with alternatives. In this section it is shown that Aristotelian method is not vulnerable to criticisms that are made both of methods that give a primary place to moral theory (such as utilitarianism) and those that eschew moral theory (such as casuistry and social science approaches). As such, it compares favourably with these other approaches that are vulnerable to at least some of these criticisms. Second, the appropriateness of Aristotelian method is indicated through outlining how it would deal with a particular case. Finally, it is argued that the success of Aristotle's philosophy is suggestive of both the credibility and appropriateness of his method.</p
