    Genetic Diversity Among Banana streak virus Isolates from Australia

    Banana streak virus (BSV) is an important pathogen of bananas and plantains (Musa spp.) throughout the world. We have cloned and sequenced part of the genomes of four isolates of BSV from Australia, designated BSV-RD, BSV-Cav, BSV-Mys, and BSV-GF. These isolates originated from banana cvs. Red Dacca, Williams, Mysore, and Goldfinger, respectively. All clones contained a sequence covering part of open reading frame III and the intergenic region of the badnavirus genome. The sequences were compared with those of other badnaviruses, including BSV-Onne, a previously characterized isolate from Nigeria. The BSV-RD sequence was virtually identical to that of BSV-Onne, differing by only two nucleotides over 1,292 bp. However, BSV-Cav, -Mys, and -GF were divergent in nucleotide sequence. Phylogenetic analyses using conserved sequences in the ribonuclease H domain revealed that all BSV isolates were more closely related to each other than to any other badnavirus. BSV-Cav was most closely related to BSV-Onne, and there was 95.1% identity between the two amino acid sequences. Other relationships between the BSV isolates were less similar, with sequence identities ranging from 66.4 to 78.2%, which is a magnitude comparable to the distance between some of the recognized badnavirus species.     Ecologic analysis of some immune-related disorders, including type 1 diabetes, in Australia: latitude, regional ultraviolet radiation, and disease prevalence.

    The apparent immune-suppressive effect of ultraviolet radiation (UVR) has suggested that this environmental exposure may influence the development of immune-related disorders. Self-reported prevalence rates of type 1 diabetes mellitus, rheumatoid arthritis (RA), eczema/dermatitis, and asthma, from the 1995 Australian National Health Survey, were therefore examined by latitude and ambient level of UVR. A positive association of type 1 diabetes mellitus prevalence was found with both increasing southern latitude of residence (r = 0.77; p = 0.026) and decreasing regional annual ambient UVR (r= -0.80; p = 0.018); a 3-fold increase in prevalence from the northernmost region to the southernmost region was evident. In contrast, asthma correlated negatively with latitude (r = -0.72; p = 0.046), although the change in asthma prevalence from the north to the south of Australia was only 0.7-fold. For both RA and eczema/dermatitis, there were no statistically significant associations between latitude/UVR and disease prevalence. These ecologic data provide some support for a previously proposed beneficial effect of UVR on T-helper 1-mediated autoimmune disorders such as type 1 diabetes. The inverse association of type 1 diabetes prevalence with UVR is consistent with that previously reported for another autoimmune disease, multiple sclerosis, in Australia, and also with type 1 diabetes latitudinal gradients in the Northern Hemisphere. The finding also accords with photoimmunologic evidence of UVR-induced immunosuppression and may suggest a beneficial effect of UVR in reducing the incidence of such autoimmune conditions. In light of this study, analytic epidemiologic studies investigating risk of immune disorders in relation to personal UVR exposure in humans are required

    An important aspect of intergenerational justice concerns the specification of a 'currency of advantage' that can be used to evaluate distributive outcomes across time. Environmental theorists have introduced several innovative currencies of justice in recent years, such as ecological space and critical natural capital. However they have often downplayed the application of established currencies (such as welfare, resources or capabilities) to issues of futurity. After exploring the merits of a number of rival currencies, it is argued that the currency of 'capabilities to function' provides a promising basis for a theory of justice that takes seriously the rights and duties of intergenerational justice

    Adiabatic Domain Wall Motion and Landau-Lifshitz Damping

    Recent theory and measurements of the velocity of current-driven domain walls in magnetic nanowires have re-opened the unresolved question of whether Landau-Lifshitz damping or Gilbert damping provides the more natural description of dissipative magnetization dynamics. In this paper, we argue that (as in the past) experiment cannot distinguish the two, but that Landau-Lifshitz damping nevertheless provides the most physically sensible interpretation of the equation of motion. From this perspective, (i) adiabatic spin-transfer torque dominates the dynamics with small corrections from non-adiabatic effects; (ii) the damping always decreases the magnetic free energy, and (iii) microscopic calculations of damping become consistent with general statistical and thermodynamic considerations

    Analysis of Coaxial Soil Cell in Reflection and Transmission

    Accurate measurement of moisture content is a prime requirement in hydrological, geophysical and biogeochemical research as well as for material characterization and process control. Within these areas, accurate measurements of the surface area and bound water content is becoming increasingly important for providing answers to many fundamental questions ranging from characterization of cotton fiber maturity, to accurate characterization of soil water content in soil water conservation research to bio-plant water utilization to chemical reactions and diffusions of ionic species across membranes in cells as well as in the dense suspensions that occur in surface films. In these bound water materials, the errors in the traditional time-domain-reflectometer, “TDR”, exceed the range of the full span of the material’s permittivity that is being measured. Thus, there is a critical need to re-examine the TDR system and identify where the errors are to direct future research. One promising technique to address the increasing demands for higher accuracy water content measurements is utilization of electrical permittivity characterization of materials. This technique has enjoyed a strong following in the soil-science and geological community through measurements of apparent permittivity via time-domain-reflectometery as well in many process control applications. Recent research however, is indicating a need to increase the accuracy beyond that available from traditional TDR. The most logical pathway then becomes a transition from TDR based measurements to network analyzer measurements of absolute permittivity that will remove the adverse effects that high surface area soils and conductivity impart onto the measurements of apparent permittivity in traditional TDR applications. This research examines the theoretical basis behind the coaxial probe, from which the modern TDR probe originated from, to provide a basis on which to perform absolute permittivity measurements. The research reveals currently utilized formulations in accepted techniques for permittivity measurements which violate the underlying assumptions inherent in the basic models due to the TDR acting as an antenna by radiating energy off the end of the probe, rather than returning it back to the source as is the current assumption. To remove the effects of radiation from the experimental results obtain herein, this research utilized custom designed coaxial probes of various diameters and probe lengths by which to test the coaxial cell measurement technique for accuracy in determination of absolute permittivity. In doing so, the research reveals that the basic models available in the literature all omitted a key correction factor that is hypothesized by this research as being most likely due to fringe capacitance. To test this theory, a Poisson model of a coaxial cell was formulated to calculate the effective extra length provided by the fringe capacitance which is then used to correct the experimental results such that experimental measurements utilizing differing coaxial cell diameters and probe lengths, upon correction with the Poisson model derived correction factor, all produce the same results thereby lending support for the use of an augmented measurement technique, described herein, for measurement of absolute permittivity, as opposed to the traditional TDR measurement of apparent permittivity

    Thermal-Structural Analysis of the MacArthur Maze Freeway Collapse

    At approximately 3:41 AM on the morning of April 29, 2007, a tractor-trailer rig carrying 8,600 gallons (32.6 m{sup 3}) of fuel overturned on Interstate 880 in Oakland, CA. The resultant fire weakened the surrounding steel superstructure and caused a 50-yard (45.7 m) long section of the above connecting ramp from Interstate 80 to Interstate 580 to fail in approximately 18 minutes. In this study, we performed a loosely-coupled thermal-structural finite element analysis of the freeway using the LLNL Engineering codes NIKE3D, DYNA3D and TOPAZ3D. First, we applied an implicit structural code to statically initialize the stresses and displacements in the roadway at ambient conditions due to gravity loading. Next, we performed a thermal analysis by approximating the tanker fire as a moving box region of uniform temperature. This approach allowed for feasible calculation of the fire-to-structure radiative view factors and convective heat transport. We used a mass scaling methodology in the thermal analysis to reduce the overall simulation time so an explicit structural analysis could be used, which provided a more computationally efficient simulation of structural failure. Our approach showed structural failure of both spans due to thermal softening under gravity loading at approximately 20 minutes for a fixed fire temperature of 1200 C and fixed thermal properties. When temperature-dependent thermal properties were applied, the south and north spans collapsed at approximately 10 minutes and 16 minutes, respectively. Finally, we performed a preliminary fully-coupled analysis of the system using the new LLNL implicit multi-mechanics code Diablo. Our investigation shows that our approach provides a reasonable first-order analysis of the system, but improved modeling of the transport properties and the girder-box beam connections is required for more accurate predictions

    Concurrent application of tretinoin (retinoic acid) partially protects against corticosteroid-induced epidermal atrophy

    Cutaneous atrophy arising from prolonged use of potent topical corticosteroids has long been a concern. Thus, it would be advantageous to find an agent which protects against atrophy produced by corticosteroids but at the same time does not impair their anti-inflammatory effects. Recent work shows that topical all- trans retinoic acid (tretinoin) prevents skin atrophy in mice treated with topical corticosteroids, but such studies have not been performed in humans. We performed an 8-week clinical, histological and biochemical study to test the ability of tretinoin to enhance efficacy and inhibit atrophogenicity of topical corticosteroids, when used in the treatment of psoriasis. In each of 20 psoriasis patients, one plaque, and its perilesional skin, was treated once daily with betamethasone dipropionate and tretinoin 0 1 , and one plaque, and its perilesional skin, treated with once daily betamethasone dipropionate and tretinoin vehicle. There was no difference in the speed or degree of improvement in plaques treated with either the topical corticosteroid tretinoin combination or with corticosteroid alone. Light microscopy revealed a 19 reduction in epidermal thickness, in corticosteroid-treated perilesional skin, as compared with a slight (1 ) increase in corticosteroid tretinoin-treated perilesional areas (P 0.067). Western blot analysis showed a 55 reduction in procollagen I aminopropeptide in perilesional skin treated with corticosteroid alone, as compared with a 45 reduction in corticosteroid tretinoin-treated perilesional skin. These data indicate that the addition of tretinoin does not impair the efficacy of a topical corticosteroid, in the treatment of psoriasis, and partially ameliorates epidermal atrophy produced by the topical corticosteroid.Peer Reviewedhttp://deepblue.lib.umich.edu/bitstream/2027.42/75727/1/j.1365-2133.1996.d01-933.x.pd

    Immunogenicity of a universal HIV-1 vaccine vectored by DNA, MVA and CHADV-63 in a Phase I/IIA clinical trial

    Background The major challenge facing both antibody and T cell-eliciting vaccines against HIV-1 is the extreme variability of the HIV-1 genome: a successful vaccine has to effectively target diverse HIV-1 strains circulating in the population and then must deal with ongoing virus escape in infected individuals. To address these issues, we assembled a vaccine immunogen HIVconsv from the functionally most conserved regions (not epitopes) of the HIV-1 proteome. Methods A gene coding for the HIVconsv immunogen was inserted into plasmid DNA (D), modified vaccinia virus Ankara (MVA; M) and non-replicating adenovirus of a chimpanzee origin ChAdV-63 (C). Currently, combined heterologous prime-boost regimens of these vaccines, namely CM, DDDCM and DDDMC, are being evaluated in a phase I/IIa trial HIV-CORE002 in healthy HIV-1/2-negative volunteers in Oxford. Results Preliminary data indicate that the vaccines are well tolerated and show high immunogenicity. Following the CM regimen, vaccine-induced T cell frequencies reached a median of 5150 (range 1475 to 16495) SFU/106 PMBC ex vivo one week post MVA vaccination. DNA priming increased subsequent T cell responses to ChAdV-63 vaccination (median: C 577 and DDDC 1328 SFU/106 PBMC) and ELISpot responses again peaked 1 week following MVA (median 4500; range 2260-7960 SFU/106 PBMC). Matrix analyses of the participants following CM vaccination showed that T cells responded to a range of peptides across the length of HIVconsv. The CM regimen elicited IFN-γ in both CD4+ and CD8+ T cell subsets and polyfunctional (IFN-γ & TNF-α) responses to HIVconsv peptides. Conclusion Presented data will be very much work in progress. Nevetheless, the HIVconsv vaccines have so far induced T cell responses superior to other HIV-1 vaccine candidates tested to date. ChAdV-63 is the first adenovirus of chimp origin delivering an HIV-1-derived immunogen that has reached the clinic. The work is supported by Medical Research Council UK
