    Lipid Markers and Compound-Specific Carbon Isotopes as Diet and Biosynthesis Reflectors in the Northern Neptune Whelk \u3ci\u3eNeptunea heros\u3c/i\u3e

    A suite of lipid biomarkers plus compound-specific carbon isotopes of major sterols were determined in muscle tissues across increasing sizes of northern Neptune whelks Neptunea heros, developing eggs and potential diets to link trophic patterns, metabolism and carbon sources on the Chukchi Sea shelf. Analysis of primary prey included the northern clam Astarte borealis, water column particulate organic matter (POM) and surface sediments near the collection sites. Sterols specific to major algal groups along with algal-derived polyunsaturated fatty acids (C20:5n-3, C20:4n-3, C22:6n-3) in whelk muscle tissue reflected the importance of algal primary production to benthic consumers and its direct incorporation. Compound-specific carbon isotope values of cholesterol and Δ5,7-sterols present in foot muscle of N. heros also suggest a transition from scavenging/detrital feeding in smaller juveniles to predatory consumption by larger adults. Sexually mature adult female N. heros (shell lengths \u3e10 cm) contained lower sterol concentrations compared to smaller whelks, which may reflect reduced consumption rates or lipid translocation during the annual reproductive cycle. Analysis of N. heros eggs at 3 developmental stages ranging from internal egg masses to mature egg cases showed significant transfer of algal-derived sterols and fatty acids which were conserved during egg maturation. The suite of individual lipid signatures and δ13C sterol values observed in N. heros suggest that the northern Neptune whelk relies on both direct lipid incorporation as well as metabolic modification of algal-derived food sources for sterol requirements that transition as animals mature and become predatory consumers

    Importance of the pleckstrin homology domain of dynamin in clathrin-mediated endocytosis

    AbstractThe GTPase dynamin plays an essential role in clathrin-mediated endocytosis [1–3]. Substantial evidence suggests that dynamin oligomerisation around the necks of endocytosing vesicles and subsequent dynamin-catalysed GTP hydrolysis is responsible for membrane fission [4,5]. The pleckstrin homology (PH) domain of dynamin has previously been shown to interact with phosphoinositides, but it has not been determined whether this interaction is essential for dynamin's function in endocytosis [6–9]. In this study, we address the in vivo function of the PH domain of dynamin by assaying the effects of deletions and point mutations in this region on transferrin uptake in COS-7 fibroblasts. Overexpression of a dynamin construct lacking its entire PH domain potently blocked transferrin uptake, as did overexpression of a dynamin construct containing a mutation in the first variable loop of the PH domain. Structural modelling of this latter mutant suggested that the lysine residue at position 535 (Lys535) may be critical in the coordination of phosphoinositides, and indeed, the purified mutant no longer interacted with lipid nanotubes. Interestingly, the inhibitory phenotype of cells expressing this dynamin mutant was partially relieved by a second mutation in the carboxy-terminal proline-rich domain (PRD), one that prevents dynamin from binding to the Src homology 3 (SH3) domain of amphiphysin. These data demonstrate that dynamin's interaction with phosphoinositides through its PH domain is essential for endocytosis. These findings also support our hypothesis that PRD–SH3 domain interactions are important in the recruitment of dynamin to sites of endocytosis

    A deep optical/near-infrared catalog of Serpens

    We present a deep optical/near-infrared imaging survey of the Serpens molecular cloud. This survey constitutes the complementary optical data to the Spitzer "Core To Disk" (c2d) Legacy survey in this cloud. The survey was conducted using the Wide Field Camera at the Isaac Newton Telescope. About 0.96 square degrees were imaged in the R and Z filters, covering the entire region where most of the young stellar objects identified by the c2d survey are located. 26524 point-like sources were detected in both R and Z bands down to R=24.5 mag and Z=23 mag with a signal-to-noise ratio better than 3. The 95% completeness limit of our catalog corresponds to 0.04 solar masses for members of the Serpens star forming region (age 2 Myr and distance 260 pc) in the absence of extinction. Adopting the typical extinction of the observed area (Av=7 mag), we estimate a 95% completeness level down to 0.1 solar masses. The astrometric accuracy of our catalog is 0.4 arcsec with respect to the 2MASS catalog. Our final catalog contains J2000 celestial coordinates, magnitudes in the R and Z bands calibrated to the SDSS photometric system and, where possible, JHK magnitudes from 2MASS for sources in 0.96 square degrees in the direction of Serpens. This data product has been already used within the frame of the c2d Spitzer Legacy Project analysis in Serpens to study the star/disk formation and evolution in this cloud; here we use it to obtain new indications of the disk-less population in Serpens.Comment: 7 page, 5 figure

    Endophilin drives the fast mode of vesicle retrieval in a ribbon synapse

    Compensatory endocytosis of exocytosed membrane and recycling of synaptic vesicle components is essential for sustained synaptic transmission at nerve terminals. At the ribbon-type synapse of retinal bipolar cells, manipulations expected to inhibit the interactions of the clathrin adaptor protein complex (AP2) affect only the slow phase of endocytosis (Ď„ = 10-15 s), leading to the conclusion that fast endocytosis (Ď„ = 1-2 s) occurs by a mechanism that differs from the classical pathway of clathrin-coated vesicle retrieval from the plasma membrane. Here we investigate the role of endophilin in endocytosis at this ribbon synapse. Endophilin A1 is a synaptically enriched N-BAR domain-containing protein, suggested to function in clathrin-mediated endocytosis. Internal dialysis of the synaptic terminal with dominant-negative endophilin A1 lacking its linker and Src homology 3 (SH3) domain inhibited the fast mode of endocytosis, while slow endocytosis continued. Dialysis of a peptide that binds endophilin SH3 domain also decreased fast retrieval. Electron microscopy indicated that fast endocytosis occurred by retrieval of small vesicles in most instances. These results indicate that endophilin is involved in fast retrieval of synaptic vesicles occurring by a mechanism that can be distinguished from the classical pathway involving clathrin-AP2 interactions

    An interdisciplinary approach to enhance children’s listening, learning, and wellbeing in the classroom : The Listen to Learn for Life (L3) Assessment Framework

    Introduction: Listening is the gateway to children learning in the mainstream classroom. However, modern classrooms are noisy and dynamic environments making listening challenging. It is therefore critical for researchers from speech and hearing, education, and health sciences to co-design and collaborate to realistically assess how children listen to learn in the classroom and to understand how listening can be improved to enhance children’s learning and wellbeing – an understanding which is currently lacking. Such highly interdisciplinary thinking demands a holistic classroom listening framework that can integrate a range of varied assessments and outcomes. Methods: An extensive review of literature into classroom listening was conducted but failed to identify a suitable framework. In this hypothesis and theory article we present a new framework that we have developed – the Listen to Learn for Life (L3) Assessment Framework. Results: The L3 Assessment Framework holistically incorporates frameworks from health, speech and hearing sciences, and education sectors. The framework accommodates a broad range of different factors that may affect listening, allowing for researchers to choose specific factors dependent on the context of use. Discussion: Selected examples of applying the framework are provided demonstrating how to assess children’s performance during different classroom activities as well as the effectiveness of a chosen intervention. For example, the framework can be used to assess the effectiveness of a wireless remote microphone intervention during group work activities for a child with autism. Conclusion: The L3 Assessment Framework provides a theoretical basis for the future development of research and practice as applied to listening in a classroom setting
