    Saving Culture Through Language: A Hermeneutic Phenomenological Study of Ojibwe Language Immersion Educator Experience

    With the near extinction of many tribal languages at the present, language immersion education offers considerable promise for the restoration of Ojibwe culture and identity. Through a series of structured interviews and longitudinal text-based dialogue with the researcher, eight educators from three school-based programs described the lived practice of working with culture in language immersion education. The study principally revealed how the lived experience of Ojibwe language immersion educators is important, challenging, and rewarding. The dynamic synergy of culture, language, and content that happens in Ojibwe language immersion requires competent and creative educators who are knowledgeable about language, cultural traditions, and teaching practice. The study concludes by noting how the lived experience of immersion school practitioners as cultural workers is marked not only by excellence in practice and professionalism, but by dedication, responsibility, and hope for the future of the culture, language and tribal nation itself

    Thermal Physics Resources on the Web

    In 1995 Dr Fekete was employed by McInnes and Walker as part of a CAUT project, “Diagnostic Tools for Concept Development”, to develop a practical and accessible database of resources that would encourage deep learning through interactive teaching in large class environments to be used in a first year lecture course on Thermal Physics. The web was chosen (http://www.physics.usyd.edu.au/teach/thermal.html) as the preferred choice of delivery as it was the most suitable medium to disseminate information to a large international body of academics. The material was specifically designed for large lecture theatre environments and included: ‱ information on how to use demonstrations interactively; ‱ a bank of questions designed to promote deeper conceptual understanding; ‱ references to research literature; and ‱ information on CD, video and film resources.     Functional characterization and identification of mouse Rad51d splice variants

    <p>Abstract</p> <p>Background</p> <p>The homologous recombination (HR) pathway is vital for maintaining genomic integrity through the restoration of double-stranded breaks and interstrand crosslinks. The RAD51 paralogs (RAD51B, RAD51C, RAD51D, XRCC2, XRCC3) are essential for this process in vertebrates, and the RAD51D paralog is unique in that it participates in both HR repair and telomere maintenance. RAD51D is also known to directly interact with the RAD51C and XRCC2 proteins. <it>Rad51d </it>splice variants have been reported in mouse and human tissues, supportive of a role for alternative splicing in HR regulation. The present study evaluated the interaction of the <it>Rad51d </it>splice isoform products with RAD51C and XRCC2 and their expression patterns.</p> <p>Results</p> <p>Yeast-2-hybrid analysis was used to determine that the <it>Mus musculus Rad51d </it>splice variant product RAD51DΔ7b (deleted for residues 219 through 223) was capable of interacting with both RAD51C and XRCC2 and that RAD51D+int3 interacted with XRCC2. In addition, the linker region (residues 54 through 77) of RAD51D was identified as a region that potentially mediates binding with XRCC2. Cellular localization, detected by EGFP fusion proteins, demonstrated that each of the splice variant products tested was distributed throughout the cell similar to the full-length protein. However, none of the splice variants were capable of restoring resistance of <it>Rad51d</it>-deficient cell lines to mitomycin C. RT-PCR expression analysis revealed that <it>Rad51dΔ3 </it>(deleted for exon 3) and <it>Rad51dΔ5 </it>(deleted for exon 5)transcripts display tissue specific expression patterns with <it>Rad51dΔ3 </it>being detected in each tissue except ovary and <it>Rad51dΔ5 </it>not detected in mammary gland and testis. These expression studies also led to the identification of two additional <it>Rad51d </it>ubiquitously expressed transcripts, one deleted for both exon 9 and 10 and one deleted for only exon 10.</p> <p>Conclusion</p> <p>These results suggest <it>Rad51d </it>alternative splice variants potentially modulate mechanisms of HR by sequestering either RAD51C or XRCC2.</p

    University students’ conceptions about familiar thermodynamic processes and the implications for instruction

    A large proportion of research in science education is either centred on or influenced by studies concerning conceptual change - in particular, the topic of students’ misconceptions. This is justified by the observation that studies involving conceptual change or troublesome knowledge capture an aspect of science education that seems to be extremely significant for successful learning and vital for developments in instructional methods. This paper is an examination of conceptions about fundamental thermodynamic concepts held by university students. A ‘pre-test’ was developed and administered to 858 first year and 80 second year university students to probe conceptions and inform a subsequent study. Questions included both multiple choice and free response types. The results indicate that the first year students experienced varied and considerable difficulties with the thermodynamic concepts presented in the pre-test, particularly with respect to heat transfer and thermal equilibrium. It is significant to note that these particular concepts appear as part of formal instruction in science in NSW, and that they are embedded in familiar everyday situations.     Deletion of the Androgen Receptor in Adipose Tissue in Male Mice Elevates Retinol Binding Protein 4 and Reveals Independent Effects on Visceral Fat Mass and on Glucose Homeostasis

    Testosterone deficiency is epidemic in obese ageing males with type 2 diabetes, but the direction of causality remains unclear. Testosterone-deficient males and global androgen receptor (AR) knockout mice are insulin resistant with increased fat, but it is unclear whether AR signaling in adipose tissue mediates body fat redistribution and alters glucose homoeostasis. To investigate this, mice with selective knockdown of AR in adipocytes (fARKO) were generated. Male fARKO mice on normal diet had reduced perigonadal fat but were hyperinsulinemic and by age 12 months, were insulin deficient in the absence of obesity. On high-fat diet, fARKO mice had impaired compensatory insulin secretion and hyperglycemia, with increased susceptibility to visceral obesity. Adipokine screening in fARKO mice revealed a selective increase in plasma and intra-adipose retinol binding protein 4 (RBP4) that preceded obesity. AR activation in murine 3T3 adipocytes downregulated RBP4 mRNA. We conclude that AR signaling in adipocytes not only protects against high-fat diet–induced visceral obesity but also regulates insulin action and glucose homeostasis, independently of adiposity.     Aromatase Inhibition Reduces Insulin Sensitivity in Healthy Men

    CONTEXT: Deficiency of aromatase, the enzyme that catalyzes the conversion of androgens to estrogens, is associated with insulin resistance in humans and mice. OBJECTIVE: We hypothesized that pharmacological aromatase inhibition results in peripheral insulin resistance in humans. DESIGN: This was a double-blind, randomized, controlled, crossover study. SETTING: The study was conducted at a clinical research facility. PARTICIPANTS: Seventeen healthy male volunteers (18–50 y) participated in the study. INTERVENTION: The intervention included oral anastrozole (1 mg daily) and placebo, each for 6 weeks with a 2-week washout period. MAIN OUTCOME MEASURE: Glucose disposal and rates of lipolysis were measured during a stepwise hyperinsulinemic euglycemic clamp. Data are mean (SEM). RESULTS: Anastrozole therapy resulted in significant estradiol suppression (59.9 ± 3.6 vs 102.0 ± 5.7 pmol/L, P = < .001) and a more modest elevation of total T (25.8 ± 1.2 vs 21.4 ± 0.7 nmol/L, P = .003). Glucose infusion rate, during the low-dose insulin infusion, was lower after anastrozole administration (12.16 ± 1.33 vs 14.15 ± 1.55 ÎŒmol/kg·min, P = .024). No differences in hepatic glucose production or rate of lipolysis were observed. CONCLUSION: Aromatase inhibition reduces insulin sensitivity, with respect to peripheral glucose disposal, in healthy men. Local generation and action of estradiol, at the level of skeletal muscle, is likely to be an important determinant of insulin sensitivity

    Secukinumab immunogenicity over 52 weeks in patients with psoriatic arthritis and ankylosing spondylitis

    Objective. Secukinumab, a fully human antiinterleukin 17A monoclonal antibody, is efficacious for the treatment of psoriatic arthritis (PsA) and ankylosing spondylitis (AS). This study examined the immunogenicity of secukinumab in patients with PsA and AS exposed to secukinumab for up to 52 weeks. Methods. Antibody bridging assays were used to assess the immunogenicity of secukinumab in patients with PsA [FUTURE 1–3 studies, and AS (MEASURE 1–4 studies)]. Evaluations were at baseline and at weeks 16 (AS only), 24, and 52. Treatment-emergent antidrug antibodies (TE-ADA) were defined as a positive ADA signal in ≄ 1 posttreatment sample in patients negative at baseline. Positive samples were analyzed for drug-neutralizing potential, and effect of TE-ADA on secukinumab pharmacokinetics, immunogenicity-related adverse events (AE), and efficacy through Week 52 were assessed. Results. Of 1414 treated PsA and 1164 treated AS patients with samples available for immunogenicity evaluation, 5 (0.35%) and 8 (0.69%), respectively, developed TE-ADA. All but 1 PsA patient were biologic-naive; two of the 5 PsA and one of the 8 AS patients received concomitant methotrexate, and two of the 8 AS patients received concomitant sulfasalazine. Associations between TE-ADA and secukinumab dose, frequency, or administration mode were not observed. Other than one PsA patient, all TE-ADA were non-neutralizing. No TE-ADA were associated with any AE. All TE-ADA were associated with normal secukinumab pharmacokinetics and none were associated with loss of secukinumab efficacy. Conclusion. Secukinumab treatment was associated with a low (&lt; 1%) incidence of immunogenicity in patients with PsA or AS.     For many users of the biomedical literature, abstracts may be the only source of information about a study. Hence, abstracts should allow readers to evaluate the objectives, key design features, and main results of the study. Several evaluations have shown deficiencies in the reporting of journal and conference abstracts across study designs and research fields, including systematic reviews of diagnostic test accuracy studies. Incomplete reporting compromises the value of research to key stakeholders. The authors of this article have developed a 12 item checklist of preferred reporting items for journal and conference abstracts of systematic reviews and meta-analyses of diagnostic test accuracy studies (PRISMA-DTA for Abstracts). This article presents the checklist, examples of complete reporting, and explanations for each item of PRISMA-DTA for Abstracts

    Life-history trait variation in a queen-size dimorphic ant

    1. Size polymorphism is often connected to alternative life-history traits, which may eventually lead to distinct size classes. In the ant Myrmica ruginodis, larger macrogyne and smaller microgyne queen morphs have been suggested to follow different reproductive strategies, which has presumably resulted in several differences in their key life-history traits. 2. In this study, we examine the association of queen-size morphs with colony queen number (monogyny vs. polygyny), dispersal and queen recruitment patterns, as well as habitat associations of the queen morphs. We do this by sampling established queens from a large number of excavated nests from several populations, estimating genetic relatedness among coexisting queens and pitfall trapping free-ranging wingless queens. 3. Our results show that associations of queen morphs with colony queen number and nest-founding strategy holds only partly. The morph frequencies vary widely across populations from practically pure macrogyne to more than 50% microgyne, but the expected association of macrogyne occurrence with monogyny and microgyne with polygyny is not universal. Dispersal and queen recruitment patterns also show that although most macrogynes participate in nuptial flights and most microgynes are recruited back to their natal nests, a fraction of both morphs use the alternative strategy. 4. The polygynous microgyne morph has been suggested to specialize in stable habitats, but our results from Finnish mesic heath forests do not support this. This study shows that factors other than just queen size also influence life-history trait variation and reproductive strategies in ants.Peer reviewe
