    Micro- and macrofossil data from the remains of forests overwhelmed and buried at Pureora and Benneydale during the Taupo eruption (c. 1850 conventional radiocarbon yr BP) were compared. Classification of relative abundance data separated the techniques, rather than the locations, because the two primary clusters comprised pollen and litter/wood. This indicates that the pollen:litter/wood within-site comparisons (Pureora and Benneydale are 20 km apart) are not reliable. Plant macrofossils represented mainly local vegetation, while pollen assemblages represented a combination of local and regional vegetation. However, using ranked abundance and presence/absence data, both macrofossils and pollen at Pureora and Benneydale indicated conifer/broadleaved forest, of similar forest type and species composition at each site. This suggests that the forests destroyed by the eruption were typical of mid-altitude west Taupo forests, and that either data set (pollen or macrofossils) would have been adequate for regional forest interpretation. The representation of c. 1850 yr BP pollen from the known buried forest taxa was generally consistent with trends determined by modern comparisons between pollen and their source vegetation, but with a few exceptions. A pollen profile from between the Mamaku Tephra (c. 7250 yr BP) and the Taupo Ignimbrite indicated that the Benneydale forest had been markedly different in species dominance compared with the forest that was destroyed during the Taupo eruption. These differences probably reflect changes in drainage, and improvements in climate and/or soil fertility over the middle Holocene

    UVB-Induced Microvesicle Particle Release in Human Skin in vivo is Diminished Following Oral Vitamin C and E Antioxidant Administration

    An important question in photobiology asks how Ultraviolet B (UVB, 290 – 320 nm) radiation, which mostly absorbs in the outer epidermis of skin, can generate a systemic response such as immunosuppression. Previous in vitro and ex vivo studies demonstrate UVB-dependent release of bioactive molecule-containing microvesicle particles (MVPs) from keratinocytes. Furthermore, MVP release is diminished upon antioxidant administration. The purpose of this study is to examine UVB-induced MVP release and antioxidant response in vivo. In this IRB-approved study, 8 male participants with Fitzpatrick type I or II skin were treated with 1000 J/m2 UVB irradiation to a 5 by 5 mm area of volar forearm skin. 4 hours later, punch biopsies and erythema measurements were performed. This procedure was repeated 8 days later following a course of oral antioxidants (vitamin C 2g/day, vitamin E 1000IU/day). On average, tissue MVP release increased 1.8-fold (+/- 0.31, P = 0.02) following UVB treatment. Following a course of oral antioxidants, the average UVB-induced tissue MVP release did not differ from the control (0.9-fold +/- 0.13, P = 0.23). There was no significant change in UVB-induced erythema between pre- and post-antioxidant administration. These studies suggest that UVB-MVP are dependent upon reactive oxygen species

    Effectiveness of a social support intervention on infant feeding practices : randomised controlled trial

    Background: To assess whether monthly home visits from trained volunteers could improve infant feeding practices at age 12 months, a randomised controlled trial was carried out in two disadvantaged inner city London boroughs. Methods: Women attending baby clinics with their infants (312) were randomised to receive monthly home visits from trained volunteers over a 9-month period (intervention group) or standard professional care only (control group). The primary outcome was vitamin C intakes from fruit. Secondary outcomes included selected macro and micro-nutrients, infant feeding habits, supine length and weight. Data were collected at baseline when infants were aged approximately 10 weeks, and subsequently when the child was 12 and 18 months old. Results: Two-hundred and twelve women (68%) completed the trial. At both follow-up points no significant differences were found between the groups for vitamin C intakes from fruit or other nutrients. At first follow-up, however, infants in the intervention group were significantly less likely to be given goats’ or soya milks, and were more likely to have three solid meals per day. At the second follow-up, intervention group children were significantly less likely to be still using a bottle. At both follow-up points, intervention group children also consumed significantly more specific fruit and vegetables. Conclusions: Home visits from trained volunteers had no significant effect on nutrient intakes but did promote some other recommended infant feeding practices

    Hepatocyte cholesterol content modulates glucagon receptor signalling

    Objective To determine whether glucagon receptor (GCGR) actions are modulated by cellular cholesterol levels. Methods We determined the effects of experimental cholesterol depletion and loading on glucagon-mediated cAMP production, ligand internalisation and glucose production in human hepatoma cells, mouse and human hepatocytes. GCGR interactions with lipid bilayers were explored using coarse-grained molecular dynamic simulations. Glucagon responsiveness was measured in mice fed a high cholesterol diet with or without simvastatin to modulate hepatocyte cholesterol content. Results GCGR cAMP signalling was reduced by higher cholesterol levels across different cellular models. Ex vivo glucagon-induced glucose output from mouse hepatocytes was enhanced by simvastatin treatment. Mice fed a high cholesterol diet had increased hepatic cholesterol and a blunted hyperglycaemic response to glucagon, both of which were partially reversed by simvastatin. Simulations identified likely membrane-exposed cholesterol binding sites on the GCGR, including a site where cholesterol is a putative negative allosteric modulator. Conclusions Our results indicate that cellular cholesterol content influences glucagon sensitivity and indicate a potential molecular basis for this phenomenon. This could be relevant to the pathogenesis of non-alcoholic fatty liver disease, which is associated with both hepatic cholesterol accumulation and glucagon resistance

    The C-tactile (CLTM) peripheral nervous system is involved in social bonding in primates and humans through its capacity to trigger the brain’s endorphin system. Since the mammalian cochlea has an unusually high density of similar neurons (type-II spiral ganglion neurons, SGNs), we hypothesise that their function may have been exploited for social bonding by co-opting head movements in response to music and other rhythmic movements of the head in social contexts. Music provides one of many cultural behavioural mechanisms for ‘virtual grooming’ in that it is used to trigger the endorphin system with many people simultaneously so as to bond both dyadic relationships and large groups. Changes in pain threshold across an activity are a convenient proxy assay for endorphin uptake in the brain, and we use this, in two experiments, to show that pain thresholds are higher when nodding the head than when sitting still

    Seeded crystallization of β-L-glutamic acid in a continuous oscillatory baffled crystallizer

    A continuously seeded l-glutamic acid cooling crystallization process, in a continuous oscillatory baffled crystallizer, was designed and operated to deliver control over polymorphic form. Different feed solution concentrations and seed loadings of β-l-glutamic acid crystals were examined. Steady-state operation, based on particle size distribution and polymorphic form, was demonstrated consistently after two residence times. Where bulk supersaturation remained in the range 2–3, the polymorphic phase purity of the thermodynamically stable β polymorph was retained. However, when the bulk supersaturation exceeded this range to values of 3–8, primary nucleation of the metastable α polymorph was observed, and product crystals were a mixed phase. In the absence of seeding the system could not be operated without significant encrustation to the vessel surface thus leading to loss of control, whereas a continuously seeded approach allowed robust processing for at least 10 h
