    The Residual Effect of Novice Primary Teachers on Reading Achievement Scores

    This dissertation was an exploratory investigation of the residual effect of novice primary teachers on reading achievement scores. The study employed the theory of pedagogical content knowledge as a basis for understanding teacher expertise and comparing the expert teacher to the novice teacher. The research sought to answer two major questions; (a) Is there a statistically significant difference between the reading achievement, (measured two years later) of students taught by teachers of differing experience levels in primary grades and the state mean for the appropriate grade level? and (b) Is there a statistically significant difference among the two-year-later reading achievement of the groups of students based on teacher experience levels? This study used student reading achievement test scores from the CTBS/5 Achievement Test published by CTB/McGraw Hill. Data were analyzed using the Statistical Package for the Social Sciences (SPSS) using a series of nine Mests and three analysis of variance tests (ANOVAs). While the findings of this study indicated that there were no statistically significant differences among the groups, the author discussed several limitations to the study. In addition, proposals for future research in the area were presented.     STAINING OF OVCA1 ANTIBODY IN HUMAN MALIGNANCIES

    poster abstractImmunohistochemistry biomarkers are currently being developed to tar-get specific proteins found in cancer cells. The biomarker and putative tumor suppressor, OvCa1, has a function that is not well characterized. Due to lack of reagents, we developed monoclonal antibodies of OvCa1 to examine mul-tiple human malignancies. Primary cancers with different histologic grades as well as with metastatic lesions were examined with the monoclonal anti-bodies. Ovarian cancer tissue samples from the IU Simon Cancer Center Tis-sue Bank were used for this study. The samples were fixed in neutral buff-ered formalin and processed into a paraffin block. The slides were microtomed, and immunohistochemistry (IHC) with the OvCa1 antibody was performed. Thirty-one low, medium, and high grade tumors as well as meta-static ovarian carcinomas were evaluated. All cases revealed a range of staining intensity with OvCa1. The results indicated that OvCa1 had the highest immunostaining in the high grade, Stage 3 to 4 ovarian carcinomas. Medium grade tumors had less OvCa1 expression, while the metastatic tu-mors had less staining than any of the other three grades. Immunostaining was observed primarily in the cytoplasm and nucleus of the tumor cells. In addition, we evaluated approximately 20 tumors from various different or-gans. These included prostate, breast, spleen, lung, colon, stomach, and kidney tumors, which were positive for immunostaining with the OvCa1 anti-body. In summary, the results indicate that all histologic grades express the biomarker, OvCa1, and the staining intensity was highest in the high grade, Stage 3 and 4 tumors. Our preliminary studies demonstrate a further need to delineate OvCa1 as a potential biomarker, which could be used for early detection and diagnosis of ovarian cancer

    Evaluation of the Electronic Clinical Dementia Rating for dementia screening

    IMPORTANCE: The Clinical Dementia Rating (CDR) is a well-validated instrument widely used to detect and stage dementia due to Alzheimer disease. The digital Electronic Clinical Dementia Rating (eCDR) can be remotely self-administered and automatically scored, with potential to facilitate efficient dementia screening and staging. OBJECTIVE: To evaluate the association of the eCDR with the CDR and other in-clinic assessments for screening older adults for cognitive impairment. DESIGN, SETTING, AND PARTICIPANTS: This multisite, cross-sectional study used baseline data from a longitudinal, observational study from 2020 to 2023, including up to 3 years of follow-up. Participants were enrolled from 3 Alzheimer Disease Research Centers and the Brain Health Registry. Participants (aged ≥55 years, with a study partner, and no acute or unstable major medical conditions) were recruited during in-clinic visits or by automated emails. EXPOSURES: Participants completed the Uniform Data Set Version 3 (UDS; including the CDR) in supervised clinical research settings, and then completed the eCDR remotely, online and unsupervised, using their own device. MAIN OUTCOMES AND MEASURES: The primary outcomes were eCDR scores (item; categorical box and global; continuous box and global), CDR scores (item; categorical box and global), and UDS assessment scores. Associations were evaluated using linear and logistic regressions. RESULTS: A total of 3565 participants were contacted, and 288 were enrolled. Among 173 participants with item-level data (mean [SD] age, 70.84 [7.65] years; 76 women [43.9%]), eCDR to CDR concordance was 90% or higher for 33 items (63%) and 70% to 89% for 13 items (25%). Box (domain) level concordance ranged from 80% (memory) to 99% (personal care). The global score concordance rate was 81%. κ statistics were fair to moderate. Among 206 participants with box and global scores (mean [SD] age, 71.34 [7.68] years; 95 women [46.1%]), eCDR continuous global score was associated with CDR global (categorical) score with an area under the receiver operating characteristic curve of 0.79 (95% CI, 0.70-0.87). Correlations between eCDR and in-clinic UDS assessments were similar to those between CDR sum of box scores and the same in-clinic assessments. CONCLUSIONS AND RELEVANCE: These findings suggest that the eCDR is valid and has potential use for screening and assessment of older adults for cognitive and functional decline related to Alzheimer disease. Instrument optimization and validation in diverse cohorts in remote settings are crucial for evaluating scalability and eCDR utility in clinical research, trials, and health care settings

    Protein stability engineering insights revealed by domain-wide comprehensive mutagenesis

    The accurate prediction of protein stability upon sequence mutation is an important but unsolved challenge in protein engineering. Large mutational datasets are required to train computational predictors, but traditional methods for collecting stability data are either low-throughput or measure protein stability indirectly. Here, we develop an automated method to generate thermodynamic stability data for nearly every single mutant in a small 56-residue protein. Analysis reveals that most single mutants have a neutral effect on stability, mutational sensitivity is largely governed by residue burial, and unexpectedly, hydrophobics are the best tolerated amino acid type. Correlating the output of various stability-prediction algorithms against our data shows that nearly all perform better on boundary and surface positions than for those in the core and are better at predicting large-to-small mutations than small-to-large ones. We show that the most stable variants in the single-mutant landscape are better identified using combinations of 2 prediction algorithms and including more algorithms can provide diminishing returns. In most cases, poor in silico predictions were tied to compositional differences between the data being analyzed and the datasets used to train the algorithm. Finally, we find that strategies to extract stabilities from high-throughput fitness data such as deep mutational scanning are promising and that data produced by these methods may be applicable toward training future stability-prediction tools
