    Genome sequence of the oleaginous yeast Rhodotorula toruloides strain CGMCC 2.1609

Available from the publisher via the DOI in this record.Most eukaryotic oleaginous species are yeasts and among them the basidiomycete red yeast, Rhodotorula (Rhodosporidium) toruloides (Pucciniomycotina) is known to produce high quantities of lipids when grown in nitrogen-limiting media, and has potential for biodiesel production. The genome of the CGMCC 2.1609 strain of this oleaginous red yeast was sequenced using a hybrid of Roche 454 and Illumina technology generating 13 × coverage. The de novo assembly was carried out using MIRA and scaffolded using MAQ and BAMBUS. The sequencing and assembly resulted in 365 scaffolds with total genome size of 33.4 Mb. The complete genome sequence of this strain was deposited in GenBank and the accession number is LKER00000000.     Two-dimensional electrophoretic comparison of metastatic and non-metastatic human breast tumors using in vitro cultured epithelial cells derived from the cancer tissues

    <p>Abstract</p> <p>Background</p> <p>Breast carcinomas represent a heterogeneous group of tumors diverse in behavior, outcome, and response to therapy. Identification of proteins resembling the tumor biology can improve the diagnosis, prediction, treatment selection, and targeting of therapy. Since the beginning of the post-genomic era, the focus of molecular biology gradually moved from genomes to proteins and proteomes and to their functionality. Proteomics can potentially capture dynamic changes in protein expression integrating both genetic and epigenetic influences.</p> <p>Methods</p> <p>We prepared primary cultures of epithelial cells from 23 breast cancer tissue samples and performed comparative proteomic analysis. Seven patients developed distant metastases within three-year follow-up. These samples were included into a metastase-positive group, the others formed a metastase-negative group. Two-dimensional electrophoretical (2-DE) gels in pH range 4–7 were prepared. Spot densities in 2-DE protein maps were subjected to statistical analyses (R/maanova package) and data-mining analysis (GUHA). For identification of proteins in selected spots, liquid chromatography-tandem mass spectrometry (LC-MS/MS) was employed.</p> <p>Results</p> <p>Three protein spots were significantly altered between the metastatic and non-metastatic groups. The correlations were proven at the 0.05 significance level. Nucleophosmin was increased in the group with metastases. The levels of 2,3-trans-enoyl-CoA isomerase and glutathione peroxidase 1 were decreased.</p> <p>Conclusion</p> <p>We have performed an extensive proteomic study of mammary epithelial cells from breast cancer patients. We have found differentially expressed proteins between the samples from metastase-positive and metastase-negative patient groups.</p

    BUILDING FACADE DOCUMENTATION USING LASER SCANNING AND PHOTOGRAMMETRY AND DATA IMPLEMENTATION INTO BIM

    A project started last year called MORE-CONNECT, which focuses on the renovation of buildings (especially building facades) using prefabricated elements. The aim of this project is to create a competitive solution consisting of a technology and processes which enable fast, cost-effective renovation with minimal difficulties to inhabitants. Significant cost savings in renovation costs lies in the usage of prefabricated elements and the reduction of construction works on site. The precision of the prefabricated element depends on the precision of the construction, project and building documentation. This article offers an overview of the possible methods for building documentation and spatial data transfer into BIM (Building Information Modelling) software. The description of methods focuses on laser scanning and photogrammetry (including RPAS based), its advantages, disadvantages and limitations according to the documented building, level of renovation, situation on site etc. The next part involves spatial data transfer into BIM software. A proposed solution is tested in a case study

    The mature spermatozoon of Khawia armeniaca, a monozoic caryophyllidean parasite of templar fish Capoeta capoeta sevangi (De Filippi, 1865) from the Lake Sevan, Armenia, has been studied using transmission electron microscopy and cytochemical technique of Thiéry (1967) for the first time. The mature spermatozoon of K. armeniaca consists of a single axoneme with the 9+‘1’ trepaxonematan structure, cortical microtubules and nucleus which are situated parallel to the longitudinal axis of the spermatozoon, and a moderately electrondense cytoplasm with glycogen particles. The cortical microtubules are arranged in one continuous semicircle beneath the plasma membrane in Region II and anterior part of Region III of the mature spermatozoon. The two opposite rows of cortical microtubules are observed in the remaining nuclear and at the beginning of the postnuclear part (Regions III, IV) of the male gamete The number of cortical microtubules is remarkably variable in the spermatozoa of various Khawia species. K. armeniaca exhibits the highest number of cortical microtubules in comparison with K. sinensis and K. rossittensis. Glycogen was detected in the cytoplasm of prenuclear (II), nuclear (III) and postnuclear (IV) regions with different ultrastructural organization of the mature spermatozoon of K. armeniaca. Variations of sperm ultrastructural characters within caryophyllideans and other cestodes are discussed

    USING REMOTE SENSING AND RPAS FOR ARCHAEOLOGY AND MONITORING IN WESTERN GREENLAND

    Since 2002, German low-cost scientific expeditions to Greenland have been performed. The objective was a geodetic survey and glaciology with GNSS technology – mainly the measurement of glacier profiles (height). The same glacier profiles along the route were measured during German expeditions in 2006, 2010, 2012 and 2015. The last international expedition was supplemented with RPAS (UAV) measurement, the testing of small corner reflectors for Terra SAR X satellite measurement and the use of image based modelling technology for historical monuments documentation, all in specific arctic conditions. The RPAS measurement was focused on the documentation of existing valuable archaeological sites near Ilulissat city and the testing of RPAS technology for the monitoring of the face of the moving glacier. Two typical church wooden constructions were documented by simple photogrammetric technology based on image correlation. Both experiments were evaluated as successfully case projects. The last part of the experiments deals with the monitoring of a moving inland glacier using SAR technology; four corner reflectors were installed on the glacier and on a massive nearby rock. Two ascending and two descending overflights of the Terra SAR X satellite in fine resolution mode were performed. The InSAR technology give inconclusive results, but some movements were detected; small and inexpensive corner reflectors of our own production have proven suitable.     The Effect of Postoperative Pain Treatment on the Incidence of Anastomotic Insufficiency after Rectal and Rectosigmoideal Surgery

    The aim of prospective study was to evaluate the pain relief in the postoperative period and consumption of opioid and non-opioid analgesics as a risk factor of the anastomotic insufficiency after rectal and rectosigmoideal resection for carcinoma. Anastomotic insufficiency is one of the most feared and life threatening early complications. No articles about the effect of the response to opioid therapy in the postoperative period on the risk of this major clinical problem have been published. We compared the effect of opioid and non-opioid analgesics in 109 patients who underwent rectal and rectosigmoideal resection in a prospective study. We evaluated the appearance of anastomotic insufficiency and clinical conditions in the relationship with the pain relief in the postoperative period and consumption of opioid and non-opioid analgesics. The pain intensity and the consumption of analgesics were significantly increased in the group of nonresponders. The rate of PONV (postoperative nausea and vomiting) in the responders and nonresponders groups was 69% and 78%, respectively. However, the differences did not reach significant level. Other clinical conditions were not significantly different between the both groups, too. The difference in the incidence of anastomotic insufficiency between both groups was highly significant, 6% cases of anastomotic insufficiency in the responders group and 19% in nonresponders group (χ2 = 7.73; p=0.0054). Nonrespoders to opioid therapy and their high consumption of second-line analgesics is a high risk factor for anastomotic insufficiency
