    Structure determination of Split-soret Cytochrome from a Desulfovibrio species isolated from a human abdominal abcess

    The determined structure of the split-soret cytochrome (SSC) isolated from Desulfovibrio desulfuricans ATCC 27774 (D.d.) revealed a new Heme arrangement, which suggests that this protein constitutes a new cytochrome class.. SSC is a 52.6kDa homodimer containing four hemes at one end of the molecule. In each monomer the two hemes have their edges overlapped within van der Waals contacts. The polypeptide chain of each monomer supplies the sixth ligand to the heme-iron of the other monomer. A similar protein was recently purified from a homologous Desulfovibrio clinical strain isolated from an abdominal wall abscess in human patient2. Crystals of this SSC were grown using vapour diffusion method in the presence of agarose gel. Diffraction data were collected using X-ray synchrotron radiation at the ESRF, beamline, ID 14-1. The structure will be solved by molecular replacement using the structure of the D.d. as a starting model

However, this industry, due to its singularity, presents several challenges and disadvantages related to the transportation of raw materials, with the large volumes that are usually involved. This project aimed to address this internal logistics situation in torrefied biomass pellets production with two different biomass storage parks, located in Portugal. The main park receives raw material coming directly from the source and stores it in large amounts as a backup and strategic storage park. The second park, with smaller dimensions, precedes the production unit and must be stocked daily. Therefore, a fleet of transport units with self-unloading cranes is required to help to unload the biomass at the main park and transport the raw material from this park to the one preceding the production unit. Thus, the main goal was to determine the dimensions of the fleet used in internal transportation operations to minimize the idle time of the transport units using a methodology already in use in the mining and quarrying industry. This methodology was analyzed and adapted to the situation presented here. The implementation of this study allows the elimination of unnecessary costs in an industry where the profit margins are low.publishersversionpublishe

    Efeito de estresse salino no processo germinativo de sementes de angico (Anadenanthera colubrina).

    O objetivo deste trabalho foi avaliar a germinação de sementes de angico (Anadenanthera colubrina) sob diferentes níveis de estresse salino. A salinidade é considerada um dos principais fatores limitantes para crescimento das plantas, e a germinação e o crescimento inicial de plântulas são os estágios de desenvolvimento mais sensíveis à salinidade, sendo influenciados significativamente pela condição de salinidade do solo. Alta concentrações de sais, especialmente de cloreto de sódio (NaCl), pode inibir a germinação por causa da diminuição do potencial osmótico, o que ocasiona prejuízos às demais fases do processo. No presente experimento, as sementes de angico foram distribuídas em substrato papel Germitest, umedecido com cada uma das soluções salinas (0 dS.m-1; 2 dS.m-1; 4 dS.m-1; 8 dS.m-1; 12 dS.m-1; 16 dS.m-1 e 18 dS.m-1). Os rolos obtidos foram mantidos em incubadora B.O.D a 25 °C durante 10 dias. Foi constatada a tolerância das sementes de angico à salinidade obtendo na condutividade de até 8 dS.m-1 germinação superior a 90%

    TiO2 nanostructured films for electrochromic paper based-devices

    (H2020-NMP-2015, grant 685758-21D H2020 ERC AdG 787410)Electrochromic titanium dioxide (TiO2) nanostructured films were grown on gold coated papers using a microwave-assisted hydrothermal method at low temperature (80 °C). Uniform nanostructured films fully covered the paper substrate, while maintaining its flexibility. Three acids, i.e., acetic, hydrochloric and nitric acids, were tested during syntheses, which determined the final structure of the produced films, and consequently their electrochromic behavior. The structural characteristics of nanostructured films were correlated with electrochemical response and reflectance modulation when immersed in 1 M LiClO4-PC (lithium perchlorate with propylene carbonate) electrolyte, nevertheless the material synthesized with nitric acid resulted in highly porous anatase films with enhanced electrochromic performance. The TiO2 films revealed a notable contrast behavior, reaching for the nitric-based film optical modulations of 57%, 9% and 22% between colored and bleached states, at 250, 550 and 850 nm, respectively in reflectance mode. High cycling stability was also obtained performing up to 1500 cycles without significant loss of the electrochromic behavior for the nitric acid material. The approach developed in this work proves the high stability and durability of such devices, together with the use of paper as substrate that aggregates the environmentally friendly, lightweight, flexibility and recyclability characters of the substrate to the microwave synthesis features, i.e., simplicity, celerity and enhanced efficiency/cost balance.publishersversionpublishe

    Viability of dietary substitution of live microalgae with dry Ulva rigida in broodstock conditioning of the Pacific oyster (Crassostrea gigas)

    The current study evaluated the microalgae replacement by dry macroalgae (Ulva rigida) in the reproductive success and biochemical composition of the Pacific oyster (Crassostrea gigas) during broodstock conditioning. Five nutritional regimes were tested: 100% macroalgae (diet 1), 50% macroalgae+50% microalgae (diet 2), 25% macroalgae+75% microalgae (diet 3) and 100% microalgae (diet 4). An unfed group was used as a negative control. The microalgae blend was composed of 33% Isochrysis galbana and 67% diatoms (75% Skeletonema costatum+25% Chaetoceros calcitrans). Gonadal maturation was reflected in the physiological condition of the individuals. All treatments, except diet 1, showed an increase in condition index and were fully matured at the end of the trial, with the best physiological condition observed in oysters fed diet 3 and diet 4. Protein and total lipid content increased during the conditioning period, whereas glycogen content decreased. Oysters conditioned with diet 3 had higher protein and total lipid content and lower glycogen content than the other treatments. In addition, diet 3 showed the highest percentage of viable veliger larvae. The current study demonstrated that it is possible to replace 25% of microalgae with macroalgae in the broodstock conditioning, minimizing the operative cost in bivalve hatcheries. © 2018.     Caracterização e Análise dos Processos Produtivos em Empresas Agro-Alimentares

    Através de um diagnóstico inicial baseado na análise do conhecimento, informação existente, recolha de informação e medição de parâmetros num conjunto de empresas (60) dos subsetores dos produtos cárneos, hortofrutícolas, lácteos e panificação apresenta-se através do presente relatório a caracterização dos processos e atividades produtivas em empresas do setor agroalimentar. Executando uma análise técnica ao estado da arte, através do estudo da organização das atividades produtivas, nível tecnológico e tendências no desenvolvimento de produtos, permite-se de acordo com a informação disponível realizar uma comparação das empresas em análise com a performance da indústria, globalmente, por região ou setor de atividade. Verifica-se através da análise ao presente relatório, quais os fatores que permitem a criação de valor no setor agroindustrial de uma forma contínua para a cadeia de valor organizacional. Por outro lado, entre os diversos pontos a melhorar, considerando as organizações em análise, destaca-se a cultura de inovação e a digitalização dos processos de produção (Indústria 4.0). Pretende-se, com a aplicação prática do conhecimento gerado pelo presente relatório, a criação de ferramentas que constituam soluções para a melhoria da gestão da produção que promovam a introdução de métodos de inovação na cultura e performance organizacional de forma a tornar as organizações do setor competitivas e com impacto nos mercados internacionais
