    Teenage and adult speech in school context: building and processing a corpus of European Portuguese

    We present a corpus of European Portuguese spoken by teenagers and adults in school context, CPE-FACES, with an overview of the differential characteristics of high school oral presentations and the challenges this data poses to automatic speech processing. The CPE-FACES corpus has been created with two main goals: to provide a resource for the study of prosodic patterns in both spontaneous and prepared unscripted speech, and to capture inter-speaker and speaking style variations common at school, for research on oral presentations. Research on speaking styles is still largely based on adult speech. References to teenagers are sparse and cross-analyses of speech types comparing teenagers and adults are rare. We expect CPE-FACES, currently a unique resource in this domain, will contribute to filling this gap in European Portuguese. Focusing on disfluencies and phrase-final phonetic-phonological processes we show the impact of teenage speech on the automatic segmentation of oral presentations. Analyzing fluent final intonation contours in declarative utterances, we also show that communicative situation specificities, speaker status and cross-gender differences are key factors in speaking style variation at school.info:eu-repo/semantics/acceptedVersio

    A panel data approach towards the effectiveness of energy policies in fostering the implementation of solar photovoltaic technology: Empirical evidence for Asia-Pacific

    Today, the growing Asia-Pacific population causes a dramatic growth in energy supply to meet energy demand. The rapid rise in energy demand is causing concern in the region. Thus, the present study scrutinizes the effect of energy policy involvement in steering-up renewable energy development by empirically assessing the role of policy instruments in encouraging residen-tial-scale and commercial-scale photovoltaic (PV) systems. The analysis is performed using a fixed effects estimator on a selected range of policy approaches (market-pull policies and tax incentives) and a technology-push policy (capital grants) in selected Asia-Pacific countries between 1998 and 2015. The return on investment is estimated to measure the incentives of feed-in tariff (FIT) tariff policies for both residential-scale and commercial-scale PV systems. This study has shown the im-portance of a strategic combination between technology-push and market-pull policies as comple-mentary to adopting technology and increasing renewable energy utilization for solar PV systems on a residential scale.     Connective tissue progenitor cell growth characteristics on textured substrates

    Growth characteristics of human connective tissue progenitor (CTP) cells were investigated on smooth and textured substrates, which were produced using MEMS (microelectromechanical systems) fabrication technology. Human bone marrow derived cells were cultured for 9 days under conditions promoting osteoblastic differentiation on polydimethylsiloxane (PDMS) substrates comprising smooth (non-patterned) surfaces (SMOOTH), 4 different cylindrical post micro-textures (POSTS) that were 7–10 μm high and 5, 10, 20, and 40 μm diameter, respectively, and channel micro-textures (CHANNELS) with curved cross-sections that were 11 μm high, 45 μm wide, and separated by 5 μm wide ridges. Standard glass-tissue culture surfaces were used as controls. Micro-textures resulted in the modification of CTP morphology, attachment, migration, and proliferation characteristics. Specifically, cells on POSTS exhibited more contoured morphology with closely packed cytoskeletal actin microfilaments compared to the more random orientation in cells grown on SMOOTH. CTP colonies on 10 μm-diameter POSTS exhibited higher cell number than any other POSTS, and a significant increase in cell number (442%) compared to colonies on SMOOTH (71%). On CHANNELS, colonies tended to be denser (229%) than on POSTS (up to 140% on 10 μm POSTS), and significantly more so compared to those on SMOOTH (104%)

    Cross-domain analysis of discourse markers in European Portuguese

    This paper presents an analysis of discourse markers in two spontaneous speech corpora for European Portuguese - university lectures and map-task dialogues - and also in a collection of tweets, aiming at contributing to their categorization, scarcely existent for European Portuguese. Our results show that the selection of discourse markers is domain and speaker dependent. We also found that the most frequent discourse markers are similar in all three corpora, despite tweets containing discourse markers not found in the other two corpora. In this multidisciplinary study, comprising both a linguistic perspective and a computational approach, discourse markers are also automatically discriminated from other structural metadata events, namely sentence-like units and disfluencies. Our results show that discourse markers and disfluencies tend to co-occur in the dialogue corpus, but have a complementary distribution in the university lectures. We used three acoustic-prosodic feature sets and machine learning to automatically distinguish between discourse markers, disfluencies and sentence-like units. Our in-domain experiments achieved an accuracy of about 87% in university lectures and 84% in dialogues, in line with our previous results. The eGeMAPS features, commonly used for other paralinguistic tasks, achieved a considerable performance on our data, especially considering the small size of the feature set. Our results suggest that turn-initial discourse markers are usually easier to classify than disfluencies, a result also previously reported in the literature. We conducted a cross-domain evaluation in order to evaluate the robustness of the models across domains. The results achieved are about 11%-12% lower, but we conclude that data from one domain can still be used to classify the same events in the other. Overall, despite the complexity of this task, these are very encouraging state-of-the-art results. Ultimately, using exclusively acoustic-prosodic cues, discourse markers can be fairly discriminated from disfluencies and SUs. In order to better understand the contribution of each feature, we have also reported the impact of the features in both the dialogues and the university lectures. Pitch features are the most relevant ones for the distinction between discourse markers and disfluencies, namely pitch slopes.     Trapping and Characterization of a Reaction Intermediate in Carbapenem Hydrolysis by \u3cem\u3eB. cereus\u3c/em\u3e Metallo-β-lactamase

    Metallo-β-lactamases hydrolyze most β-lactam antibiotics. The lack of a successful inhibitor for them is related to the previous failure to characterize a reaction intermediate with a clinically useful substrate. Stopped-flow experiments together with rapid freeze−quench EPR and Raman spectroscopies were used to characterize the reaction of Co(II)−BcII with imipenem. These studies show that Co(II)−BcII is able to hydrolyze imipenem in both the mono- and dinuclear forms. In contrast to the situation met for penicillin, the species that accumulates during turnover is an enzyme−intermediate adduct in which the β-lactam bond has already been cleaved. This intermediate is a metal-bound anionic species with a novel resonant structure that is stabilized by the metal ion at the DCH or Zn2 site. This species has been characterized based on its spectroscopic features. This represents a novel, previously unforeseen intermediate that is related to the chemical nature of carbapenems, as confirmed by the finding of a similar intermediate for meropenem. Since carbapenems are the only substrates cleaved by B1, B2, and B3 lactamases, identification of this intermediate could be exploited as a first step toward the design of transition-state-based inhibitors for all three classes of metallo-β-lactamases

    Fission yeast cell morphogenesis: identification of new genes and analysis of their role during the cell cycle.

    Abstract. To identify new genes involved in the control of cell morphogenesis in the fission yeast Schizosaccharomyces pombe we have visually screened for temperature-sensitive mutants that show defects in cell morphology. We have isolated and characterized 64 mutants defining 19 independent genes, 10 of which have not been previously described. One class of mutants, defining 12 orb genes, become round and show a complete loss of cell polarity. A second class of mutants exhibits branched or bent morphologies. These mutants show defects in either selection of the growth site, defining two tea genes, or in the maintenance of growth direction, defining five ban genes.
