    La presente relazione costituisce il compendio di quanto discusso all’interno del seminario organizzato anche con valenza formativa nell’ambito del Master di 2° livello in Ingegneria per la Sicurezza Occupazionale -MISO- e accreditato Educazione Continua in Medicina -ECM-, dal titolo “Valutazione e gestione dei rischi per la salute dei lavoratori nello scavo meccanico di gallerie”. Nei vari interventi, dopo un inquadramento sulle moderne tecniche di scavo meccanico, sono trattati i criteri di approccio per la valutazione e gestione dei principali inquinanti di formazione o generati durante la lavorazione. Sono in particolare approfonditi gli aspetti connessi con la definizione di valori di concentrazione accettabili con uno scenario normativo tuttora in marcata evoluzione. L’ultima parte fornisce indicazioni sulla gestione della sicurezza in coerenza con il moderno approccio in Prevention through Design -PtD- e in qualitĂ , in condizioni normali ed in caso di deviazioni ragionevolmente attendibili. Nel suo insieme quindi, quanto esposto puĂČ costituire riferimento aggiornato sullo stato dell’arte in materia di sicurezza per lo scavo meccanico delle gallerie

    Random planar graphs and the London street network

    In this paper we analyse the street network of London both in its primary and dual representation. To understand its properties, we consider three idealised models based on a grid, a static random planar graph and a growing random planar graph. Comparing the models and the street network, we find that the streets of London form a self-organising system whose growth is characterised by a strict interaction between the metrical and informational space. In particular, a principle of least effort appears to create a balance between the physical and the mental effort required to navigate the city

    Epstein-Barr Virus Independent Dysregulation of UBP43 Expression Alters Interferon-Stimulated Gene Expression in Burkitt Lymphoma

    Epstein-Barr virus (EBV) persists as a life-long latent infection within memory B cells, but how EBV may circumvent the innate immune response within this virus reservoir is unclear. Recent studies suggest that the latency-associated non-coding RNAs of EBV may actually induce type I (antiviral) interferon production, raising the question of how EBV counters the negative consequences this is likely to have on viral persistence. We addressed this by examining the type I interferon response in Burkitt lymphoma (BL) cell lines, the only in vitro model of the restricted program of EBV latency-gene expression in persistently infected B cells in vivo. Importantly, we observed no effect of EBV on interferon alpha-induced signaling or evidence of type I interferon production, suggesting that EBV in this latent state is silent to the cell's innate antiviral surveillance. We did uncover, however, a defect in the negative feedback control of interferon signaling in a subpopulation of BL lines as was revealed by prolonged interferon-stimulated gene transcription consistent with sustained tyrosine phosphorylation on STAT1 and STAT2. This was due to inadequate induction of expression of the ubiquitin-specific protease UBP43, which removes the ubiquitin-like ISG15 polypeptide conjugated to proteins (ISGylation) in response to type I interferons. Results here are consistent with previous findings in genetically engineered Ubp43−/− murine cells that UBP43 down-regulates interferon signaling, independent of its ISG15 isopeptidase activity, by precluding the protein kinase JAK1 from the interferon receptor. This natural deficiency in UBP43 expression may therefore provide a useful model to further probe the biological roles of UBP43 and ISGylation

    Fatty acid composition of Mediterranean buffalo milk fat

    The purpose of this research was to investigate the variation in fatty acid composition of milk fat from four buffalo (Bubalus bubalis) herds under different feeding management and ration composition. Changes in milk fatty acid composition were monitored on a weekly basis. Saturated fatty acids (65.5%) predominated in buffalo milk fat; monounsaturated and polyunsaturated fatty acids were 27.0% and 4.5%, respectively. Of saturated fatty acids, the content of palmitic acid was the highest (30.6%) followed by stearic acid (12.0%) and myristic acid (10.7%). Of the unsaturated fatty acids the content of oleic acid was the highest (26.6%). The average content of conjugated linoleic acid (0.76±0.33) was higher than the maximal values generally reported for dairy cow

    Therapeutic Strategies Targeting Urokinase and Its Receptor in Cancer

    Several studies have ascertained that uPA and uPAR do participate in tumor progression and metastasis and are involved in cell adhesion, migration, invasion and survival, as well as angiogenesis. Increased levels of uPA and uPAR in tumor tissues, stroma and biological fluids correlate with adverse clinic–pathologic features and poor patient outcomes. After binding to uPAR, uPA activates plasminogen to plasmin, a broad-spectrum matrix-and fibrin-degrading enzyme able to facilitate tumor cell invasion and dissemination to distant sites. Moreover, uPAR activated by uPA regulates most cancer cell activities by interacting with a broad range of cell membrane receptors. These findings make uPA and uPAR not only promising diagnostic and prognostic markers but also attractive targets for developing anticancer therapies. In this review, we debate the uPA/uPAR structure–function relationship as well as give an update on the molecules that interfere with or inhibit uPA/uPAR functions. Additionally, the possible clinical development of these compounds is discussed

    Stable or plastic body shape? Emys orbicularis hatchlings-juveniles growth patterns under different ecological conditions

    To test if habitat could shape shell features of freshwater turtles starting from birth, we tested hatchlings of Emys orbicularis, randomly chosen from four clutches of the same locality, under two treatments, stagnant (pond habitat) vs lotic (river habitat) water regimes. Due to mortality at the beginning of the experiment, we finally considered only two clutches. Turtle shells were characterized initially at hatching and then after one year of treatment by means of traditional and geometric morphometrics, and phenotypic trajectories analyses were conducted on both types of data. Carapace growth was isometric both in proportion and shape, while plastron showed an allometric pattern: length increased more than width, while the contour showed an expansion at the rear and a compression of the front. The hydrodynamic conditions during growth did not seem to affect the phenotypic trajectories of the shell proportions or the plastron. On the contrary, a significant effect was found on the phenotypic trajectory of the carapace: the size component (but not the angle one) was positively influenced by the "lentic" treatment, suggesting a different growth rate between the two environmental conditions

    FETR-ALS Study Protocol: A Randomized Clinical Trial of Fecal Microbiota Transplantation in Amyotrophic Lateral Sclerosis

    Background and Rationale: Among the key players in the pathogenesis of Amyotrophic Lateral Sclerosis (ALS), microglia and T regulatory lymphocytes (Treg) are candidate cells for modifying the course of the disease. The gut microbiota (GM) acts by shaping immune tolerance and regulating the Treg number and suppressive function, besides circulating neuropeptides, and other immune cells that play in concert through the gut-brain axis. Previous mouse models have shown an altered enteric flora in early stage ALS, pointing to a possible GM role in ALS pathogenesis. Fecal Microbial Transplantation (FMT) is a well-known therapeutic intervention used to re-establish the proper microenvironment and to modulate enteric and systemic immunity. Methods: We are going to perform a multicenter randomized double-blind clinical trial employing FMT as a therapeutic intervention for ALS patients (NCT0376632). Forty-two ALS patients, at an early stage, will be enrolled with a 2:1 allocation ratio (28 FMT-treated patients vs. 14 controls). Study duration will be 12 months per patient. Three endoscopic procedures for intestinal biopsies in FMT and control groups are predicted at baseline, month 6 and month 12; at baseline and at month 6 fresh feces from healthy donors will be infused at patients in the intervention arm. The primary outcome is a significant change in Treg number between FMT-treated patients and control arm from baseline to month 6. Secondary outcomes include specific biological aims, involving in-depth analysis of immune cells and inflammatory status changes, central and peripheral biomarkers of ALS, besides comprehensive analysis of the gut, saliva and fecal microbiota. Other secondary aims include validated clinical outcomes of ALS (survival, forced vital capacity, and modifications in ALSFRS-R), besides safety and quality of life. Expected Results: We await FMT to increase Treg number and suppressive functionality, switching the immune system surrounding motorneurons to an anti-inflammatory, neuroprotective status. Extensive analysis on immune cell populations, cytokines levels, and microbiota (gut, fecal and saliva) will shed light on early processes possibly leading the degenerative ALS course. Conclusions: This is the first trial with FMT as a potential intervention to modify immunological response to ALS and disease progression at an early stage
