    Analysis of retinoblastoma protein function in the regulation of apoptosis = Analyse der Funktion des Retinoblastoma-Proteins in der Apoptose-Regulation

    Ziel: Im Rahmen dieser Arbeit sollte die Rolle des Retinoblastoma-Proteins (RB) in der negativen Regulation von programmiertem Zelltod untersucht werden. Die Analyse des Einflusses verschiedener aktivitätserhaltender RB-Punktmutanten auf individuelle Apoptose-Signalwege sollte Aufschluss über Mechanismen der Apoptose-Regulation durch RB geben. Thesen: Die negative Regulation von Zellwachstum und Apoptose sind zwei unabhängige Funktionen des Retinoblastoma-Proteins (RB). RB-vermittelter Zellzyklusarrest ist nicht grundsätzlich mit einer erhöhten Resistenz gegenüber Apoptose-aktivierenden Stimuli verbunden RB-vermittelter Zellzyklusarrest kann die Aktivierung von Caspasen nach DNA Schäden hemmen. Die negative Regulation von Apoptose durch RB kann unabhängig von der Regulation von Genaktivität erfolgen. Die Expression von Caspase-resistentem RB (RB-MI) verleiht murinen Fibroblasten-Zellen selektiven Schutz vor TNFR1-aktivierter Apoptose. Diese Resistenz lässt sich auf eine RB-MI-abhängige Unterbrechung des mitochondrialen Apoptose-Signalwegs zurückführen, die durch die Abwesenheit von Caspase-Spaltprodukt des Bid-Proteins gekennzeichnet ist

    Intramyocardial hemorrhage: An enigma for cardiac MRI?

    Cardiovascular magnetic resonance (CMR) is a useful noninvasive technique for determining the presence of microvascular obstruction (MVO) and intramyocardial hemorrhage (IMH), frequently occurring in patients after reperfused myocardial infarction (MI). MVO, or the so-called no-reflow phenomenon, is associated with adverse ventricular remodeling and a poor prognosis during follow-up. Similarly, IMH is considered a severe damage after revascularization by percutaneous primary coronary intervention (PPCI) or fibrinolysis, which represents a worse prognosis. However, the pathophysiology of IMH is not fully understood and imaging modalities might help to better understand that phenomenon. While, during the past decade, several studies examined the distribution patterns of late gadolinium enhancement with different CMR sequences, the standardized CMR protocol for assessment of IMH is not yet well established. The aim of this review is to evaluate the available literature on this issue, with particular regard to CMR sequences. New techniques, such as positron emission tomography/magnetic resonance imaging (PET/MRI), could be useful tools to explore molecular mechanisms of the myocardial infarction healing process

    Breast cancer or metastasis? An unusual case of metastatic malignant pleural mesothelioma to the breast

    Background: Metastases to the breast from extramammary malignancies are very rare, and ruling out the diagnosis of primary breast tumor is important in order to decide on clinical management and predict prognosis. Case presentation: Clinical examination revealed in a 49-year-old hairdresser a 3-cm hard lump adherent to the underlying layers in the right breast. Trucut biopsy was performed. Histology showed a solid proliferation of medium-sized neoplastic polygonal cells. Immunohistochemical analysis showed tumor cells diffusely positive for cytokeratin 8/18 and calretinin and focally positive for cytokeratin 5/6 and Wilms’ tumor 1, e-cadherin, and human bone marrow endothelial-1. Estrogen receptors and progesterone receptors were negative. The final diagnosis was metastatic epithelioid malignant pleural mesothelioma. Conclusions: Immunohistochemistry is an important tool for a conclusive diagnosis of malignant pleural mesothelioma. Owing to the degree of histological and immunohistochemical overlap, a high level of clinical suspicion is essential in order to avoid unnecessary mutilating surgery

    The Application of Barocaloric Solid-State Cooling in the Cold Food Chain for Carbon Footprint Reduction

    In this paper, the application of solid-state cooling based on the barocaloric effect in the cold food supply chain is investigated. Barocaloric solid-state technology is applied to the final links of the cold food supply chain regarding the steps of retail and domestic conservation. In this context, effective barocaloric cooling entails the refrigeration of food at 5 °C (273 K) and as such is a promising cooling technology due to its energy efficiency and environmental friendliness. The categories of food involved in this investigation are meat and fresh food products like soft cheese, yogurt, and milk. The energy performance of the barocaloric system is analyzed and compared with a commercial vapor compression refrigerator of a similar size, both operating using R600a under the same working conditions. Based on the results of this comparison, it is concluded that barocaloric cooling is a favorable technology for application in the final links of the cold food supply chain if the system operates in an ABR cycle at frequencies between 1.25 and 1.50 Hz with a regenerator comprising acetoxy silicone rubber as the solid-state refrigerant and a 50%EG–50% water mixture as the heat transfer fluid flowing at an optimal velocity of 0.15 m s−1. Thus, an appropriate tradeoff between the temperature span, cooling power, and coefficient of performance is guaranteed. Under these conditions, the barocaloric system outperforms the domestic vapor compression cooler operating using R600a
