    Solid-phase extraction followed by dispersive liquid-liquid microextraction for the sensitive determination of ecstasy compounds and amphetamines in biological samples

    A novel approach for the determination of ecstasy and amphetamines (3,4-methylenedioxymethylamphetamine (MDMA, Ecstasy), 3,4-methylenedioxyamphetamine (MDA), 3,4-methylenedioxyethylamphetamine (MDEA) and 3,4-methylenedioxypropylamphetamine (MDPA)) in biological samples is presented. The analytes were extracted from the matrix and transferred to a small volume of a high density, water insoluble solvent using solid-phase extraction (SPE) followed by dispersive liquid-liquid microextraction (DLLME). This combination not only resulted in a high enrichment factor, but also it could be used in complex matrices (biological samples). Some important extraction parameters, such as sample solution flow rate, sample pH, type and volume of extraction and disperser solvents as well as the salt addition, were studied and optimized. Under the optimized conditions, the calibration graphs were linear in the range of 0.5-500 µg L-1 and 1.0-500 µg L-1 with detection limits in the range of 0.1-0.3 µg L-1 and 0.2-0.7 µg L-1 in urine and plasma samples, respectively. The results showed that SPE-DLLME is a suitable method for the determination of ecstasy components and amphetamines in biological and water samples. KEY WORDS: Dispersive liquid-liquid microextraction, Solid-phase extraction, Ecstasy compounds, Amphetamines, Gas chromatography, Biological samples Bull.     Fermi-Bose Mixtures Near Broad Interspecies Feshbach Resonances

    In this Letter we have studied dressed bound states in Fermi-Bose mixtures near broad interspecies resonance, and implications on many-body correlations. We present the evidence for a first order phase transition between a mixture of Fermi gas and condensate, and a fully paired mixture where extended fermionic molecules occupy a single pairing channel instead of forming a molecular Fermi surface. We have further investigated the effect of Fermi surface dynamics, pair fluctuations and discussed the validity of our results.Comment: 5 pages, 4 figure

    In vitro propagation of four Iranian grape varieties: Influence of genotype and pretreatment with arbuscular mycorrhiza

    There is a great demand for table grape saplings, mainly for commercial varieties indicating that micropropagation could be an effective method for their mass propagation. Internal contamination in woody plant species is an important problematic issue and arbuscular mycorrhizal fungi (AMF) have been known as potential plant biological protectors. In the present study, the glasshouse grown mother plants of four grape varieties ('Asgari', 'Khalili', 'Keshmeshi', and 'Shahroudi') were inoculated with AMF as pre-treatment. The fungi strains were Glomus mosseae, G. fasciculatum, G. intraradices and a mixture of all three species. The comparative in vitro performance of these genotypes was evaluated following optimization of in vitro growth conditions for each genotype. Furthermore, the positive effect of AMF inoculation of stock plant on micropropagation process was studied. Changes in biochemical features (total chlorophylls, total phenols and total sugars), growth parameters (root length and total leaf area) and in vitro behavior of AMF pretreated as well as control explants were recorded. The mycorrhizal association with grapevine roots was confirmed following root staining and evaluation of colonization rate. The results revealed a distinct difference and clear genotypic effect on various in vitro parameters of studied grape genotypes. The utilized inocula were found to have the capability of mycorrhizal association with grapevine roots, leading to enhancing phenolics as a defense mechanism, increasing sugars and chlorophyll and finally growth of whole plant corresponding to the grape variety and AMF strain. These results confirmed that health and physiological conditions of the stock plants are important parameters for in vitro grape culture establishment and suggest the integration of mycorrhizal technology with tissue culture to accomplish better results

    Effects of Paper-Mill Sludge as a Mulch versus Topsoil Incorporation on Potassium Uptake and the Grain Yield of Rain-Fed Wheat in a High Specific Surface Loess Soil with Illite Dominance in Clay Fraction

    A field experiment with rain-fed winter wheat investigated the nutritional aspects of paper-mill sludge as a mulch and incorporated into the topsoil. Treatments with chemical fertilizers were also used as controls. Paper-mill sludge used as mulch with high rate (100 MG ha−1) and also the combined N and K mineral fertilizer treatments increased yield when a low potassium otherwise caused potassium deficiency in wheat with high specific surface soil. High soil Ca : K molar ratio by incorporation lowered potassium uptake and yield, with visual symptoms of potassium deficiency. A very high Gapon selectivity coefficient (KG) for K exchange against Ca + Mg (16.58 (L/mole)0.5) produced a nonlinear normalized exchange isotherm in favor of potassium with these soils containing high illite. Ca and K which are released by sludge decomposition are diverged in soil when mobilized by rain infiltration, lowering Ca : K molar ratio. Low soil Ca : K molar ratio may be expected by surface sludge application relative to incorporation, due to greater rain infiltration through upper soil layers and their effluent pore volumes per unit depth. Ca from triple superphosphate by the P, N, and K mineral fertilizers combined also reduced potassium uptake and yield relative to N and K combined

    Anesthesia is considered to be the best way to ensure animal welfare during handling etc. Clove oil and electronarcosis are among the most popular chemical and physical anesthetics used in fisheries and biology;however there is a little information available on the effect of anesthesia using direct electric current in fish. In the present study, the impacts of clove oil (30 mg.l-1) and electro-anesthesia (constant direct current, 0.5 v.cm-1) (DC) were assessed in common carp, Cyprinus carpio during narcotic stage in two experimental sets. Experiment 1 was conducted to quantitatively compare induction of anesthesia and recovery time. The biochemical and hematological effects of anesthetics were analyzed as experiment 2. The results revealed that induction and recovery times in fish anesthetized with electric current were significantly lower than clove oil treatment.Significant differences were observed in cortisol concentrations, red blood cells and mean corpuscular hemoglobin concentration between anesthetic methods. Our results suggest that constant direct current electronarcosis was more quickly than clove oil and associated with lower side effects in common carp

    Effects of different packaging methods on microbial, [chemical] and sensory properties of Nile tilapia (Oreochromis niloticus Linnaeus, 1758) fillets during refrigerator storage

    The effect of three different packaging methods including Modified Atmosphere Packaging (MAP), Vacuum Packaging and normal Packaging was investigated on the quality of Nile tilapia fresh fillets stored in the refrigerator's temperature. The packaged samples were examined for 10 days with regard to the changes in chemical (TVN, PV, pH), microbial (total viable count) and sensory evaluations. The results indicated that the samples packed in MAP condition had higher quality than that of other methods at the end of the storage period. In addition, the slower destructive impacts and microbial growth was observed in MAP. The results of present study suggest that packaging tilapia under MAP conditions results in the increase in the durability, storing, and distribution period for fillets

    Goldilocks mixing in oceanic shear-induced turbulent overturns

    We present a new, simple and physically motivated parameterization, based on the ratio of Thorpe and Ozmidov scales, for the irreversible turbulent flux coefficient ΓM=M/ϵ , i.e. the ratio of the irreversible rate M at which the background potential energy increases in a stratified flow due to macroscopic motions to the dissipation rate of turbulent kinetic energy ϵ . Our parameterization covers all three key phases (crucially, in time) of a shear-induced stratified turbulence life cycle: the initial, ‘hot’ growing phase, the intermediate energetically forced phase and the final ‘cold’ fossilization decaying phase. Covering all three phases allows us to highlight the importance of the intermediate one, to which we refer as the ‘Goldilocks’ phase due to its apparently optimal (and so neither too hot nor too cold, but just right) balance, in which energy transfer from background shear to the turbulent mixing is most efficient. The value of ΓM is close to 1/3 during this phase, which we demonstrate appears to be related to an adjustment towards a critical or marginal Richardson number for sustained turbulence ∼0.2--0.25 . Importantly, although buoyancy effects are still significant at leading order for the turbulent dynamics during this intermediate phase, the marginal balance in the flow ensures that the turbulent mixing of the (density) scalar is nevertheless effectively ‘locked’ to the turbulent mixing of momentum. We present supporting evidence for our parameterization through comparison with six oceanographic datasets that span various turbulence generation regimes and a wide range of geographical location and depth. Using these observations, we highlight the significance of parameterizing an inherently variable flux coefficient for capturing the turbulent flux associated with rare energetic, yet fundamentally shear-driven (and so not strongly stratified) overturns that make a disproportionate contribution to the total mixing. We also highlight the importance of representation of young turbulent patches in the parameterization for connecting the small scale physics to larger scale applications of mixing such as ocean circulation and tracer budgets. Shear-induced turbulence is therefore central to irreversible mixing in the world's oceans, apparently even close to the seafloor, and it is critically important to appreciate the inherent time dependence and evolution of mixing events: history matters to mixing
