    Gas Sensing Properties of Rutile-tio2 (100) Films Prepared by Pulsed Laser Deposition

    Gas sensing property of TiO2 thin films have been demonstrated in rutile-TiO2 (100) films grown on the a-Al2O3 (0001)substrates by Pulsed Laser Deposition (PLD). High quality rutile-TiO2 (100) films were successfully grown on a-Al2O3(0001) with the substrate temperature at 500oC under 15 mTorr of O2 gas pressure. The thickness and crystallinity ofTiO2 films were evaluated by Rutherford backscattering spectrometry combined with channeling (RBS/C) and X-raydiffraction using q-2q scans. To evaluate CO2 gas sensing property of TiO2 films, the dependence of the changing ofelectrical resistivity on the temperature was measured. It's found that high crystallinity rutile-TiO2 (100) films on the a-Al2O3 (0001) substrate kept at 100oC exhibits good gas sensing property for CO2 gas

    Self‐Emergent Protocells Generated in an Aqueous Solution with Binary Macromolecules through Liquid‐Liquid Phase Separation

    生体内の高分子混雑に着目した新規の細胞モデルの創成に成功. 京都大学プレスリリース. 2020-12-16.Recently, liquid–liquid phase separation (LLPS) has attracted considerable attention among researchers in the life sciences as a plausible mechanism for the generation of microstructures inside cells. LLPS occurs through multiple nonspecific interactions and does not always require a lock‐and‐key interaction with a binary macromolecular solution. The remarkable features of LLPS include the non‐uniform localization and concentration of solutes, resulting in the ability to isolate certain chemical systems and thereby parallelize multiple chemical reactions within the limited space of a living cell. We report that, by using the macromolecules, poly(ethylene glycol) (PEG) and dextran, that exhibit LLPS in an aqueous solution, cell‐sized liposomes are spontaneously formed therein in the presence of phospholipids. In this system, LLPS is generated through the depletion effect of macromolecules. The results showed that cell‐like microdroplets entrapping DNA wrapped by a phospholipid layer emerge in a self‐organized manner

    Clinical efficacy and cytokine network-modulating effects of tacrolimus in myasthenia gravis

    金沢大学保健管理センター115To clarify the long-term efficacy, safety and the cytokine network-modulating effects of tacrolimus in myasthenia gravis, medical records of 86 newly diagnosed consecutive patients and nine steroid-dependent patients were retrospectively reviewed, and peripheral blood mononuclear cells (PBMC) were cultured for the cytokine profile. Steroid reduction effects were observed by using tacrolimus, and no serious adverse effects were observed. The culture study showed reduced IL-12, IL-17, IFN-γ, GM-CSF, TNF-α and MIP-1β, and elevated IL-10 in the PBMC from patients who received tacrolimus, which suggests inhibition of T cells and macrophages, and enhancement of type 1 regulatory T cells. © 2008 Elsevier B.V.     A nitrogen isoscape of phytoplankton in the western North Pacific created with a marine nitrogen isotope model

    The nitrogen isotopic composition (δ15N) of phytoplankton varies substantially in the ocean reflecting biogeochemical processes such as N2 fixation, denitrification, and nitrate assimilation by phytoplankton. The δ15N values of zooplankton or fish inherit the values of the phytoplankton on which they feed. Combining δ15N values of marine organisms with a map of δ15N values (i.e., a nitrogen isoscape) of phytoplankton can reveal the habitat of marine organisms. Remarkable progress has been made in reconstructing time-series of δ15N values of migratory fish from various tissues, such as otoliths, fish scales, vertebrae, and eye lenses. However, there are no accurate nitrogen isoscapes of phytoplankton due to observational heterogeneity, preventing improvement in the accuracy of estimating migratory routes using the fish δ15N values. Here we present a nitrogen isoscape of phytoplankton in the western North Pacific created with a nitrogen isotope model. The simulated phytoplankton is relatively depleted in 15N at the subtropical site (annual average δ15N value of phytoplankton of 0.6‰), where N2 fixation occurs, and at the subarctic site (2.1‰), where nitrate assimilation by phytoplankton is low due to iron limitation. The simulated phytoplankton is enriched in 15N at the Kuroshio–Oyashio transition site (3.9‰), where nitrate utilization is high, and in the region around the Bering Strait site (6.7‰), where partial nitrification and benthic denitrification occur. The simulated δ15N distributions of nitrate, phytoplankton, and particulate organic nitrogen are consistent with δ15N observations in the western North Pacific. The seamless nitrogen isoscapes created in this study can be used to improve our understanding of the habitat of marine organisms or fish migration in the western North Pacific

    EFFICACY OF INTERFERON THERAPY FOR CHRONIC HEPATITIS C : A COOPERATIVE STUDY IN ELEVEN HOSPITALS

    We investigated the influences of liver histology,serum levels of hepatitis C virus (HCV) and HCV genotypes on responsiveness to interferon (IFN) therapy in 342 patients with chronic hepatitis C. Either 9 million units (MU) of lymphoblastoid alpha IFN or 3 MU of recombinant IFN-alpha was administered daily for 2 weeks and then three times a week for 22 weeks. IFN responses were divided into three groups on the basis of the results of polymerase chain reaction (PCR) assay detecting HCV-RNA in serum. Complete response (CR) was defined as sustained elimination of HCV for at least 6 months after treatment,partial response (PR) as HCV elimination for a limited period,non-response (NR) as continuously positive for HCV-RNA in serum. Quantitation of pre-treament HCV-RNA amount in serum was determined by competitive PCR assay in 47 patients. HCV genotyping was performed in 114 patients by PCR with genotype-specific primers. CR was obtained in 97 patients (28.4%),PR in 104 (30.4%) and NR in 141 (41.2%). IFN responses,represented by CR/PR/NR,were 15/18/11 in 44 patients with chronic persistent hepatitis (CPH),72/65/73 in 210 patients with chronic aggressive hepatitis (CAH) 2a,and 10/21/57 in 88 patients with CAH2b. CR rate was lower in patients with CAH2b (11.4%) compared to those with CPH (34.1%) or CAH2a (34.3%). Averages of pre-treatment serum HCV-RNA amount (copies/50μl) were 10³·⁵⁵ in 13 CRs,10⁴·⁵⁶ in 17 PRs,and 10⁵·⁹⁵ in 17 NRs. There was a positive correlation between pre-treatment HCV-RNA levels and IFN unresponsiveness. HCV genotyping in 114 patients revealed that HCV type Ⅰ infection was observed in one,type Ⅱ in 94,type Ⅲ in 11,type Ⅳ in 6 and mixed (types Ⅱ and Ⅳ) in 2 patients,and their IFN responses (CR/PR/NR) were 0/0/1,28/26/40,3/5/3,1/3/2 and 0/1/1,respectively

    Successful Endoscopic Injection Sclerotherapy of High-Risk Gastroesophageal Varices in a Cirrhotic Patient with Hemophilia A

    A 68-year-old man with hemophilia A and liver cirrhosis caused by hepatitis C virus was referred to our hospital to receive prophylactic endoscopic treatment for gastroesophageal varices (GOV). He had large, tense, and winding esophageal varices (EV) with cherry red spots extending down to lesser curve, predicting the likelihood of bleeding. Esophageal endoscopic injection sclerotherapy (EIS) was performed with a total 15 mL of 5% ethanolamine oleate with iopamidol (EOI). Radiographic imaging during EIS demonstrated that 5% EOI reached the afferent vein of the varices. He was administered sufficient factor VIII concentrate before and after EIS to prevent massive bleeding from the varices. Seven days after EIS, upper gastrointestinal endoscopy (UGIE) showed that the varices were eradicated almost completely. Eighteen months after EIS, the varices continued to diminish. We report a successful case of safe and effective EIS for GOV in a high-risk cirrhotic patient with hemophilia A

    Autoantibody-induced internalization of nicotinic acetylcholine receptor α3 subunit exogenously expressed in human embryonic kidney cells

    Autoantibody against nicotinic acetylcholine receptor (nAChR) α3 subunit has been implicated in the pathogenesis of paraneoplastic neurological syndrome. To examine the effect of anti-α3 subunit autoantibody on cell-surface nAChRs, we established human embryonic kidney 293 cells stably co-expressing α3 and β4 subunits. Upon incubation with seropositive patient\u27s serum, this cell line showed co-accumulation of patient\u27s IgG and α3 subunits in the cytoplasm. These data support the hypothesis that anti-α3 subunit autoantibody induces internalization of cell-surface nAChRs and thereby impairs synaptic transmission. © 2012 Elsevier B.V. All rights reserved
