    The Metabolic Consequences of Hepatic AMP-Kinase Phosphorylation in Rainbow Trout

    AMP-activated protein kinase (AMPK), a phylogenetically conserved serine/threonine protein kinase, is proposed to function as a “fuel gauge” to monitor cellular energy status in response to nutritional environmental variations. However, in fish, few studies have addressed the metabolic consequences related to the activation of this kinase. This study demonstrates that the rainbow trout (Oncorhynchus mykiss) possesses paralogs of the three known AMPK subunits that co-diversified, that the AMPK protein is present in the liver and in isolated hepatocytes, and it does change in response to physiological (fasting-re-feeding cycle) and pharmacological (AICAR and metformin administration and incubations) manipulations. Moreover, the phosphorylation of AMPK results in the phosphorylation of acetyl-CoA carboxylase, a main downstream target of AMPK in mammals. Other findings include changes in hepatic glycogen levels and several molecular actors involved in hepatic glucose and lipid metabolism, including mRNA transcript levels for glucokinase, glucose-6-phosphatase and fatty acid synthase both in vivo and in vitro. The fact that most results presented in this study are consistent with the recognized role of AMPK as a master regulator of energy homeostasis in living organisms supports the idea that these functions are conserved in this piscine model

    As vesículas extracelulares urinárias (uEV) são nanomoléculas de origem celular variada, as quais são secretadas para o espaço extracelular. De fato, vesículas extracelulares urinárias vem sendo intensamente estudadas como potenciais biomarcadores em estudo sobre hipertensão, nefropatias, glomerulonefrites, câncer e doenças urogenitais. Apesar dos intensos estudos, ainda há falta de padronização nos relatos científicos sobre as condições de armazenamento e a influência do tempo de armazenamento das amostras em -80oC. Assim, as condições de armazenamento da urina podem influenciar a identificação e quantificação da uEV. Portanto, o objetivo deste trabalho é analisar a diferença na viabilidade de uEV frescas e congeladas a partir de amostras de urina de indivíduos saudáveis. Avaliamos as uEVs totais e derivadas de podócitos por citometria de fluxo nas urinas frescas e após um mês congeladas à -80oC. As amostras foram adquiridas e quantificadas no citômetro de fluxo Cytoflex (BC, USA) e a análise de dados foi feita no software FlowJo Versão 9.0. Como resultado, vimos que houve uma diminuição significativa da quantidade de vesículas extracelulares urinárias encontradas nas amostras que foram congeladas por um mês se comparadas às amostras frescas, indicando perda de viabilidade dessas urinas pelo tempo de congelamento, o que indica prejuízo em sua utilização como forma de controle de pacientes saudáveis em outros estudos que possam vir a ser realizados com amostras de urinas congeladasUrinary extracellular vesicles (uEV) are nanomolecules of varied cellular origin, which are secreted into the extracellular space. In fact, extracellular vesicles have been intensely studied as potential biomarkers in studies on hypertension, nephropathy, glomerulonephritis, cancer and urogenital diseases. Despite intense studies, there is still a lack of standardization in scientific reports on the storage conditions and the influence of sample time storage at -80oC. Thus, urine storage conditions may influence the identification and quantification of uEVs. Therefore, the objective of this work is to analyze the difference in the viability of fresh and frozen uEVs from urine samples of healthy individuals. Samples were acquired and quantified using a Cytoflex flow cytometer (BC, USA) and data analysis was performed using FlowJo Version 9.0 software. As a result, we saw that there was a significant decrease in the amount of urinary extracellular vesicles found in the urines that were frozen for one month if compared to the fresh ones, indicating loss of viability of these urines due to storage time, which indicates a disadvantage in their use in other studies that may be performed with frozen urine samples28 f
