    Fluid dynamics of aortic root dilation in Marfan syndrome

    Aortic root dilation and propensity to dissection are typical manifestations of the Marfan Syndrome (MS), a genetic defect leading to the degeneration of the elastic fibres. Dilation affects the structure of the flow and, in turn, altered flow may play a role in vessel dilation, generation of aneurysms, and dissection. The aim of the present work is the investigation in-vitro of the fluid dynamic modifications occurring as a consequence of the morphological changes typically induced in the aortic root by MS. A mock-loop reproducing the left ventricle outflow tract and the aortic root was used to measure time resolved velocity maps on a longitudinal symmetry plane of the aortic root. Two dilated model aortas, designed to resemble morphological characteristics typically observed in MS patients, have been compared to a reference, healthy geometry. The aortic model was designed to quantitatively reproduce the change of aortic distensibility caused by MS. Results demonstrate that vorticity released from the valve leaflets, and possibly accumulating in the root, plays a fundamental role in redirecting the systolic jet issued from the aortic valve.     On the improved adhesion of NiTi wires embedded in polyester and vinylester resins

    This paper discusses the effect of different surface treatments on shape memory alloy wires embedded in PolyEster (PE) and VinylEster (VE) polymeric matrices. In particular, two types of chemical etching and a chemical bonding with a silane coupling agent have been performed on the surfaces of the wires. Pull-out tests have been carried out on samples made from a specifically designed Teflon mould. Considering the best results of the pull-out tests obtained with PE resin, the debonding induced by strain recovery of 4%, 5% and 6% pre-strained NiTi wires has been evaluated with the wires being subjected to different surface treatment conditions and then being embedded in the PE matrix. The results prove that the wires functionalised and embedded in the PE resin show the maximum pull-out forces and the highest interfacial adhesion. Finally, it has been found that debonding induced by strain recovery is strongly related to the propagation towards the radial direction of sharp cracks at the debonding region

    Honey bee brood ring-test: method for testing pesticide toxicity on honeybee brood in laboratory conditions

    contribution to session IV Test methodology The Experimental unit of entomology (INRA, France) developed a new in vitro method to assess effects of pesticides on honey bee larvae. The method consists in rearing bee larvae in plastic cells. The larvae are fed with diet containing 50% of fresh royal jelly and 50% of an aqueous sugar and yeast extract solution, and reared in an incubator at 35 °C and 96% relative humidity. According to that method, 9 tests (7 in 2008 and 2 in 2005) were carried out in 7 laboratories and different countries. The objective of these trials was to assess the LD50 for dimethoate 48 hours after an acute exposure. The LD50 values ranged from 1.5 μg a.i./larva to 8.8 μg a.i./larva, with 2 tests with particularly high values (5.0 and 8.8 μg a.i./larva). In 7 tests, these values ranged from 1.5 μg a.i./larva to 3.1 μg a.i./larva. Such variability may be due to the colony origin, the season and larva heterogeneity at grafting.     Guidelines for the use and interpretation of assays for monitoring autophagy (4th edition)1.

    In 2008, we published the first set of guidelines for standardizing research in autophagy. Since then, this topic has received increasing attention, and many scientists have entered the field. Our knowledge base and relevant new technologies have also been expanding. Thus, it is important to formulate on a regular basis updated guidelines for monitoring autophagy in different organisms. Despite numerous reviews, there continues to be confusion regarding acceptable methods to evaluate autophagy, especially in multicellular eukaryotes. Here, we present a set of guidelines for investigators to select and interpret methods to examine autophagy and related processes, and for reviewers to provide realistic and reasonable critiques of reports that are focused on these processes. These guidelines are not meant to be a dogmatic set of rules, because the appropriateness of any assay largely depends on the question being asked and the system being used. Moreover, no individual assay is perfect for every situation, calling for the use of multiple techniques to properly monitor autophagy in each experimental setting. Finally, several core components of the autophagy machinery have been implicated in distinct autophagic processes (canonical and noncanonical autophagy), implying that genetic approaches to block autophagy should rely on targeting two or more autophagy-related genes that ideally participate in distinct steps of the pathway. Along similar lines, because multiple proteins involved in autophagy also regulate other cellular pathways including apoptosis, not all of them can be used as a specific marker for bona fide autophagic responses. Here, we critically discuss current methods of assessing autophagy and the information they can, or cannot, provide. Our ultimate goal is to encourage intellectual and technical innovation in the field
