    Vision Stations: Addressing Corrective Vision Needs With Low-cost Technologies

    Eyeglasses, required for functional vision by nearly half the world\u27s population, are still needed by more than a billion people. There are a number of constraints on the provision of eyeglasses: product cost, durability, and appearance; traditional approaches to evaluating refraction; and sustainably scaling potential distribution methods. We offer our experience with an immigrant population in a US urban setting using a Vision Station. The station allowed for immediate provision of adjustable glasses using self-refraction, ordering of custom lenses from a low-cost website, and referral to primary and eye care physicians for those with medical eye concerns. As with models in development by other groups, Vision Stations connect people with the life-changing provision of functional vision

Although addressing this issue is challenging, it can be seen as encouraging that many householders with below-Standard incomes are already part of the workforce. It is possible that many householders have adequate levels of education and experience but face other barriers that keep their wages low or raise their expenses. Identifying and addressing such barriers is the next step in bringing household incomes and costs into balance

    LHC Limits on the Top-Higgs in Models with Strong Top-Quark Dynamics

    LHC searches for the standard model Higgs Boson in WW or ZZ decay modes place strong constraints on the top-Higgs state predicted in many models with new dynamics preferentially affecting top quarks. Such a state couples strongly to top-quarks, and is therefore produced through gluon fusion at a rate enhanced relative to the rate for the standard model Higgs boson. A top-Higgs state with mass less than 300 GeV is excluded at 95% CL if the associated top-pion has a mass of 150 GeV, and the constraint is even stronger if the mass of the top-pion state exceeds the top-quark mass or if the top-pion decay constant is a substantial fraction of the weak scale. These results have significant implications for theories with strong top dynamics, such as topcolor-assisted technicolor, top-seesaw models, and certain Higgsless models.Comment: 9 pages, 6 pdf figures included.     Enhanced conformational space sampling improves the prediction of chemical shifts in proteins.

    A biased-potential molecular dynamics simulation method, accelerated molecular dynamics (AMD), was combined with the chemical shift prediction algorithm SHIFTX to calculate (1)H(N), (15)N, (13)Calpha, (13)Cbeta, and (13)C' chemical shifts of the ankyrin repeat protein IkappaBalpha (residues 67-206), the primary inhibitor of nuclear factor kappa-B (NF-kappaB). Free-energy-weighted molecular ensembles were generated over a range of acceleration levels, affording systematic enhancement of the conformational space sampling of the protein. We have found that the predicted chemical shifts, particularly for the (15)N, (13)Calpha, and (13)Cbeta nuclei, improve substantially with enhanced conformational space sampling up to an optimal acceleration level. Significant improvement in the predicted chemical shift data coincides with those regions of the protein that exhibit backbone dynamics on longer time scales. Interestingly, the optimal acceleration level for reproduction of the chemical shift data has previously been shown to best reproduce the experimental residual dipolar coupling (RDC) data for this system, as both chemical shift data and RDCs report on an ensemble and time average in the millisecond range

    Trypanosoma cruzi glutathione-dependent peroxidase I (TcGPXI) can reduce fatty acid, phospholipid, and short chain organic hydroperoxides utilizing a novel redox cycle in which enzyme activity is linked to the reduction of trypanothione, a parasite-specific thiol, by glutathione. Here we show that TcGPXI activity can also be linked to trypanothione reduction by an alternative pathway involving the thioredoxin-like protein tryparedoxin. The presence of this new pathway was first detected using dialyzed soluble fractions of parasite extract. Tryparedoxin was identified as the intermediate molecule following purification, sequence analysis, antibody studies, and reconstitution of the redox cycle in vitro. The system can be readily saturated by trypanothione, the rate-limiting step being the interaction of trypanothione with the tryparedoxin. Both tryparedoxin and TcGPXI operate by a ping-pong mechanism. Overexpression of TcGPXI in transfected parasites confers increased resistance to exogenous hydroperoxides. TcGPXI contains a carboxyl-terminal tripeptide (ARI) that could act as a targeting signal for the glycosome, a kinetoplastid-specific organelle. Using immunofluorescence, tagged fluorescent proteins, and biochemical fractionation, we have demonstrated that TcGPXI is localized to both the glycosome and the cytosol. The ability of TcGPXI to use alternative electron donors may reflect their availability at the corresponding subcellular sites

    Using electronic health records to support clinical trials: a report on stakeholder engagement for EHR4CR

    Background. The conduct of clinical trials is increasingly challenging due to greater complexity and governance requirements as well as difficulties with recruitment and retention. Electronic Health Records for Clinical Research (EHR4CR) aims at improving the conduct of trials by using existing routinely collected data, but little is known about stakeholder views on data availability, information governance, and acceptable working practices. Methods. Senior figures in healthcare organisations across Europe were provided with a description of the project and structured interviews were subsequently conducted to elicit their views. Results. 37 structured interviewees in Germany, UK, Switzerland, and France indicated strong support for the proposed EHR4CR platform. All interviewees reported that using the platform for assessing feasibility would enhance the conduct of clinical trials and the majority also felt it would reduce workloads. Interviewees felt the platform could enhance trial recruitment and adverse event reporting but also felt it could raise either ethical or information governance concerns in their country. Conclusions. There was clear support for EHR4CR and a belief that it could reduce workloads and improve the conduct and quality of trials. However data security, privacy, and information governance issues would need to be carefully managed in the development of the platform
