    Common promoter variant in cyclooxygenase-2 represses gene expression: evidence of role in acute-phase inflammatory response

    Objective: Cyclooxygenase (COX)-2 is a key regulatory enzyme in the synthesis of prostanoids associated with trauma and inflammation. We investigated the COX-2 gene for functional variants that may influence susceptibility to disease. Methods and results: The promoter of COX-2 was screened for variants in healthy subjects by use of polymerase chain reaction-based methods. Promoter activity was investigated by using reporter expression experiments in human lung fibroblasts. Patients undergoing coronary artery bypass graft surgery, with measurements of plasma markers linked to COX-2 activity, were genotyped for association studies. A common COX-2 promoter variant, -765G>C, was found and shown to be carried by >25% of a group of healthy UK subjects. The -765C allele had significantly lower promoter activity compared with -765G, basally (28±3% lower, P<0.005) and in serum-stimulated cells (31±2% lower, P<0.005). In patients subjected to coronary artery bypass graft surgery, the magnitude of rise in levels of C-reactive protein (CRP) was strongly genotype dependent. Compared with -765G homozygotes, patients carrying the -765C allele had significantly lower plasma CRP levels at 1 to 4 days after surgery (14% lower at the peak of CRP levels on day 3, P<0.05 for all time points).     Since multivariate lifetime data frequently occur in applications, various properties of multivariate distributions have been previously considered to model and describe the main concepts of aging commonly considered in the univariate setting. The generalization of univariate aging notions to the multivariate case involves, among other factors, appropriate definitions of multivariate quantiles and related notions, which are able to correctly describe the intrinsic characteristics of the concepts of aging that should be generalized, and which provide useful tools in the applications. A new multivariate version of the well-known New Better than Used in Expectation univariate aging notion is provided, by means of the concepts of the upper corrected orthant and multivariate excess-wealth function. Some of its properties are described, with particular attention paid to those that can be useful in the analysis of real data sets. Finally, through an example it is illustrated how the new multivariate aging notion influences the final results in the analysis of data on tumor growth from the Comprehensive Cohort Study performed by the German Breast Cancer Study Grou

    Activation of CD22; a Potential Novel Marker for Ovarian Cancer

    CD22 in the human genome codes for a protein of the same name, which is normally expressed only on the surface of B-lymphocytes. The primary hypothesis is that inappropriate activation of CD22 enables cell metastasis to lymph nodes and bone. The aim of this research is to determine whether CD22 is activated in cancerous ovarian cell lines, whether CD22 is expressed on the surface of these cells, and to compare the expression of CD22 expression on B-cell lines with cancerous ovarian cells to determine expression patterns and characterize normal and aberrant CD22 receptor substrate interactions on bone marrow stromal cells. This research will improve understanding of aberrant expression of CD22 on tumors, which may potentially be used for antibody-directed therapy for ovarian cancer

    Microplastics on the menu: Plastics pollute Indonesian Manta Ray and Whale Shark feeding grounds

    The implications of plastic pollution, including microplastics, on marine ecosystems and species are increasingly seen as an environmental disaster. Yet few reports focus on filter-feeding megafauna in regions heavily impacted by plastic pollution, such as Indonesia in the Coral Triangle, a global marine biodiversity hotspot. Here, we evaluate plastic abundance and characterize debris from feeding grounds for manta rays Mobula alfredi and whale sharks Rhincodon typus in three coastal locations in Indonesia: Nusa Penida Marine Protected Area, Komodo National Park, and Pantai Bentar, East Java. A 200 μm plankton net was used to sample the top 0.5 m of the water column (‘trawl survey’) and floating plastics were assessed along ∼440 m long transects (‘visual survey’) during the Indonesian north-west (wet) and south-east (dry) monsoon seasons during 2016–2018. Microplastics were identified visually, measured and categorized from trawl samples, and larger floating plastics were counted and categorized visually from boats. Plastic abundance ranged widely from 0.04 to 0.90 pieces m–3 (trawl survey) and 210 to 40,844 pieces km–2 (visual survey). Results from linear models showed significant seasonal and location differences in estimated plastic abundance for trawl and visual surveys in Nusa Penida and Komodo. Plastic abundance was up to ∼ 44 times higher in the wet than the dry season, with the largest seasonal effect observed in Nusa Penida. Overall, small pieces  50% combined) were the most prevalent plastics. Theoretical plastic ingestion rates were calculated using estimated filtration volumes of manta rays and whale sharks and the mean plastic abundance in their feeding grounds. Upper plastic ingestion estimates for manta rays were ∼63 and 25 pieces h–1 for Nusa Penida and Komodo locations, respectively, and ∼137 pieces h–1 for whale sharks in Java. Analysis of manta ray egested material confirmed plastic ingestion, the consequences of which might include exposure to toxic plastic additives and adhered persistent organic pollutants. Communicating this information to communities who stand to benefit from healthy megafauna populations might help local governments as they work toward reducing plastics in the marine environment

    Measuring the gap in ARPES experiments

    Angle-resolved photoemission spectroscopy (ARPES) is considered as the only experimental tool from which the momentum distribution of both the superconducting and pseudo-gap can be quantitatively derived. The binding energy of the leading edge of the photoemission spectrum, usually called the leading edge gap (LEG), is the model-independent quantity which can be measured in the modern ARPES experiments with the very high accuracy--better than 1 meV. This, however, may be useless as long as the relation between the LEG and the real gap is unknown. We present a systematic study of the LEG as a function of a number of physical and experimental parameters. The absolute gap values which have been derived from the numerical simulation prove, for example that the nodal direction in the underdoped Bi-2212 in superconducting state is really the node--the gap is zero.     Determining patterns in the composition of dissolved organic matter in fresh waters according to land use and management

    In fresh waters, the origins of dissolved organic matter (DOM) have been found to exert a fundamental control on its reactivity, and ultimately, its ecosystem functional role. A detailed understanding of landscape scale factors that control the export of DOM to aquatic ecosystems is, therefore, pivotal if the effects of DOM flux to fresh waters are to be fully understood. In this study we present data from a national sampling campaign across the United Kingdom in which we explore the variability in DOM composition in three broad landscape types defined by similar precipitation, geology, land use and management, hydrology, and nutrient enrichment status. We characterised samples from fifty-one sites, grouping them into one of three major underlying classifications: circumneutral streams underlain by clay and mudstone (referred to as ‘clay’), alkaline streams underlain by Cretaceous Chalk or by Carboniferous or Jurassic Limestone (‘limestone’), and acidic streams in peatland catchments underlain by a range of low permeability lithologies (‘peat’). DOM composition was assessed through organic matter stoichiometry (organic carbon: organic nitrogen; organic carbon: organic phosphorus; C/N(P)DOM) and metrics derived from ultra-violet (UV)/visible spectroscopic analysis of DOM such as specific UV absorption (a254 nm; SUVA254). We found similar SUVA254, C/NDOM and DOM/a254 relationships within classifications, demonstrating that despite a large degree of heterogeneity within environments, catchments with shared environmental character and anthropogenic disturbance export DOM with a similar composition and character. Improving our understanding of DOM characterisation is important to help predict shifts in stream ecosystem function, and ecological responses to enrichment or mitigation efforts and how these may result in species composition shifts and biodiversity loss in freshwater ecosystems

    Evolution of the pairing pseudogap in the spectral function with interplane anisotropy

    We study the pairing pseudogap in the spectral function as a function of interplane coupling. The analytical expressions for the self-energy in the critical regime are obtained for any degree of anisotropy. The frequency dependence of the self-energy is found to be qualitatively different in two and three dimensions, and the crossover from two to three dimensional behavior is discussed. In particular, by considering the anisotropy of the Fermi velocity and gap along the Fermi surface, we can qualitatively explain recent photoemission experiments on high temperature superconductors concerning the temperature dependent Fermi arcs seen in the pseudogap phase.Comment: 20 pages, revtex, 5 encapsulated postscript figures include

    Q fever prevention and vaccination: Australian livestock farmers' knowledge and attitudes to inform a One Health approach

    Background: Livestock farmers are at risk of Q fever, a zoonotic disease transmitted to humans from animals such as cattle, sheep and goats. Australia bears substantial Q fever burden, particularly among farmers. A One Health approach engages cross-sectoral collaboration among animal, human and environmental health and is the preferred framework for Q fever prevention. Methods: Cattle, sheep and goat farmers were invited to participate in an online survey in 2019 to gauge perceptions about Q fever and its prevention. Participants were recruited via membership newsletters and social media. Descriptive analyses and logistic regressions were performed. Results: A total of 351 farmers completed the survey. Most respondents (80%) had been farming for ≥20 years, with sheep and beef cattle their primary stock. 71% reported knowledge of Q fever, and 85% identified transmission through contaminated dust inhalation was highly likely. The majority of respondents (97%) were aware of Q fever vaccine, and 95% agreed it was effective in preventing disease, yet 42% remained unvaccinated. Reported barriers to vaccination included poor access to a trained doctor and time and cost related to vaccination. Most farmers (≥91%) believed that subsidized vaccination and improved awareness would promote higher uptake. Conclusion: While Q fever knowledge among respondents was good, their practices related to airborne transmission prevention were poor. Livestock farmers would benefit from adherence to dust and aerosol transmission prevention practices. One Health partnership between government and industry is needed to promote Q fever awareness and address low vaccination rates among livestock farmers by funding vaccination programs.Md Rezanur Rahaman, Helen Marshall, Adriana Milazzo, Deane Crabb, Peng B
