All-weather ice information system for Alaskan arctic coastal shipping
A near real-time ice information system designed to aid arctic coast shipping along the Alaskan North Slope is described. The system utilizes a X-band Side Looking Airborne Radar (SLAR) mounted aboard a U.S. Coast Guard HC-130B aircraft. Radar mapping procedures showing the type, areal distribution and concentration of ice cover were developed. In order to guide vessel operational movements, near real-time SLAR image data were transmitted directly from the SLAR aircraft to Barrow, Alaska and the U.S. Coast Guard icebreaker Glacier. In addition, SLAR image data were transmitted in real time to Cleveland, Ohio via the NOAA-GOES Satellite. Radar images developed in Cleveland were subsequently facsimile transmitted to the U.S. Navy's Fleet Weather Facility in Suitland, Maryland for use in ice forecasting and also as a demonstration back to Barrow via the Communications Technology Satellite
Development of nociceptive synaptic inputs to the neonatal rat dorsal horn: glutamate release by capsaicin and menthol
To study the postnatal development of nociceptive synaptic inputs in the superficial dorsal horn of the neonatal rat spinal cord, we examined the effect of capsaicin and menthol on glutamatergic mEPSCs in postnatal day (P) 0-1, P5-6 and P9-11 slices of spinal cord. Capsaicin (100 nM to 2 muM) increased the mEPSC frequency in a concentration-dependent manner at all ages tested, with a significant enhancement of the effect between P5 and P10. This effect was sensitive to vanilloid receptor (VR) antagonists. The elevation in mEPSC frequency occurred at concentrations of capsaicin (100 nM) that did not alter the distribution of mEPSC amplitudes and was abolished by a dorsal rhizotomy, demonstrating that capsaicin acts via presynaptic VR1 receptors localized on primary afferents. Menthol significantly increased the mEPSC frequency with a similar developmental pattern to capsaicin without consistently affecting mEPSC amplitude. The increase in mEPSC frequency following capsaicin did not depend on transmembrane calcium influx since it persisted in zero [Ca2+](o). The facilitation of spontaneous glutamate release by capsaicin was sufficient to evoke action potentials in neonatal dorsal horn neurons but was accompanied by a block of EPSCs evoked by electrical stimulation of the dorsal root. These results indicate that VR1-expressing nociceptive primary afferents; form functional synaptic connections in the superficial dorsal horn from birth and that activation of the VR1 receptor increases spontaneous glutamate release via an undetermined mechanism. In addition, the data suggest that immature primary afferents express functional menthol receptors that are capable of modulating transmitter release. These results have important functional implications for infant pain processing
Coyotes Go âBridge and Tunnelâ: A Narrow Opportunity to Study the Socio-ecological Impacts of Coyote Range Expansion on Long Island, NY Pre- and Post-Arrival
Currently, Long Island, NY is without a breeding population of northeastern coyote (Canis latras var.), yet recent evidence of dispersing individuals on the island, coupled with the âdoggedâ momentum of coyote range expansion across North America, suggests a Long Island coyote population is close at hand. We highlighted the fleeting opportunity to takes advantage of this natural experiment by developing a multidisciplinary research framework to investigate the ecological and social impacts of the coyote, pre- and post- range expansion. We reviewed coyote spatial ecology, community ecology, and human dimensions research and identified three components of future investigation: predicting future occupancy, monitoring colonization, testing hypotheses of trophic cascades by leveraging and expanding existing ecological data, and exploring attitudes towards coyotes to better understand and mitigate human-wildlife conflicts. Each proposed component will integrate for a comprehensive investigation to advance theory and applied management of northeastern coyotes
A posteriori inclusion of parton density functions in NLO QCD final-state calculations at hadron colliders: The APPLGRID Project
A method to facilitate the consistent inclusion of cross-section measurements
based on complex final-states from HERA, TEVATRON and the LHC in proton parton
density function (PDF) fits has been developed. This can be used to increase
the sensitivity of LHC data to deviations from Standard Model predictions. The
method stores perturbative coefficients of NLO QCD calculations of final-state
observables measured in hadron colliders in look-up tables. This allows the
posteriori inclusion of parton density functions (PDFs), and of the strong
coupling, as well as the a posteriori variation of the renormalisation and
factorisation scales in cross-section calculations.
The main novelties in comparison to original work on the subject are the use
of higher-order interpolation, which substantially improves the trade-off
between accuracy and memory use, and a CPU and computer memory optimised way to
construct and store the look-up table using modern software tools.
It is demonstrated that a sufficient accuracy on the cross-section
calculation can be achieved with reasonably small look-up table size by using
the examples of jet production and electro-weak boson (Z, W) production in
proton-proton collisions at a center-of-mass energy of 14 TeV at the LHC.
The use of this technique in PDF fitting is demonstrated in a PDF-fit to HERA
data and simulated LHC jet cross-sections as well as in a study of the jet
cross-section uncertainties at various centre-of-mass energies
The mitogen-activated protein kinome from Anopheles gambiae: identification, phylogeny and functional characterization of the ERK, JNK and p38 MAP kinases
<p>Abstract</p> <p>Background</p> <p><it>Anopheles gambiae </it>is the primary mosquito vector of human malaria parasites in sub-Saharan Africa. To date, three innate immune signaling pathways, including the nuclear factor (NF)-kappaB-dependent Toll and immune deficient (IMD) pathways and the Janus kinase/signal transducers and activators of transcription (Jak-STAT) pathway, have been extensively characterized in <it>An. gambiae</it>. However, in addition to NF-kappaB-dependent signaling, three mitogen-activated protein kinase (MAPK) pathways regulated by JNK, ERK and p38 MAPK are critical mediators of innate immunity in other invertebrates and in mammals. Our understanding of the roles of the MAPK signaling cascades in anopheline innate immunity is limited, so identification of the encoded complement of these proteins, their upstream activators, and phosphorylation profiles in response to relevant immune signals was warranted.</p> <p>Results</p> <p>In this study, we present the orthologs and phylogeny of 17 <it>An. gambiae </it>MAPKs, two of which were previously unknown and two others that were incompletely annotated. We also provide detailed temporal activation profiles for ERK, JNK, and p38 MAPK in <it>An. gambiae </it>cells <it>in vitro </it>to immune signals that are relevant to malaria parasite infection (human insulin, human transforming growth factor-beta1, hydrogen peroxide) and to bacterial lipopolysaccharide. These activation profiles and possible upstream regulatory pathways are interpreted in light of known MAPK signaling cascades.</p> <p>Conclusions</p> <p>The establishment of a MAPK "road map" based on the most advanced mosquito genome annotation can accelerate our understanding of host-pathogen interactions and broader physiology of <it>An. gambiae </it>and other mosquito species. Further, future efforts to develop predictive models of anopheline cell signaling responses, based on iterative construction and refinement of data-based and literature-based knowledge of the MAP kinase cascades and other networked pathways will facilitate identification of the "master signaling regulators" in biomedically important mosquito species.</p
Evaluation of Steelhead Kelt Passage into the Bonneville Dam Second Powerhouse Corner Collector Prior to the Juvenile Migration Seasons, 2007 and 2008
This report documents the results of a steelhead kelt passage study conducted by the PNNL for the U.S. Army Corps of Engineers at Bonneville Dam in early spring 2007 and 2008. At the Second Powerhouse, a surface flow outlet called the corner collector (B2CC) may be an effective non-turbine passage route for steelhead kelt moving downstream in early spring before the main juvenile emigration season. The goal of this project was to inform management decisions regarding B2CC operations by estimating the number of kelt using the B2CC for downstream passage at Bonneville Dam prior to the juvenile spring migration season. Variation in migration behaviors used by Arctic Terns (Sterna paradisaea) breeding across a wide latitudinal gradient
Arctic Terns (Sterna paradisaea) share a few routes to undertake the longest annual migrations of any organism. To understand how the wide spatial range of their breeding colonies may affect their migration strategies (e.g., departure date), we tracked 53 terns from five North American colonies distributed across 30° of latitude and 90° of longitude. While birds from all colonies arrived in Antarctic waters at a similar time, terns nesting in the Arctic colonies migrated back north more slowly and arrived to their breeding grounds later than those nesting in the colony farther south. Arrival dates in Antarctic waters coincided with the start of favorable foraging conditions (i.e., increased ocean productivity), and similarly arrival dates at breeding colonies coincided with the start of local favorable breeding conditions (i.e., disappearance of snow and ice). Larger birds followed a more direct southbound migration route than smaller birds. On both southbound and northbound migrations, daily distances traveled declined as time spent in contact with the ocean increased, suggesting a trade-off between resting/foraging and traveling. There was more unexplained variation in behavior among individuals than among colonies, and one individual had a distinctive stop around Brazil. Terns nesting in the Arctic have a narrow time window for breeding that will likely increase with continuing declines in sea ice and snow. Departing Arctic Terns likely have few clues about the environmental conditions they will encounter on arrival, and their response to environmental changes at both poles may be assisted by large individual variation in migration strategy
Process Evaluation in Action: Lessons Learned from Alabama REACH 2010
The CDC-funded Alabama Racial and Ethnic Approaches to Community Health (REACH 2010) project is designed to reduce and eliminate disparities in breast and cervical cancer between African American and white women in six rural and three urban counties in Alabama. In this manuscript, we report on the development, implementation, results, and lessons learned from a process evaluation plan initiated during the Phase I planning period of the Alabama REACH 2010 program. The process evaluation plan for Alabama REACH 2010 focused on four main areas of activity that coincided with program objectives: assessing coalition development, building community capacity, conducting a needs assessment, and developing a community action plan. Process evaluation findings indicated that progress made by Alabama REACH 2010 was due, in part, to evaluative feedback. We conclude that process evaluation can be a powerful tool for monitoring and measuring the administrative aspect of a complex, community-based health intervention
ACE Inhibitor and Angiotensin Receptor Blocker Use During Pregnancy:Data from the ESC Registry Of Pregnancy and Cardiac Disease (ROPAC)
Angiotensin-converting enzyme inhibitors (ACE-Is) and angiotensin receptor blockers (ARBs) are not recommended during the second and third trimester because of the significant risk of congenital anomalies associated with their use. However, data are scarce, especially regarding their use in the first trimester and about the impact of stopping just before pregnancy. Our study illustrates the profile of the women who used ACE-Is or ARBs during pregnancy and evaluates the impact on perinatal outcomes. The Registry of Pregnancy and Cardiac Disease is a prospective, global registry of pregnancies in women with structural heart disease. Outcomes were compared between women who used ACE-Is or ARBs and those who did not. Multivariable regression analysis was performed to assess the effect of ACE-I or ARB use on the occurrence of congenital anomalies. ACE-Is (n = 35) and/or ARBs (n = 8) were used in 42 (0.7%) of the 5,739 Registry of Pregnancy and Cardiac Disease pregnancies. Women who used ACE-Is or ARBs more often came from a low-or-middle-income country (57% vs 40%, p = 0.021), had chronic hypertension (31% vs 6%, p <0.001), or a left ventricular ejection fraction <40% (33% vs 4%, p <0.001). In the multivariable analysis, ACE-I use during the first trimester was associated with an increased risk of congenital anomaly (odds ratio 3.2, 95% confidence interval 1.0 to 9.6). Therefore, ACE-Is should be avoided during pregnancy, also in the first trimester, because of a higher risk of congenital anomalies. However, there is no need to stop long before pregnancy. Preconception counseling is crucial to discuss the potential risks of these medications, to evaluate the clinical condition and, if possible, to change or stop the medication.</p
Unique Clinical and Pathological Features in HLA-DRB1*0401ârestricted MBP 111â129âspecific Humanized TCR Transgenic Mice
Amino acid residues 111â129 represent an immunodominant epitope of myelin basic protein (MBP) in humans with human leukocyte antigen (HLA)-DRB1*0401 allele(s). The MBP 111â129âspecific T cell clone MS2-3C8 was repeatedly isolated from a patient with multiple sclerosis (MS), suggesting an involvement of MS2-3C8 T cells in the pathogenesis. To address the pathogenic potential of the MS2-3C8 T cell clone, we generated transgenic (Tg) mice expressing its T cell receptor and restriction element, HLA-DRB1*0401, to examine the pathogenic characteristics of MS2-3C8 Tg T cells by adoptive transfer into HLA-DRB1*0401 Tg mice. In addition to the ascending paralysis typical of experimental autoimmune encephalomyelitis, mice displayed dysphagia due to restriction in jaw and tongue movements and abnormal gait. In accordance with the clinical phenotype, infiltrates of MS2-3C8 Tg T cells and inflammatory lesions were predominantly located in the brainstem and the cranial nerve roots in addition to the spinal cord and spinal nerve roots. Together, these data suggest a pathogenic role of MBP-specific T cells in inflammatory demyelination within the brainstem and cranial nerve roots during the progression of MS. This notion may help to explain the clinical and pathological heterogeneity of MS
