    Elephant Endotheliotropic Herpesvirus Impact in the European Asian Elephant (Elephas maximus) Population: Are Hereditability and Zoo-Associated Factors Linked with Mortality?

    EEHV is a ubiquitous virus, which most likely has co-evolved with elephants and is shed by healthy individuals and maintained in the herds. Yet, the factors determining calf susceptibility to the virus remain unknown. Here, we explored the impact of EEHV-HD in the European captive Asian elephant population in a retrospective statistical study spanning the last 35 years. We show that EEHV-HD was implicated in more than half of all deaths recorded in calves older than one months old. Moreover, the median age across EEHV-HD fatalities was significantly lower compared to other death causes. Finally, we investigated if heredity and zoo-associated factors could be linked to a higher susceptibility of calves to this disease. We used a univariable logistic regression model to evaluate if either fathers, mothers, or zoos could, separately, be considered as risk factors to the development of the disease. Afterwards, we used a two multivariable model, combining: (1) fathers and zoos, and (2) mothers and zoos. Overall, we found that two fathers, one mother, and four zoos had three or more times higher risk of their calves becoming sick when compared to all others, pointing us to the presence of a management or environmental element, which can have paternal and maternal influence and leads to calf susceptibility or resistance to EEHV-HD

    Prediction of total quarter milk somatic cell counts based on foremilk sampling

    Determination of somatic cell count (SCC) is used worldwide in dairy practice to describe the hygienic status of the milk and the udder health of cows. When SCC is tested on a quarter level to detect single quarters with high SCC levels of cows for practical reasons, mostly foremilk samples after prestimulation (i.e. cleaning of the udder) are used. However, SCC is usually different in different milk fractions. Therefore, the goal of this study was the investigation of the use of foremilk samples for the estimation of total quarter SCC. A total of 378 milkings in 19 dairy cows were performed with a special milking device to drain quarter milk separately. Foremilk samples were taken after udder stimulation and before cluster attachment. SCC was measured in foremilk samples and in total quarter milk. Total quarter milk SCC could not be predicted precisely from foremilk SCC measurements. At relatively high foremilk SCC levels (>300Ă—103 cells/ml) foremilk SCC were higher than total quarter milk. At around (50-300)Ă—103 cells/ml foremilk and total quarter SCC did not differ considerably. Most interestingly, if foremilk SCC was lower than 50Ă—103 cells/ml the total quarter SCC was higher than foremilk SCC. In addition, individual cows showed dramatic variations in foremilk SCC that were not very well related to total quarter milk SCC. In conclusion, foremilk samples are useful to detect high quarter milk SCC to recognize possibly infected quarters, only if precise cell counts are not required.     Innovative e-Learning Training Modules to Improve Animal Welfare during Transport and Slaughter of Pigs: A Pretest–Posttest Study to Pre-Evaluate the General Didactical Concept

    In addition to the information on the possession of a certificate of competence, there are no concrete obligations for repetitive training for personnel handling live animals at transport and slaughter. Deficiencies in the animal-welfare-friendly handling of pigs are known. The developed pilot modules “Handling of pigs” and “Electrical stunning” were tested in a pretest–posttest study in German and Romanian using questions of knowledge before and after the implementation of the modules. In this study, 45 and 46 datasets of participants could be analyzed. The mean percentages of correctly answered questions in the posttest increased by 5.6% in the module “Handling of pigs” and by 10.6% in the module “Electrical stunning”. A significant interaction was found for the language match and trend categories in the module “Handling of pigs”. No Romanian native speaker had a positive trend in this module. For both modules separately, participant education level significantly interacted with the language match and the presence or absence of a certificate of competence. Comparing the percentages of the correct given answers, significant interactions in the subgroups were more common in the module “Electrical stunning”. One question in “Electrical stunning” was correctly answered significantly more often in the posttest. Because of the positive mean trends of knowledge within this pre-evaluation, we assume the didactical concept was suitable for our target groups. Holders of a certificate of competence also gave more correct answers in the post-test. This underlines the importance of repetitive training.     Sensitivity and Specificity for the Detection of Clinical Mastitis by Automatic Milking Systems in Bavarian Dairy Herds

    In automatic milking systems (AMSs), the detection of clinical mastitis (CM) and the subsequent separation of abnormal milk should be reliably performed by commercial AMSs. Therefore, the objectives of this cross-sectional study were (1) to determine the sensitivity (SN) and specificity (SP) of CM detection of AMS by the four most common manufacturers in Bavarian dairy farms, and (2) to identify routinely collected cow data (AMS and monthly test day data of the regional Dairy Herd Improvement Association (DHIA)) that could improve the SN and SP of clinical mastitis detection. Bavarian dairy farms with AMS from the manufacturers DeLaval, GEA Farm Technologies, Lely, and Lemmer-Fullwood were recruited with the aim of sampling at least 40 cows with clinical mastitis per AMS manufacturer in addition to clinically healthy ones. During a single farm visit, cow-level milking information was first electronically extracted from each AMS and then all lactating cows examined for their udder health status in the barn. Clinical mastitis was defined as at least the presence of visibly abnormal milk. In addition, available DHIA test results from the previous six months were collected. None of the manufacturers provided a definition for clinical mastitis (i.e., visually abnormal milk), therefore, the SN and SP of AMS warning lists for udder health were assessed for each manufacturer individually, based on the clinical evaluation results. Generalized linear mixed models (GLMMs) with herd as random effect were used to determine the potential influence of routinely recorded parameters on SN and SP. A total of 7411 cows on 114 farms were assessed; of these, 7096 cows could be matched to AMS data and were included in the analysis. The prevalence of clinical mastitis was 3.4% (239 cows). When considering the 95% confidence interval (95% CI), all but one manufacturer achieved the minimum SN limit of >80%: DeLaval (SN: 61.4% (95% CI: 49.0%–72.8%)), GEA (75.9% (62.4%–86.5%)), Lely (78.2% (67.4%–86.8%)), and Lemmer-Fullwood (67.6% (50.2%–82.0%)). However, none of the evaluated AMSs achieved the minimum SP limit of 99%: DeLaval (SP: 89.3% (95% CI: 87.7%–90.7%)), GEA (79.2% (77.1%–81.2%)), Lely (86.2% (84.6%–87.7%)), and Lemmer-Fullwood (92.2% (90.8%–93.5%)). All AMS manufacturers’ robots showed an association of SP with cow classification based on somatic cell count (SCC) measurement from the last two DHIA test results: cows that were above the threshold of 100,000 cells/mL for subclinical mastitis on both test days had lower chances of being classified as healthy by the AMS compared to cows that were below the threshold. In conclusion, the detection of clinical mastitis cases was satisfactory across AMS manufacturers. However, the low SP will lead to unnecessarily discarded milk and increased workload to assess potentially false-positive mastitis cases. Based on the results of our study, farmers must evaluate all available data (test day data, AMS data, and daily assessment of their cows in the barn) to make decisions about individual cows and to ultimately ensure animal welfare, food quality, and the economic viability of their farm

    Neuropathological and molecular comparison between clinical and asymptomatic bovine spongiform encephalopathy cases

    Interest in the proper neuropathological and molecular characterization of bovine spongiform encephalopathy (BSE) has increased since asymptomatic and atypical cases were detected in the cattle population by active disease surveillance. In this respect we investigated a total of 95 confirmed BSE cases originating from different active and passive surveillance categories (clinical suspects, emergency-slaughter, fallen stock and routinely slaughter) in Switzerland for their neuropathological and molecular phenotype. We looked for measurable differences between these categories in lesion profile, severity of spongiform change, degree of astrocytosis as well as immunohistochemical and molecular patterns of the disease-associated isoform of the prion protein (PrPd) in the caudal brainstem. Our results indicate significantly higher intensities of spongiform change in clinically affected compared to asymptomatic BSE cases. Similar effects were in trend observed for the intensities of PrPd deposition and astrocytosis, whereas the frequencies of morphological PrPd types and the molecular patterns in Western immunoblot were not different. Importantly, none of the animals included in this study revealed features of atypical BSE. Taken together, this study suggests that both clinically affected as well as asymptomatic Swiss BSE cases in cattle share the neuropathological and molecular phenotype of classical BSE and that asymptomatic classical BSE cases are at a pre-clinical stage of the disease rather than representing a true sub-clinical form of BS

    Using scenario tree modelling for targeted herd sampling to substantiate freedom from disease

    Background: In order to optimise the cost-effectiveness of active surveillance to substantiate freedom from disease, a new approach using targeted sampling of farms was developed and applied on the example of infectious bovine rhinotracheitis (IBR) and enzootic bovine leucosis (EBL) in Switzerland. Relevant risk factors (RF) for the introduction of IBR and EBL into Swiss cattle farms were identified and their relative risks defined based on literature review and expert opinions. A quantitative model based on the scenario tree method was subsequently used to calculate the required sample size of a targeted sampling approach (TS) for a given sensitivity. We compared the sample size with that of a stratified random sample (sRS) with regard to efficiency. Results: The required sample sizes to substantiate disease freedom were 1,241 farms for IBR and 1,750 farms for EBL to detect 0.2% herd prevalence with 99% sensitivity. Using conventional sRS, the required sample sizes were 2,259 farms for IBR and 2,243 for EBL. Considering the additional administrative expenses required for the planning of TS, the risk-based approach was still more cost-effective than a sRS (40% reduction on the full survey costs for IBR and 8% for EBL) due to the considerable reduction in sample size. Conclusions: As the model depends on RF selected through literature review and was parameterised with values estimated by experts, it is subject to some degree of uncertainty. Nevertheless, this approach provides the veterinary authorities with a promising tool for future cost-effective sampling designs

    Representative Seroprevalences of Brucellosis in Humans and Livestock in Kyrgyzstan

    Kyrgyzstan reported 77.5 new human brucellosis cases per 100,000 people in 2007, which is one of the highest incidences worldwide. In Kyrgyzstan, the currently used diagnostic tests in humans and animals are the Rose Bengal Test and the Huddleson test. A national representative cross-sectional study using cluster sampling proportional to size in humans, cattle, sheep, and goats was undertaken to assess the apparent seroprevalence in humans and animals. A total of 4,936 livestock sera and 1,774 human sera were tested in Naryn, Chuy, and Osh Oblasts. The overall apparent seroprevalences of brucellosis were 8.8% in humans (95% CI 4.5-16.5), 2.8% (95% CI 1.6-4.9%) in cattle, 3.3% (95% CI 1.5-6.9%) in sheep, and 2.5% (95% CI 1.4-4.5%) in goats. Naryn Oblast had the highest seroprevalences in humans and sheep. More men than women were seropositive (OR=1.96; P<0.001). Human seroprevalence was significantly associated with small ruminant seroprevalence but not with cattle seroprevalence. Annual incidence of human brucellosis exposure, measured by serological tests, was more than ten times higher than the annual incidence of reported clinical brucellosis cases. This indicates an under-reporting of human brucellosis cases, even if only a fraction of seropositive people have clinical symptoms. In conclusion, this study confirms the high seroprevalence of brucellosis in Kyrgyzstan and warrants rapid effective intervention, among others, by mass vaccination of sheep and goats but also of cattl

    Retrospective Study of Salinomycin Toxicosis in 66 Cats

    We examined 66 cats with salinomycin intoxication. Salinomycin caused different LMN signs of varying degrees of severity in all cases. Changes in blood work were unspecific, with the most frequent being increased serum creatine kinase activity, leukocytosis, and increased liver enzymes. Pathological electrodiagnostic findings: fibrillation potentials and positive sharp waves were detected in 10 cases, motor nerve conductance velocity was mildly decreased in 8/12 cats, and sensory nerve conductance velocity and repetitive nerve stimulation were normal in all examined cases. In five cases the peripheral neuropathy was confirmed by pathohistology. Fluid therapy and supportive care were used as therapy and 52 cats recovered completely. The probability for complete remission was significantly different between mildly and severely affected cases. It seems that the severity of clinical signs and prognosis correlate well with the amount of toxin ingested. We conclude that early recognition and decontamination combined with supportive care results in complete recovery
