    Partition of unity interpolation using stable kernel-based techniques

    In this paper we propose a new stable and accurate approximation technique which is extremely effective for interpolating large scattered data sets. The Partition of Unity (PU) method is performed considering Radial Basis Functions (RBFs) as local approximants and using locally supported weights. In particular, the approach consists in computing, for each PU subdomain, a stable basis. Such technique, taking advantage of the local scheme, leads to a significant benefit in terms of stability, especially for flat kernels. Furthermore, an optimized searching procedure is applied to build the local stable bases, thus rendering the method more efficient

    We present an algorithm to approximate large dataset by Radial Basis Function (RBF) techniques. The method couples a fast domain decomposition procedure with a localized stabilization method. The resulting algorithm can efficiently deal with large problems and it is robust with respect to the typical instability of kernel methods

    Late movement of basin-edge lobate scarps on Mercury

    Basin-edge lobate scarps are a sub-type of tectonic shortening structure on the surface of Mercury that have formed at the edge of volcanic units that fill or partly fill impact basins. We have performed a global survey of these features and find that they are widespread in basins across the planet. We obtained model ages from crater size–frequency distribution analysis for a subset of our surveyed basins, for both the smooth plains infill and for the last resolvable tectonic activity on the associated basin-edge scarps. Our results indicate that some of these lobate scarps were still accumulating strain in the late Mansurian (approximately 1 Ga). From a photogeological assessment, we find that the orientations of these basin-edge lobate scarps are similar to those reported for the global population of lobate scarps in earlier studies, appearing to align ∌north–south at low latitudes and ∌east–west at higher latitudes. However, reassessing these landforms’ orientation with artificially illuminated topographic data does not allow us to rule out the effect of illumination bias. We propose that these landforms, the result of crustal shortening in response to global contraction, formed along the interface between the basin floor and the smooth plains unit, which acted as a mechanical discontinuity along which shortening strains were concentrated

    Emerging roles of purinergic signaling in anti-cancer therapy resistance

    Cancer is a complex disease with a rapid growing incidence and often characterized by a poor prognosis. Although impressive advances have been made in cancer treatments, resistance to therapy remains a critical obstacle for the improvement of patients outcome. Current treatment approaches as chemo-, radio-, and immuno-therapy deeply affect the tumor microenvironment (TME), inducing an extensive selective pressure on cancer cells through the activation of the immune system, the induction of cell death and the release of inflammatory and damage-associated molecular patterns (DAMPS), including nucleosides (adenosine) and nucleotides (ATP and ADP). To survive in this hostile environment, resistant cells engage a variety of mitigation pathways related to metabolism, DNA repair, stemness, inflammation and resistance to apoptosis. In this context, purinergic signaling exerts a pivotal role being involved in mitochondrial function, stemness, inflammation and cancer development. The activity of ATP and adenosine released in the TME depend upon the repertoire of purinergic P2 and adenosine receptors engaged, as well as, by the expression of ectonucleotidases (CD39 and CD73) on tumor, immune and stromal cells. Besides its well established role in the pathogenesis of several tumors and in host–tumor interaction, purinergic signaling has been recently shown to be profoundly involved in the development of therapy resistance. In this review we summarize the current advances on the role of purinergic signaling in response and resistance to anti-cancer therapies, also describing the translational applications of combining conventional anticancer interventions with therapies targeting purinergic signaling

    A systems biology approach to dissection of the effects of small bicyclic peptidomimetics on a panel of Saccharomyces cerevisiae mutants

    In recent years, an approach called “chemical genetics” has been adopted in drug research to discover and validate new targets and to identify and optimize leads by high throughput screening. In this work, we tested the ability of a library of small peptidomimetics to induce phenotypic effects with functional implications on a panel of strains of the budding yeast Saccharomyces cerevisiae, both wild type and mutants, for respiratory function and multidrug resistance. Further elucidation of the function of these peptidomimetics was assessed by testing the effects of the compound with the most prominent inhibitory activity, 089, on gene expression using DNA microarrays. Pathway analysis showed the involvement of such a molecule in inducing oxidative damage through alterations in mitochondrial functions. Transcriptional experiments were confirmed by increased levels of ROS and activation of mitochondrial membrane potential. Our results demonstrate the influence of a functional HAP1 gene in the performance of S. cerevisiae as a model system

    A methodological approach exploiting modern techniques for forest road network planning

    A well-developed road network allows all forest activities, including wood harvesting, firefighting and recreational activities. However, forest road construction and maintenance involve economic and environmental costs. For these reasons, forest road network planning is a fundamental phase of forest management, maximising the benefits and reducing costs and impacts. Thanks to modern technologies in data collection both for terrestrial and forest characteristics, new methods and tools have been developed to improve and facilitate road planning. The aim of this study was the development of a Decision Support System for helping managers during forest road network planning, exploiting Multi-Criteria Analysis, an Analytic Hierarchy Process and Geographic Information Systems. Three steps characterised the study: Þ an in-depth survey of the existing forest road network Þ an accessibility evaluation, based on a commonly applied Italian definition, taking into account the morphological characteristics of the land Þ an estimation of the accessibility requirements through the analysis of experts’ opinions, defined as Road Needs Index, based on different factors These phases were applied to a forest property located in northern Italy, and some improvements were proposed simulating a manager’s approach during planning. The results showed interesting features in accessibility evaluation, which identified three different classes of accessibility, represented in a map. The estimation of Road Needs Index assigned a class regarding road requirements to each forest management unit: »low«, »medium«, »high« and »very high«. This information was merged, becoming a useful tool to identify the forest areas with the highest problems in relation to the forest road network

    We investigate adaptivity issues for the approximation of Poisson equations via radial basis function-based partition of unity collocation. The adaptive residual subsampling approach is performed with quasi-uniform node sequences leading to a flexible tool which however might suffer from numerical instability due to ill-conditioning of the collocation matrices. We thus develop a hybrid method which makes use of the so-called variably scaled kernels. The proposed algorithm numerically ensures the convergence of the adaptive procedure

    Study of the bone pathology in early mucolipidosis II (I-cell disease).

    Histological examination of the bones obtained on autopsy of a 5-month-old child with mucolipidosis II (I-cell disease) revealed inhibition of the growth plate calcification with defective vascular invasion and signs of hyperparathyroidism. These findings are the chondro-osseous basis of the early radiological ricket-like appearance of bones in the neonatal period or soon thereafter. Whether the early skeletal abnormalities of mucolipidosis II result from a primary enzymatic defect of cartilage and bone cells or from factors controlling bone metabolism deserves further study

    WIYN Open Cluster Study 1: Deep Photometry of NGC 188

    We have employed precise V and I photometry of NGC 188 at WIYN to explore the cluster luminosity function (LF) and study the cluster white dwarfs (WDs). Our photometry is offset by V = 0.052 (fainter) from Sandage (1962) and Eggen & Sandage (1969). All published photometry for the past three decades have been based on these two calibrations, which are in error by 0.05 +- 0.01. We employ the Pinsonneault etal (1998) fiducial main sequence to derive a cluster distance modulus of 11.43 +- 0.08. We report observations that are >= 50% complete to V = 24.6 and find that the cluster central-field LF peaks at M_I ~ 3 to 4. This is unlike the solar neighborhood LF and unlike the LFs of dynamically unevolved portions of open and globular clusters, which rise continuously until M_I ~ 9.5. Although we find that >= 50% of the unresolved cluster objects are multiple systems, their presence cannot account for the shape of the NGC 188 LF. For theoretical reasons (Terlevich 1987; Vesperini & Heggie 1997) having to do with the survivability of NGC 188 we believe the cluster is highly dynamically evolved and that the missing low luminosity stars are either in the cluster outskirts or have left the cluster altogether. We identify nine candidate WDs, of which we expect three to six are bona fide cluster WDs. The luminosities of the faintest likely WD indicates an age (Bergeron, Wesemael, & Beauchamp 1995) of 1.14 +- 0.09 Gyrs. This is a lower limit to the cluster age and observations probing to V = 27 or 28 will be necessary to find the faintest cluster WDs and independently determine the cluster age. While our age limit is not surprising for this ~6 Gyr old cluster, our result demonstrates the value of the WD age technique with its very low internal errors.     This article offers a comprehensive and systematic review of the literature on Global Value Chains (GVCs). The GVC framework has received growing attention in the last decade, providing theoretical concepts and analytical tools to understand and assess patterns of value creation in view of the new international division of labour. In this area, a broad overlap of research interest exists with the international business (IB) literature. Yet, few interactions between the two fields of study have been recorded so far. Performing a quantitative content analysis on all academic publications on GVCs in the period 1994−2018, this paper describes the evolution of GVC studies, emphasizing points of contact and potential synergies with the IB literature. It also identifies research opportunities along the four key dimensions of the GVC framework: geographical and industrial scope, governance, upgrading, and institutional context
