    Efficacy of roxithromycin with gamma globulin in children with mycoplasma pneumonia and its effect on immunity

    Purpose: To determine the efficacy of roxithromycin plus gamma globulin in the treatment of children with mycoplasma pneumonia (MPP) and its effect on immune function.Methods: From January 2019 to January 2021, 100 children with MPP assessed for eligibility in Qingdao Women and Children's Hospital, Shandong Province, China, were recruited and randomized (1:1) to receive either gamma globulin (control group) or roxithromycin plus gamma globulin (study group). Levels of tumor necrosis factor (TNF)-α, immunoglobulin (Ig)A, IgM, and IgG were evaluated. Clinical indices, including fever reduction, cough disappearance, duration of hospital stay, etc were also assessed.Results: The study group had a significantly higher clinical efficacy (88 %) than the control group (68 %) (p < 0.05). After treatment, patients in the study group showed lower levels of tumor necrosis factor (TNF)-α than those in the control group (p < 0.05). The eligible patients given roxithromycin plus gamma globulin showed significantly higher levels of immunoglobulin (Ig)A, IgM, and IgG versus those given gamma globulin alone (p < 0.05). Patients in the study group had a shorter time lapse before fever reduction, cough disappearance, lung sign disappearance, and duration of hospital stay than those in the control group (p < 0.05).Conclusion: Roxithromycin plus gamma globulin demonstrate significant benefits in the treatment of children with MPP by mitigating inflammatory response, enhancing immune function, and also significantly alleviating clinical symptoms. Thus, the combination treatment shows good potentials for use in clinical practice

    Molecular Dynamic Simulation to Explore the Molecular Basis of Btk-PH Domain Interaction with Ins(1,3,4,5)P4

    Bruton’s tyrosine kinase contains a pleckstrin homology domain, and it specifically binds inositol 1,3,4,5-tetrakisphosphate (Ins(1,3,4,5)P4), which is involved in the maturation of B cells. In this paper, we studied 12 systems including the wild type and 11 mutants, K12R, S14F, K19E, R28C/H, E41K, L11P, F25S, Y40N, and K12R-R28C/H, to investigate any change in the ligand binding site of each mutant. Molecular dynamics simulations combined with the method of molecular mechanics/Poisson-Boltzmann solvent-accessible surface area have been applied to the twelve systems, and reasonable mutant structures and their binding free energies have been obtained as criteria in the final classification. As a result, five structures, K12R, K19E, R28C/H, and E41K mutants, were classified as “functional mutations,” whereas L11P, S14F, F25S, and Y40N were grouped into “folding mutations.” This rigorous study of the binding affinity of each of the mutants and their classification provides some new insights into the biological function of the Btk-PH domain and related mutation-causing diseases

    A Natural Monoterpene Enol for Dispersive Liquid–Liquid Microextraction Based on Solidification of Floating Organic Droplets for Determination of Benzophenone Compounds in Water Samples

    In the current study, an effective and simple procedure of extraction for the four benzophenone compounds from water samples was achieved by dispersive liquid–liquid microextraction (DLLME) based on the solidification of floating organic droplets (SFO) with a natural monoterpene enol as the extractant. As a natural, high solidification point, inexpensive, and environmentally friendly hydrophobic solvent, α−terpineol was selected firstly as an extractant for DLLME and could be collected and transferred easily after extraction by solidification at a lower temperature. Several main parameters closely related to extraction efficiencies, such as volume of extractant, extraction time, pH and salt concentration of the sample solution, temperature, and time of the solidification process, were investigated in detail. The results showed that the established method had good extraction performance for benzophenone compounds with enrichment factors in the range of 29–47. Furthermore, the linearities were over the range of 2–2000 µg/L, and the limits of detection were 0.12–0.53 µg/L for four benzophenone compounds. The recoveries ranged from 80.2% to 108.4%, with RSDs (intra- and inter-assay) less than 8.5%. At last, the method applicability was investigated by the determination of the benzophenone compounds in aqueous solutions, and satisfactory recoveries (83.0–107.0%) were acquired. Taken together, α−terpineol, a natural monoterpene enol, was first used as an extractant of DLLME-SFO, which provided an alternative method with simplicity and rapidity for the determination of benzophenone compounds in aqueous samples

The study subjects are patients over 18 years of age with differentiated thyroid cancer who were given radioactive iodine-131 therapy as well as endocrine therapy after radical surgery for thyroid cancer. The intervention group will receive MTM-mhealth, and the realization of health education will rely on the smart terminal WeChat platform. Routine discharge education will be given to the control group at discharge. The primary outcome will be change in thyroid-stimulating hormone (TSH) from baseline and at 3 and 6 months of follow-up, and secondary outcomes will include change in self-management behavior, social cognitive and psychological, and metabolic control.DiscussionThis study will explore a feasible mHealth intervention program applied to a population of DTC patients using the Multi-theory model of health behavior change (MTM) as a guide, with the aim of evaluating the MTM-based intervention program for clinical outcome improvement in DTC patients, as well as determining the effectiveness of the MTM-based intervention program in improving self-management skills in DTC patients.
