All-trans retinoic acid ameliorates glycemic control in diabetic mice via modulating pancreatic islet production of vascular endothelial growth factor-A.
Patients with type 1 diabetes mellitus are associated with impairment in vitamin A metabolism. This study evaluated whether treatment with retinoic acid, the biologically active metabolite of vitamin A, can ameliorate diabetes. All-trans retinoic acid (atRA) was used to treat streptozotocin (STZ)-induced diabetic mice which revealed atRA administration ameliorated blood glucose levels of diabetic mice. This hyperglycemic amelioration was accompanied by an increase in the amount of β cells co-expressed Pdx1 and insulin and by restoration of the vascular laminin expression. The atRA-induced production of vascular endothelial growth factor-A from the pancreatic islets was possibly the key factor that mediated the restoration of islet vascularity and recovery of β-cell mass. Furthermore, the combination of islet transplantation and atRA administration significantly rescued hyperglycemia in diabetic mice. These findings suggest that vitamin A derivatives can potentially be used as a supplementary treatment to improve diabetes management and glycemic control
Bose condensation of upper-branch exciton-polaritons in a transferrable microcavity
Exciton-polaritons are composite bosonic quasiparticles arising from the
strong coupling of excitonic transitions and optical modes. Exciton-polaritons
have triggered wide exploration in the past decades not only due to their rich
quantum phenomena such as superfluidity, superconductivity and quantized
vortices but also due to their potential applications for unconventional
coherent light sources and all-optical control elements. Here, we report the
observation of Bose-Einstein condensation of the upper polariton branch in a
transferrable WS monolayer microcavity. Near the condensation threshold, we
observe a nonlinear increase in upper polariton intensity. This sharp increase
in intensity is accompanied by a decrease of the linewidth and an increase of
the upper polariton temporal coherence, all of which are hallmarks of
Bose-Einstein condensation. By simulating the quantum Boltzmann equation, we
show that the upper polariton condensation only occurs for a particular range
of particle density. We can attribute the creation of Bose condensation of the
upper polariton to the following requirements: 1) the upper polariton is more
excitonic than the lower one; 2) there is relatively more pumping in the upper
branch; and 3) the conversion time from the upper to the lower polariton branch
is long compared to the lifetime of the upper polaritons
Label-free quantitative proteomics of CD133-positive liver cancer stem cells
Abstract
Background
CD133-positive liver cancer stem cells, which are characterized by their resistance to conventional chemotherapy and their tumor initiation ability at limited dilutions, have been recognized as a critical target in liver cancer therapeutics. In the current work, we developed a label-free quantitative method to investigate the proteome of CD133-positive liver cancer stem cells for the purpose of identifying unique biomarkers that can be utilized for targeting liver cancer stem cells. Label-free quantitation was performed in combination with ID-based Elution time Alignment by Linear regression Quantitation (IDEAL-Q) and MaxQuant.
Results
Initially, IDEAL-Q analysis revealed that 151 proteins were differentially expressed in the CD133-positive hepatoma cells when compared with CD133-negative cells. We then analyzed these 151 differentially expressed proteins by MaxQuant software and identified 10 significantly up-regulated proteins. The results were further validated by RT-PCR, western blot, flow cytometry or immunofluorescent staining which revealed that prominin-1, annexin A1, annexin A3, transgelin, creatine kinase B, vimentin, and EpCAM were indeed highly expressed in the CD133-positive hepatoma cells.
Conclusions
These findings confirmed that mass spectrometry-based label-free quantitative proteomics can be used to gain insights into liver cancer stem cells.http://deepblue.lib.umich.edu/bitstream/2027.42/113089/1/12953_2012_Article_407.pd
Study on the Process of Pressing Camellia Oleifera Seeds with Daily 10t Twin-screw Oil Press
In order to improve the quality and reduce the loss of the nutritional content of camellia oil with the traditional press, this paper studied the process of pressing Camellia Oleifera Seeds with 10t/d Twin-Screw Oil Press. The results demonstrated that the best conditions for this article were: 9% moisture content of Camellia Oleifera seed kernels and 8% content of Camellia oleifera seed shell. The oil content of Camellia oleifera seed cake was reduced to 2.49%, and the produced camellia oil could meet the standard of first -class oil. In comparison to the camellia oil sold on the market, the retention rate of α-tocopherol in the camellia oil obtained through this process was higher, and the nutritional value of camellia oil has been further improved, which could provide technical guidance for the high-value processing of camellia oil in China
Analysis of EGFR signaling pathway in nasopharyngeal carcinoma cells by quantitative phosphoproteomics
<p>Abstract</p> <p>Background</p> <p>The epidermal growth factor receptor (EGFR) is usually overexpressed in nasopharyngeal carcinoma (NPC) and is associated with pathogenesis of NPC. However, the downstream signaling proteins of EGFR in NPC have not yet been completely understood at the system level. The aim of this study was identify novel downstream proteins of EGFR signaling pathway in NPC cells.</p> <p>Results</p> <p>We analyzed EGFR-regulated phosphoproteome in NPC CNE2 cells using 2D-DIGE and mass spectrometry analysis after phosphoprotein enrichment. As a result, 33 nonredundant phosphoproteins including five known EGFR-regulated proteins and twenty-eight novel EGFR-regulated proteins in CNE2 were identified, three differential phosphoproteins were selectively validated, and two differential phosphoproteins (GSTP1 and GRB2) were showed interacted with phospho-EGFR. Bioinformatics analysis showed that 32 of 33 identified proteins contain phosphorylation modification sites, and 17 identified proteins are signaling proteins. GSTP1, one of the EGFR-regulated proteins, associated with chemoresistance was analyzed. The results showed that GSTP1 could contribute to paclitaxel resistance in EGF-stimulated CNE2 cells. Furthermore, an EGFR signaling network based on the identified EGFR-regulated phosphoproteins were constructed using Pathway Studio 5.0 software, which includes canonical and novel EGFR-regulated proteins and implicates the possible biological roles for those proteins.</p> <p>Conclusion</p> <p>The data not only can extend our knowledge of canonical EGFR signaling, but also will be useful to understand the molecular mechanisms of EGFR in NPC pathogenesis and search therapeutic targets for NPC.</p
Effects of the Timing of Note Taking on Repeated Listening among Advanced Chinese Japanese Learners: Focusing on the Timing of Note Taking and Working Memory Span
This study aimed to investigate the effects of the timing of note taking on repeated listening for advanced Chinese learners of Japanese. The two independent variables were participants’ working memory capacity and the timing of the note taking. The main results were as follows: In the free recall test, there was a marginal significance that participants with a low working-memory capacity demonstrated better performance when taking notes during the first trail than participants with a high working-memory capacity. However in the fill-in-blank test, regardless of the timing of the note taking, a difference according to the size of working memory capacity was not found. Further, it was found that taking notes during the first trail left a stronger memory trace, whereas taking notes during the second trail was helpful for the understanding of target passages. These results suggest that learners with a smaller memory span can perform better with note taking and learners with a larger memory span are better of taking notes after the first trail

Single-Cell Spatial Transcriptomics Unveils Platelet-Fueled Cycling Macrophages for Kidney Fibrosis.
With the increasing incidence of kidney diseases, there is an urgent need to develop therapeutic strategies to combat post-injury fibrosis. Immune cells, including platelets, play a pivotal role in this repair process, primarily through their released cytokines. However, the specific role of platelets in kidney injury and subsequent repair remains underexplored. Here, the detrimental role of platelets in renal recovery following ischemia/reperfusion injury and its contribution to acute kidney injury to chronic kidney disease transition is aimed to investigated. In this study, it is shown that depleting platelets accelerates injury resolution and significantly reduces fibrosis. Employing advanced single-cell and spatial transcriptomic techniques, macrophages as the primary mediators modulated by platelet signals is identified. A novel subset of macrophages, termed cycling M2, which exhibit an M2 phenotype combined with enhanced proliferative activity is uncovered. This subset emerges in the injured kidney during the resolution phase and is modulated by platelet-derived thrombospondin 1 (THBS1) signaling, acquiring profibrotic characteristics. Conversely, targeted inhibition of THBS1 markedly downregulates the cycling M2 macrophage, thereby mitigating fibrotic progression. Overall, this findings highlight the adverse role of platelet THBS1-boosted cycling M2 macrophages in renal injury repair and suggest platelet THBS1 as a promising therapeutic target for alleviating inflammation and kidney fibrosis
AN ASSESSMENT OF TRADITIONAL UIGHUR MEDICINE IN CURRENT XINJIANG REGION (CHINA)
Background: The main objectives of this study were to assess the current research and development of traditional Uighur medicine in Xinjiang (China), and to evaluate the promising pharmacological products of traditional Uighur medicine for further studies.
Materials and Methods: Traditional Uighur medicine data of medicine registry, patent, and academic publications was collected and analyzed.
Results: Data showed that, among the registered and studied traditional Uighur medicine, the main therapeutic areas of traditional Uighur medicine focused on skin disease, urogenital disease, rheumatism and digestive system disease. The representative traditional Uighur patent medicine included the following: BaixuanXiatare Tablets, Kaliziran Tincture and Vernoniaanthelmintica Injection (Psoriasis and vitiligo); Xi-payimazibiziLiquid (prostatitis); KursiKaknaq (urinary tract infection); Tongzhisurunjiang Capsules (anti-rheumatism medicine); HuganBuzure Granules (digestive system disease). Moreover, ten Uighur herbs were widely used, including: ResinaScammoniae, Folium FumicisDentati, HerbaDracocephali, Semen AmygdaliDulcis, HerbaChamomillae, FructusPimpinellaeanisi, Cortex Foeniculi, FructusVernoniae, FructusApii, and Radix AnacycliPyrethri.
Conclusion: This study concluded by indicating that traditional Uighur medicine with excellent curative effect should be screened in details for their phytochemical properties and pharmacological activity to discover new bioactive constituents
Application of Medical Statistical and Machine Learning Methods in the Age Estimation of Living Individuals
In the study of age estimation in living individuals, a lot of data needs to be analyzed by mathematical statistics, and reasonable medical statistical methods play an important role in data design and analysis. The selection of accurate and appropriate statistical methods is one of the key factors affecting the quality of research results. This paper reviews the principles and applicable principles of the commonly used medical statistical methods such as descriptive statistics, difference analysis, consistency test and multivariate statistical analysis, as well as machine learning methods such as shallow learning and deep learning in the age estimation research of living individuals, and summarizes the relevance and application prospects between medical statistical methods and machine learning methods. This paper aims to provide technical guidance for the age estimation research of living individuals to obtain more scientific and accurate results.</p
