    Phenolic content and radical scavenging properties of natural extracts for traditional care of domestic animals in Mediterranean areas

    Extracts of plants traditionally used for treatments on animals were tested for total phenolic, non tannic phenolic, condensed tannins and total flavonoid content. Antioxidant properties were determined using 2,2’-azinobis (3-ethylbenzothiazoline-6-sulphonic acid) diammonium salt (ABTS) and 1,1-diphenyl-2-picrylhydrazyl (DPPH) chemical assays following DMSO extraction. We also assessed the toxicity of extracts on cell viability and determined their ability to modulate the production of intracellular reactive oxygen species

    Advanced SBAS-DInSAR technique for controlling large civil infrastructures: an application to the Genzano di Lucania dam

    Monitoring surface deformation on dams is commonly carried out by in situ geodetic surveying, which is time consuming and characterized by some limitations in space coverage and frequency. More recently microwave satellite-based technologies, such as advanced-DInSAR (Differential Synthetic Aperture Radar Interferometry), have allowed the integration and improvement of the observation capabilities of ground-based methods thanks to their effectiveness in collecting displacement measurements on many non-destructive control points, corresponding to radar reflecting targets. The availability of such a large number of points of measurement, which are distributed along the whole structure and are characterized by millimetric accuracy on displacement rates, can be profitably adopted for the calibration of numerical models. These models are implemented to simulate the structural behaviour of a dam under conditions of stress thus improving the ability to maintain safety standards. In this work, after having analysed how advanced DInSAR can effectively enhance the results from traditional monitoring systems that provide comparable accuracy measurements on a limited number of points, an FEM model of the Genzano di Lucania earth dam is developed and calibrated. This work is concentrated on the advanced DInSAR technique referred to as Small BAseline Subset (SBAS) approach, benefiting from its capability to generate deformation time series at full spatial resolution and from multi-sensor SAR data, to measure the vertical consolidation displacement of the Genzano di Lucania earth dam

    DInSAR deformation time series for monitoring urban areas: The impact of the second generation SAR systems

    We investigate the capability improvement of the DInSAR techniques to map deformation phenomena affecting urban areas, by performing a comparative analysis of the deformation time series retrieved by applying the full resolution Small BAseline Subset (SBAS) DInSAR technique to selected sequences of SAR data acquired by the ENVISAT, RADARSAT-1 and COSMO-SkyMed (CSK) SAR data. The presented study, focused on the city of Napoli (Italy), allows us to quantify the dramatic increase of the DInSAR coherent pixel density achieved by exploiting the high resolution X-Band CSK SAR images with respect to the RADARSAT-1 and ENVISAT products, respectively; this permits us to analyze nearly all the structures located within the investigated urbanized area and, in many cases, also portions of a same building. © 2012 IEEE

    Testicular degeneration and infertility following arbovirus infection

    Arboviruses can cause a variety of clinical signs including febrile illness, arthritis, encephalitis and hemorrhagic fever. The recent Zika epidemic highlighted the possibility that arboviruses may also negatively affect the male reproductive tract. In this study, we focused on bluetongue virus (BTV), the causative agent of bluetongue and one of the major arboviruses of ruminants. We show that rams that recovered from bluetongue displayed signs of testicular degeneration and azoospermia up to 100 days after the initial infection. Importantly, testicular degeneration was induced in rams experimentally infected with either a high (BTV-1IT2006) or low (BTV-1IT2013) virulence strain of BTV. Rams infected with the low virulent BTV strain displayed testicular lesions in the absence of other major clinical signs. Testicular lesions in BTV-infected rams were due to viral replication in the endothelial cells of the peritubular areas of the testes, resulting in stimulation of a type-I IFN response, reduction of testosterone biosynthesis by Leydig cells, and destruction of Sertoli cells and the blood-testis barrier in more severe cases. Hence, BTV induces testicular degeneration and disruption of spermatogenesis by replicating solely in the endothelial cells of the peritubular areas unlike other gonadotropic viruses. This study shows that a naturally occurring arboviral disease can cause testicular degeneration and affect male fertility at least temporarily

    Treatment with embryonic stem-like cells into osteochondral defects in sheep femoral condyles

    Background Articular cartilage has poor intrinsic capacity for regeneration because of its avascularity and very slow cellular turnover. Defects deriving from trauma or joint disease tend to be repaired with fibrocartilage rather than hyaline cartilage. Consequent degenerative processes are related to the width and depth of the defect. Since mesenchymal stem cells (MSCs) deriving from patients affected by osteoarthritis have a lower proliferative and chondrogenic activity, the systemic or local delivery of heterologous cells may enhance regeneration or inhibit the progressive loss of joint tissue. Embryonic stem cells (ESCs) are very promising, since they can self-renew for prolonged periods without differentiation and can differentiate into tissues from all the 3 germ layers. To date only a few experiments have used ESCs for the study of the cartilage regeneration in animal models and most of them used laboratory animals. Sheep, due to their anatomical, physiological and immunological similarity to humans, represent a valid model for translational studies. This experiment aimed to evaluate if the local delivery of male sheep embryonic stem-like (ES-like) cells into osteochondral defects in the femoral condyles of adult sheep can enhance the regeneration of articular cartilage. Twenty-two ewes were divided into 5 groups (1, 2, 6, 12 and 24 months after surgery). Newly formed tissue was evaluated by macroscopic, histological, immunohistochemical (collagen type II) and fluorescent in situ hybridization (FISH) assays. Results Regenerated tissue was ultimately evaluated on 17 sheep. Samples engrafted with ES-like cells had significantly better histologic evidence of regeneration with respect to empty defects, used as controls, at all time periods. Conclusions Histological assessments demonstrated that the local delivery of ES-like cells into osteochondral defects in sheep femoral condyles enhances the regeneration of the articular hyaline cartilage, without signs of immune rejection or teratoma for 24 months after engraftment.</br

    A Canine Gait Analysis Protocol for Back Movement Assessment in German Shepherd Dogs

    Objective-To design and test a motion analysis protocol for the gait analysis of adult German Shepherd (GS) dogs with a focus in the analyses of their back movements. Animals-Eight clinically healthy adult large-sized GS dogs (age, 4 +/- 1.3 years; weight, 38.8 +/- 4.2 kg). Procedures-A six-camera stereo-photogrammetric system and two force platforms were used for data acquisition. Experimental acquisition sessions consisted of static and gait trials. During gait trials, each dog walked along a 6 m long walkway at self-selected speed and a total of six gait cycles were recorded. Results-Grand mean and standard deviation of ground reaction forces of fore and hind limbs are reported. Spatial-temporal parameters averaged over gait cycles and subjects, their mean, standard deviation and coefficient of variance are analyzed. Joint kinematics for the hip, stifle and tarsal joints and their average range of motion (ROM) values, and their 95% Confidence Interval (CI) values of kinematics curves are reported. Conclusions and Clinical Relevance-This study provides normative data of healthy GS dogs to form a preliminary basis in the analysis of the spatial-temporal parameters, kinematics and kinetics during quadrupedal stance posture and gait. Also, a new back movement protocol enabling a multi-segment back model is provided. Results show that the proposed gait analysis protocol may become a useful and objective tool for the evaluation of canine treatment with special focus on the back movement

    A GeoNode-based platform for an effective exploitation of advanced DInSAR measurements

    This work presents the development of an efficient tool for managing, visualizing, analysing, and integrating with other data sources, the deformation time-series obtained by applying the advanced differential interferometric synthetic aperture radar (DInSAR) techniques. To implement such a tool we extend the functionalities of GeoNode, which is a web-based platform providing an open source framework based on the Open Geospatial Consortium (OGC) standards, that allows development of Geospatial Information Systems (GIS) and Spatial Data Infrastructures (SDI). In particular, our efforts have been dedicated to enable the GeoNode platform to effectively analyze and visualize the spatio/temporal characteristics of the DInSAR deformation time-series and their related products. Moreover, the implemented multi-thread based new functionalities allow us to efficiently upload and update large data volumes of the available DInSAR results into a dedicated geodatabase. The examples we present, based on Sentinel-1 DInSAR results relevant to Italy, demonstrate the effectiveness of the extended version of the GeoNode platform

    Infections in lung cancer patients undergoing immunotherapy and targeted therapy: an overview on the current scenario

    Patients with a diagnosis of lung cancer are often vulnerable to infection, and the risk is increased by tumor-associated immunosuppression and the effects of the treatments. Historically, links between the risk of infection and cytotoxic chemotherapy due to neutropenia and respiratory syndromes are well established. The advent of tyrosine kinase inhibitors (TKIs) and immune-checkpoint inhibitors (ICIs) targeting the programmed cell death-1 (PD-1)/programmed cell death- ligand 1 (PD-L1) axis and cytotoxic T-lymphocyte antigen-4 (CTLA-4) have changed the treatment paradigm for lung cancer patients. Our understanding of the risk of infections while administrating these drugs is evolving, as are the biological mechanisms that are responsible. In this overview, we focus on the risk of infection with the use of targeted therapies and ICIs, summarizing current evidence from preclinical and clinical studies and discussing their clinical implications

    Cell-Free DNA Variant Sequencing Using CTC-Depleted Blood for Comprehensive Liquid Biopsy Testing in Metastatic Breast Cancer

    Keup and colleagues provide liquid biopsy preliminary results by sequencing variants in circulating tumor cells (CTCs) and cell-free deoxyribonucleic acid (cfDNA) "all from one tube" format, in order to use the same blood sample under the same isolation conditions of both analytes to reach an unbiased comparability and consistency. We appreciated the attempt of the authors to improve technical procedures in liquid biopsy research area, but we wanted to raise several issues related to cfDNA detection, reporting our research experience. This is a feasibility study as the authors analyzed only one sample from a small case series at an advanced line of treatment. In the clinical practice to monitor the disease and predict the treatment response, the analysis should be done at multiple time points. We have previously demonstrated that the quantity and the integrity of the cfDNA are not useful to determine the evolution of early breast cancer (bc), maybe due to the fact that cfDNA is not strictly related to cancer but also to an inflammatory status. Given that a high content of cfDNA could reflect inflammatory processes, we decided to investigate the role of stimulator of interferon gene (STING), an important regulator of cancer cell growth and senescence, in bc tissue in relation to cfDNA. STING biomarker analyzed by immunohistochemistry on tumor tissue could reflect a circulating inflammatory status and needs to be further investigated, not only on CTCs but also on cfDNA. One of the major issues of cfDNA is to decide what to analyze on it, in terms of type of cells and genetic alterations. Considering that multiple tests could be done to study gene copy number alterations, mutations, and variant fusions, the proper molecular test should be chosen, on the basis of the clinical need, starting from the treatment choice to disease monitoring
