On cost-effective reuse of components in the design of complex reconfigurable systems
Design strategies that benefit from the reuse of system components can reduce costs while maintaining or increasing dependability—we use the term dependability to tie together reliability and availability. D3H2 (aDaptive Dependable Design for systems with Homogeneous and Heterogeneous redundancies) is a methodology that supports the design of complex systems with a focus on reconfiguration and component reuse. D3H2 systematizes the identification of heterogeneous redundancies and optimizes the design of fault detection and reconfiguration mechanisms, by enabling the analysis of design alternatives with respect to dependability and cost. In this paper, we extend D3H2 for application to repairable systems. The method is extended with analysis capabilities allowing dependability assessment of complex reconfigurable systems. Analysed scenarios include time-dependencies between failure events and the corresponding reconfiguration actions. We demonstrate how D3H2 can support decisions about fault detection and reconfiguration that seek to improve dependability while reducing costs via application to a realistic railway case study
Strong contribution of diatom resting spores to deep-sea carbon transfer in naturally iron-fertilized waters downstream of South Georgia
Biogeochemical and diatom export fluxes are presented from two bathypelagic sediment trap deployments in the Antarctic Zone of the Southern Ocean. One of the sediment traps was deployed in very productive, naturally iron-fertilized waters downstream of South Georgia (P3, 2000 m) and compared to a deployment in moderately productive waters upstream of the island system (P2, 1500 m). At both sites significant diatom export events occurred in spring (November) and contained mostly empty cells that were associated with low particulate organic carbon (POC) fluxes. A summer export pulse occurred one month later at P2 (end February/March) compared to P3 (end January). Diatom fluxes at P3 were one order of magnitude higher than at P2, a difference mainly attributed to the short and intense export of resting spores from Chaetoceros Hyalochaete and Thalassiosira antarctica species. Aside from these resting spores, diatom export assemblages at both sites were dominated by empty Fragilariopsis kerguelensis frustules. The fraction of diatoms exported as empty frustules was considerably lower at P3 (52%) than P2 (91%). This difference was related to the flux of intact diatom resting spores at P3 and may partially explain the lower Si:C export stoichiometry observed at P3 (1.1) compared to P2 (1.5). Through the enumeration of full diatom frustules and subsequent biomass calculations we estimate that diatom resting spores account for 42% of annual POC flux in the productive waters downstream of South Georgia. At both sites the contribution of diatom vegetative stages to POC fluxes was considerably lower (<5%). From these analyses we conclude that resting spore export contributes towards the slightly higher bathypelagic (POC) flux at P3 (40.6 mmol m−2 y−1) compared to P2 (26.4 mmol m−2 y−1). We compared our sediment trap records with previously published diatom assemblage data from the mixed layer and surface sediments (3760 m) around South Georgia. The relative proportion of diatom resting spores within diatom assemblages increases as a function of depth and is explained by selective preservation of their robust frustules. Our study highlights the significance of diatom resting spore export as a carbon vector out of the mixed layer. Furthermore, the contribution or resting spores to POC flux in the bathypelagic ocean and sediments suggests they play a particularly important role in sequestering biologically fixed CO2 over climatically relevant timescales
Nanoplastics affect moulting and faecal pellet sinking in Antarctic krill (Euphausia superba) juveniles
Plastic debris has been identified as a potential threat to Antarctic marine ecosystems, however, the impact of nanoplastics (<1 μm) is currently unexplored. Antarctic krill (Euphausia superba) is a keystone species of Southern Ocean pelagic ecosystems, which plays a central role in the Antarctic food webs and carbon (C) cycle. Krill has been shown to rapidly fragment microplastic beads through the digestive system, releasing nanoplastics with unknown toxicological effects. Here we exposed krill juveniles to carboxylic (COOH, anionic) and amino- (NH2, cationic) polystyrene nanoparticles (PS NPs) and we investigated lethal and sub-lethal endpoints after 48 h. The analysis of PS NP suspensions in Antarctic sea water (SW) media showed that PS-COOH formed large agglomerates (1043 ± 121 nm), while PS-NH2 kept their nominal size (56.8 ± 3 nm) during the exposure time. After 48 h, no mortality was found but increase in exuviae production (12.6 ± 1.3%) and reduced swimming activity were observed in juveniles exposed to PS-NH2. The microbial community composition in SW supports the release of krill moults upon PS NP exposure and stimulates further research on the pivotal role of krill in shaping Southern Ocean bacterial assemblages. The presence of fluorescent signal in krill faecal pellets (FPs) confirmed the waterborne ingestion and egestion of PS-COOH at 48 h of exposure. Changes in FP structure and properties were also associated to the incorporation of PS NPs regardless of their surface charge. The effects of PS NPs on krill FP properties were compared to Control 0 h as a reference for full FPs (plastic vs food) and Control 48 h as a reference for more empty-like FPs (plastic vs lack of food). Exposure to PS NPs led to a FP sinking rate comparable to Control 48 h, but significantly lower than Control 0 h (58.40 ± 23.60 m/d and 51.23 ± 28.60 m/d for PS-COOH and PS-NH2; 168.80 ± 74.58 m/d for Control 0 h). Considering the important role played by krill in the food web and C export in the Southern Ocean, the present study provides cues about the potential impact of nanoplastics on Antarctic pelagic ecosystems and their biogeochemical cycles

Ganglion-specific splicing of TRPV1 underlies infrared sensation in vampire bats.
Vampire bats (Desmodus rotundus) are obligate blood feeders that have evolved specialized systems to suit their sanguinary lifestyle. Chief among such adaptations is the ability to detect infrared radiation as a means of locating hotspots on warm-blooded prey. Among vertebrates, only vampire bats, boas, pythons and pit vipers are capable of detecting infrared radiation. In each case, infrared signals are detected by trigeminal nerve fibres that innervate specialized pit organs on the animal's face. Thus, vampire bats and snakes have taken thermosensation to the extreme by developing specialized systems for detecting infrared radiation. As such, these creatures provide a window into the molecular and genetic mechanisms underlying evolutionary tuning of thermoreceptors in a species-specific or cell-type-specific manner. Previously, we have shown that snakes co-opt a non-heat-sensitive channel, vertebrate TRPA1 (transient receptor potential cation channel A1), to produce an infrared detector. Here we show that vampire bats tune a channel that is already heat-sensitive, TRPV1, by lowering its thermal activation threshold to about 30 °C. This is achieved through alternative splicing of TRPV1 transcripts to produce a channel with a truncated carboxy-terminal cytoplasmic domain. These splicing events occur exclusively in trigeminal ganglia, and not in dorsal root ganglia, thereby maintaining a role for TRPV1 as a detector of noxious heat in somatic afferents. This reflects a unique organization of the bat Trpv1 gene that we show to be characteristic of Laurasiatheria mammals (cows, dogs and moles), supporting a close phylogenetic relationship with bats. These findings reveal a novel molecular mechanism for physiological tuning of thermosensory nerve fibres
The role of diatom resting spores in pelagic–benthic coupling in the Southern Ocean
Natural iron fertilization downstream of Southern Ocean island plateaus supports large phytoplankton blooms and promotes carbon export from the mixed layer. In addition to sequestering atmospheric CO2, the biological carbon pump also supplies organic matter (OM) to deep-ocean ecosystems. Although the total flux of OM arriving at the seafloor sets the energy input to the system, the chemical nature of OM is also of significance. However, a quantitative framework linking ecological flux vectors to OM composition is currently lacking. In the present study we report the lipid composition of export fluxes collected by five moored sediment traps deployed in contrasting productivity regimes of Southern Ocean island systems (Kerguelen, Crozet and South Georgia) and compile them with quantitative data on diatom and faecal pellet fluxes. At the three naturally iron-fertilized sites, the relative contribution of labile lipids (mono- and polyunsaturated fatty acids, unsaturated fatty alcohols) is 2–4 times higher than at low productivity sites. There is a strong attenuation of labile components as a function of depth, irrespective of productivity. The three island systems also display regional characteristics in lipid export. An enrichment of zooplankton dietary sterols, such as C27Δ5, at South Georgia is consistent with high zooplankton and krill biomass in the region and the importance of faecal pellets to particulate organic carbon (POC) flux. There is a strong association of diatom resting spore fluxes that dominate productive flux regimes with energy-rich unsaturated fatty acids. At the Kerguelen Plateau we provide a statistical framework to link seasonal variation in ecological flux vectors and lipid composition over a complete annual cycle. Our analyses demonstrate that ecological processes in the upper ocean, e.g. resting spore formation and grazing, not only impact the magnitude and stoichiometry of the Southern Ocean biological pump, but also regulate the composition of exported OM and the nature of pelagic–benthic coupling
Light Concentrators for Borexino and CTF
Light concentrators for the solar neutrino experiment Borexino and the
Counting Test Facility (CTF) have been developed and constructed. They increase
the light yield of these detectors by a factor of 2.5 and 8.8, respectively.
Technical challenges like long term stability in various media, high
reflectivity and radiopurity have been addressed and the concepts to overcome
these difficulties will be described. Gamma spectroscopy measurements of the
concentrators show an upper limit of 12e-6 Bq/g for uranium and a value of
120e-6 Bq/g for thorium. Upper limits on other possible contaminations like
26Al are presented. The impact of these results on the performance of Borexino
and the CTF are discussed and it is shown that the design goals of both
experiments are fulfilled.Comment: submitted to Nuclear Instruments and Methods in Physics Researc
The health impact of hazardous waste landfills and illegal dumps contaminated sites: An epidemiological study at ecological level in Italian Region
Background and aim: The implementation of idoneous management of hazardous waste, in contrast to illegal practices, is one of the environment and health priorities of the WHO. The aim of the present study, based on a collaborative agreement between the Italian National Health Institute and a Prosecution Office located in Naples North, was to evaluate the health effects of illegal landfills and burning of urban and hazardous waste in the territory of the Prosecution Office. Methods: The municipalities included in the study territory were investigated with respect to the regional population. Regression analyses were performed in the study area between four classes of an environmental municipal indicator of waste risk (MRI) previously defined, computing the relative risks (RRs) in 2–4 MRI classes, with respect to the first MRI class (the least impacted). The prevalence of reproductive outcomes and cause-specific mortality and hospitalization were analyzed in the general population and in the 0–19-year-old population using SAS software. Results: An increase of mortality and hospitalization risk in both the genders of the whole area, with respect to regional population, were found for overall all cancer cases, cancer of the stomach, the liver, the lung and the kidney, and ischemic heart diseases. An increase of mortality for leukemias in the 0-19-year-old population and in hospitalization risk for certain conditions originating in the perinatal period were observed. Correlation between MRI and the risk of mortality from breast tumors in women (MRI class 2: RR = 1.06; MRI class 3: RR = 1.15; MRI class 4: RR = 1.11) and between MRI and the risk of hospitalization from testis tumors (MRI class 2: RR = 1.25; MRI class 3: RR = 1.31; MRI class 4: RR = 1.32) were found. The hospitalization risk from breast tumors and asthma exceeded significantly in both genders of three and four MRI classes. Among the 0-19-year-old population, correlation between MRI and hospitalization from leukemias (MRI class 2: RR = 1.48; MRI class 3: RR = 1.60; MRI class 4: RR = 1.41) and between MRI and the prevalence of preterm birth (MRI class 2: RR = 1.17; MRI class 3: RR = 1.08; MRI class 4: RR = 1.25) were found. Conclusion: A correlation between health outcomes and the environmental pressure by uncontrolled waste sites was found. Notwithstanding the limitation of the study, the results promote implementing the actions of environmental remediation and the prosecution of illegal practices
Hormonal therapy with megestrol in inoperable hepatocellular carcinoma characterized by variant oestrogen receptors
Variant liver oestrogen receptor transcripts in hepatocellular carcinoma are associated with aggressive clinical course and unresponsiveness to tamoxifen. To evaluate the impact on survival and on tumour growth of megestrol (progestin drug acting at post-receptorial level) we enrolled 45 patients with HCC characterized by variant liver oestrogen receptors in a prospective, randomized study with megestrol vs. placebo. Presence of variant oestrogen receptors was determined by RT/PCR. 24 patients were randomized to no treatment and 21 to therapy with megestrol 160 mg day−1. Results were analysed by Kaplan-Meier and Cox methods. Survival of hepatocellular carcinoma characterized by variant oestrogen receptors was extremely poor (median survival 7 months); megestrol significantly improved survival (18 months) (P = 0.0090). Tumour growth at one year was significantly slowed down in megestrol-treated patients (P = 0.0212). Bilirubin levels, presence of portal thrombosis, HBV aetiology and treatment were identified at univariate analysis as factors significantly associated with survival; at multivariate analysis, only megestrol therapy (P = 0.0003), presence of HBV infection (P = 0.0009) and presence of portal vein thrombosis (P = 0.0051) were factors independently related with survival. (1) Megestrol slows down the aggressive tumour growth of patients with hepatocellular carcinoma characterized by variant estrogen receptors and (2) is also able to favourably influence the course of disease, more than doubling median survival. © 2001 Cancer Research Campaignhttp://www.bjcancer.co
Minimal Standard Heterotic String Models
Three generation heterotic-string vacua in the free fermionic formulation
gave rise to models with solely the MSSM states in the observable Standard
Model charged sector. The relation of these models to Z_2 x Z_2 orbifold
compactifications dictates that they produce three pairs of untwisted Higgs
multiplets. The reduction to one pair relies on the analysis of supersymmetric
flat directions, that give superheavy mass to the dispensable Higgs states. We
explore the removal of the extra Higgs representations by using the free
fermion boundary conditions and hence directly at the string level, rather than
in the effective low energy field theory. We present a general mechanism that
achieves this reduction by using asymmetric boundary conditions between the
left- and right-moving internal fermions. We incorporate this mechanism in
explicit string models containing three twisted generations and a single
untwisted Higgs doublet pair. We further demonstrate that an additional effect
of the asymmetric boundary conditions is to substantially reduce the
supersymmetric moduli space.Comment: 20 pages, LaTeX; added reference
