    Medicinal plants as a source of future anti-pruritic drugs: A comprehensive review

    Pruritus is a distressing sensation of the skin that provokes the desire to scratch. Medicinal plants have been proposed as a worthful source for identifying new bioactive molecules. The aim of this study was to evaluate some medicinal plants and their phytochemicals used in the management of pruritus. Medicinal plants including Avena sativa, Borago officinalis, Capsicum frutescens, Curcuma longa, Fumaria spp., Mentha x piperita and Oenothera biennis showed the promising anti-pruritic activity in human studies. In experimental studies, Angelica sinensis, Betula platyphylla, Matricaria chamomilla, Rumex Japonicus, Saururus chinensis and Vaccinium myrtillus are among the best medicinal plants for management of pruritus. Essential oils, alkaloids, saponins, sterols, terpens, phenolic compounds, and fatty acids were the bioactive constituents of herbs which exhibited their anti-pruritic activity through different mechanisms. The most predominant mechanisms involved in activity of plant-derived molecules in pruritis include reducing serum IgE and proinflammatory cytokines, stabilizing mast cells, suppressing the Th2 cellular response, suppressing the expression of substance P and NF- κB, inhibiting prostaglandin E2 production, and activating receptors involved in itch sensation.     Determination of Synthetic Food Colors, Caffeine, Sodium Benzoate and Potassium Sorbate in Sports Drinks

    Purpose: To employ high performance liquid chromatography (HPLC) techniques to determine edible additives in commercial sports drinks.Methods: A total of 105 samples including 21 different brands of sports drinks were purchased from markets in Tehran, Iran. The process conditions included a C18 column, a mobile phase consisting of aqueous ammonium acetate buffer (pH = 4.2) and acetonitrile (80:20 v/v) at a flow rate of 0.8 ml min–1. The injection volume was 20 μL and the UV detector was set at 225 nm.Results: Mean recovery was between 95 and 106 %. The mean concentrations of sodium benzoate and potassium sorbate in Iranian brands (147.72 and 11.54 ppm, respectively) were significantly higher than in foreign brands (19.43 and 4.91 ppm, respectively, p < 0.05). The maximum amount of sodium benzoate and potassium sorbate was 251.50 ± 18.69 and 96.38 ± 38.56 ppm, respectively, while caffeine content was in the range of 293.48 ± 14.33 - 607.32 ± 135.33 ppm.Conclusion: HPLC permits the detection of sodium benzoate and potassium sorbate at very low concentrations. Furthermore, caffeine was detected in all the sport drinks.     The Contents of Sesamol in Iranian Sesame Seeds

    Abstract Sesamol is a sesame lignan. Sesame lignans have multiple functions, including antioxidant activity and also seem to have potential as a source of phytostrogens. This study was condncted to evaluate sesamol contents of 7 brands of Iranian sesame seeds (Sesamum indicume L.). The brands were named Karaj 29, Darab 14, Ultan, Dezful, Varamin, Branching Naz, and Nonbranching Naz. After method validation, the methanolic extracts of seeds were investigated by HPLC. Their mean of total sesamol was found to be 4.67±0.92 mg/g (ranging between 2.75 and 6.13 mg/g). The brands Karaj 29 with 5.84±0.25 mg/g, Dezful with 5.48±0.08 mg/g, and Varamin with 5.4±0.1mg/g had the highest content, and Darab 14 with the content of 3.30±0.57mg/g had the lowest (p<0.05). Iranian sesame can be considered to be a good source of natural antioxidants for medicinal and commercial uses

    Determination of 8 Synthetic Food Dyes by Solid Phase Extraction and Reversed-Phase High Performance Liquid Chromatography

    Purpose: To develop a simple, selective, and sensitive extraction and assay method for the analysis of eight food dyes.Methods: All sulphonated colors were extracted by an NH2-aminopropyl-modified silica SPE cartridge, and thereafter determined by reverse phase high performance liquid chromatography (RP-HPLC) using a C18 column with gradient elution of ammonium acetate buffer (pH 6.7), methanol, and acetonitrile. The analysis was carried out on a UV detector with two optimized method settings within 17 min.Results: Certificated standard material (CRM) was used to validate the method, and significant difference was not observed between the results and assigned values. Maximum LOD was 1.154 ppm at 250 nm for Brilliant blue, and 0.873 ppm for carmoisine. Recovery was 94.2 % for Brilliant blue in jelly powder.Conclusion: This method was successfully applied to determine colorants in various (30) water-soluble foods, including fruit flavored drinks, sugar confectionery, sweets, etc.Keywords: Synthetic colors, Food, Fruit flavored drinks, Solid phase extraction, RP-HPL

    Standardization of Tragopogon graminifolius DC. Extract Based on Phenolic Compounds and Antioxidant Activity

    Tragopogon graminifolius DC. (TG), Compositae family, is traditionally used for the treatment of various diseases like gastrointestinal and hepatic disorders. The aim of the present study is to standardize extracts from TG used for preparation of different dosage forms in traditional Iranian medicine (TIM) based on phenolic compounds. For this purpose, total phenolic content and some phenolic compounds were determined in ethanolic extracts from aerial part and root of TG by HPLC method. Furthermore, antioxidant activity was evaluated using DPPH-HPLC methods. Caffeic acid, gallic acid, -coumaric acid, ferulic acid, and catechin were detected in root and aerial part of TG. -Coumaric acid (6.357 ± 0.014 mg⋅g −1 ) was dominant phenolic compound in aerial part followed by ferulic acid (1.24 ± 0.018 mg⋅g −1 ). Also, -coumaric acid (2.685 ± 0.031 mg⋅g −1 ) was highly abundant in root, followed by catechin (2.067 ± 0.021 mg⋅g −1 ). Antioxidant activity of root extract (460.45 ± 0.78 g Vit.E.E⋅mL −1 ) was better than that of aerial part. Generally, phenolic compounds are one of the major constituents of TG and could be used as markers for standardization of dosage forms prepared from this plant. Also, TG demonstrated significant antioxidant activity using DPPH-HPLC method. Phenolic compounds of TG may be responsible for its marked antioxidant properties

    Simultaneous Determination of Preservatives in Dairy Products by HPLC and Chemometric Analysis

    Cheese and yogurt are two kinds of nutritious dairy products that are used worldwide. The major preservatives in dairy products are sodium benzoate, potassium sorbate, and natamycin. The maximum permitted levels for these additives in cheese and yogurt are established according to Iranian national standards. In this study, we developed a method to detect these preservatives in dairy products by reversed phase chromatography with UV detection in 220 nm, simultaneously. This method was performed on C18 column with ammonium acetate buffer (pH=5) and acetonitrile (73 : 27 v/v) as mobile phase. The method was carried out on 195 samples in 5 kinds of commercial cheeses and yogurts. The results demonstrated insufficient separation where limit of detection (LOD) and limit of quantitation (LOQ) ranged from 0.326 to 0.520 mg/kg and 0.989 to 1.575 mg/kg in benzoate and sorbate, respectively. The correlation coefficient of each calibration curve was mostly higher than 0.997. All samples contained sodium benzoate in various ranges. Natamycin and sorbate were detected in a remarkable amount of samples, while, according to Iranian national standard, only sorbate is permitted to be added in processed cheeses as a preservative.     Antioxidant Capacity of Plasma after Pomegranate Intake in Human Volunteers

    Dietary antioxidants including phenolic compounds are believed to be effective nutrients in the prevention of oxidative stress related disease. Pomegranate has been used for centuries in ancient cultures for its medicinal purpose and is widely acknowledged for antioxidant properties. The present study was designed to assess the effect of pomegranate fresh fruit consumption on the plasma antioxidant capacity. Thirty healthy volunteers were recruited for the study. Volunteers were randomly divided into three groups (pomegranate, vitamin E and water consumption). Blood samples were collected, after at least 12 hours overnight fast, the day before beginning supplementation period and the day after supplementation had finished. Total antioxidant capacity measurement by FRAP method and clinical laboratory test were performed for all volunteers in two selected times. The obtained data revealed that consumption of 100 grams pomegranate and vitamin E per day for ten days resulted in a significant rise (14.05%, 8.28%) plasma antioxidant capacity respectively, but this difference was not significant for water group

    Evaluation of Antioxidants in Bone Mineral Density of Iranian

    AbstractObjective(s) Bone is a dynamic tissue that is continuously renewed throughout life by the process of bone remodeling. Antioxidant system might be involved in the pathogenesis of bone loss, so the aim of this study was to evaluate the total antioxidant capacity (TAC), vitamin C and vitamin E levels of plasma besides measuring enzymatic antioxidants, superoxide dismutase (SOD), catalase (CAT) and glutathione reductase (GR) enzymes activity in Iranian osteoporotic women comparing to the control group.Materials and MethodsBone mineral density (BMD) of the femoral neck and lumbar spine was measured by dual x-ray absorptiometry. The participants were divided into groups: a) total participants (-3.9 ≤ T–score ≤ 3.6) including 192 women, b) the control group (T-score ≥ -1) including 76 women, c) the total patients (T-score < -1) including 76 women. Then, plasma TAC, vitamin C levels, SOD and GR activities, erythrocyte CAT were measured using spectrophotometrical methods separately, and for vitamin E by HPLC analysis.ResultsComparing the control group and osteoporotic women showed that: a) plasma levels for vitamin C and erythrocyte CAT were markedly lower in the patients than in the controls, but plasma activity of TAC, SOD and GR were significantly higher, respectively. b) the differences were higher between control and patients with severe disease (T-score <-1.7) comparing to patients in the group with milder disease (-1.7 ≤ T-score <-1). c) Femoral neck BMD adjusted with age and BMI showed a positive and significant correlation with plasma levels of vitamin C in all subjects, but this relation was reverse or negative for TAC, SOD and GR.ConclusionIt seems that a physiologic increase in the amount of some antioxidants occurs in osteoporosis; even though this amount may not be sufficient for the human body requirements
