Rare coding variants in PLCG2, ABI3, and TREM2 implicate microglial-mediated innate immunity in Alzheimer's disease
We identified rare coding variants associated with Alzheimer’s disease (AD) in a 3-stage case-control study of 85,133 subjects. In stage 1, 34,174 samples were genotyped using a whole-exome microarray. In stage 2, we tested associated variants (P<1×10-4) in 35,962 independent samples using de novo genotyping and imputed genotypes. In stage 3, an additional 14,997 samples were used to test the most significant stage 2 associations (P<5×10-8) using imputed genotypes. We observed 3 novel genome-wide significant (GWS) AD associated non-synonymous variants; a protective variant in PLCG2 (rs72824905/p.P522R, P=5.38×10-10, OR=0.68, MAFcases=0.0059, MAFcontrols=0.0093), a risk variant in ABI3 (rs616338/p.S209F, P=4.56×10-10, OR=1.43, MAFcases=0.011, MAFcontrols=0.008), and a novel GWS variant in TREM2 (rs143332484/p.R62H, P=1.55×10-14, OR=1.67, MAFcases=0.0143, MAFcontrols=0.0089), a known AD susceptibility gene. These protein-coding changes are in genes highly expressed in microglia and highlight an immune-related protein-protein interaction network enriched for previously identified AD risk genes. These genetic findings provide additional evidence that the microglia-mediated innate immune response contributes directly to AD development
A Characterization System for the Monitoring of ELI-NP Gamma Beam
The ELI-NP (Extreme Light Infrastructure-Nuclear Physics) facility, currently under
construction near Bucharest (Romania), is the pillar of the project ELI dedicated to the generation of
high-brilliance gamma beams and high-power laser pulses that will be used for frontier research in
nuclear physics. To develop an experimental program at the frontiers of the present-day knowledge,
two pieces of equipment will be deployed at ELI-NP: a high power laser system consisting of two
10 PW lasers and a high brilliance gamma beam system. The ELI-NP Gamma beam system will
deliver an intense gamma beam with unprecedented specifications in terms of photon flux, brilliance
and energy bandwidth in an energy range from 0.2 to 20 MeV. Such a gamma beam requires special
devices and techniques to measure and monitor the beam parameters during the commissioning and
the operational phase. To accomplish this task, the Gamma Beam Characterization System, equipped
with four elements, was developed: a Compton spectrometer (CSPEC), to measure and monitor the
photon energy spectrum; a nuclear resonant scattering system (NRSS), for absolute beam energy
calibration and inter-calibration of the other detectors; a beam profile imager (GPI) to be used for
alignment and diagnostics purposes; and finally a sampling calorimeter (GCAL), for a fast combined
measurement of the beam average energy and intensity. The combination of the measurements
performed by GCAL and CSPEC allows fully characterizing the gamma beam energy distribution
and intensity with a precision at the level of few per mill, enough to demonstrate the fulfillment
of the required parameters. This article presents an overview of the gamma beam characterization
system with focus on these two detectors, which were designed, assembled and are currently under
test at INFN-Firenze. The layout and the working principle of the four devices is described, as well
as some of the main results of detector test

Comportamento de risco dos conscritos do Exército Brasileiro, 1998: uma apreciação da infecção pelo HIV segundo diferenciais sócio-econômicos
Uma pesquisa de comportamento de risco ao HIV foi conduzida entre conscritos (17-20 anos) do Exército em 1998. A amostra (30.318) foi selecionada em três estratos: 1) municípios das regiões Norte e Centro-Oeste (N/CO); 2) municípios da região Sul (S); 3) municípios dos estados do Rio de Janeiro e São Paulo (RJ/SP), estimando-se a prevalência de infecção pelo HIV em cada estrato. O objetivo deste trabalho foi o de apreciar os resultados por nível sócio-econômico (NSE). A análise estatística utilizou um índice de comportamento sexual de risco para HIV e modelos de regressão logística. O estrato N/CO apresentou os piores indicadores - os relativos ao NSE, ao comportamento sexual de risco ou aos problemas relacionados às infecções sexualmente transmissíveis (IST) - assim como a maior proporção de infectados por HIV. Os melhores indicadores foram encontrados no estrato RJ/SP. O Sul mostrou resultados intermediários. O grau de escolaridade teve papel relevante. Os conscritos com primeiro grau incompleto apresentaram o pior índice de comportamento sexual de risco, o qual se mostrou importante preditor dos problemas relacionados às IST, incluindo a infecção por HIV

A sample of 30,318 subjects was selected in three strata, pertaining to counties from: 1) the North and Central-West (N/CW); 2) South (S); and the states of Rio de Janeiro and São Paulo. HIV prevalence rates were estimated in all strata. The objective of this paper was to analyze the results according to differences in socioeconomic status (SES). The statistical analysis used an index of sexual risk behavior and logistic regression models. The N/CW stratum showed the worst indicators for SES, sexual risk behavior, and sexually transmitted infections (STIs), as well as the highest HIV seroprevalence rate. The best indicators for all variables were found in the RJ/SP stratum. The South showed intermediate results. Level of schooling also played a relevant role. In all three strata the conscripts with an incomplete high school education displayed the worst sexual risk behavior index, shown to be a relevant predictor of STI-related problems, including HIV infection

Tracker-in-Calorimeter (TIC) Project: A Calorimetric New Solution for Space Experiments
A space-based detector dedicated to measurements of γ-rays and charged particles has to achieve a balance between different instrumental requirements. A good angular resolution is necessary for the γ-rays, whereas an excellent geometric factor is needed for the charged particles. The tracking reference technique of γ-ray physics is based on a pair-conversion telescope made of passive material (e.g., tungsten) coupled with sensitive layers (e.g., silicon microstrip). However, this kind of detector has a limited acceptance because of the large lever arm between the active layers, needed to improve the track reconstruction capability. Moreover, the passive material can induce fragmentation of nuclei, thus worsening charge reconstruction performances. The Tracker-In-Calorimeter (TIC) project aims to solve all these drawbacks. In the TIC proposal, the silicon sensors are moved inside a highly-segmented isotropic calorimeter with a couple of external scintillators dedicated to charge reconstruction. In principle, this configuration has a good geometrical factor, and the angle of the γ-rays can be precisely reconstructed from the lateral profile of the electromagnetic shower sampled, at different depths in the calorimeter, by silicon strips. The effectiveness of this approach has been studied with Monte Carlo simulations and validated with beam test data of a small prototype
