    Scaling Manifold Ranking Based Image Retrieval

    Manifold Ranking is a graph-based ranking algorithm being successfully applied to retrieve images from multimedia databases. Given a query image, Manifold Ranking computes the ranking scores of images in the database by exploiting the relationships among them expressed in the form of a graph. Since Manifold Ranking effectively utilizes the global structure of the graph, it is significantly better at finding intuitive results compared with current approaches. Fundamentally, Manifold Ranking requires an inverse matrix to compute ranking scores and so needs O(n^3) time, where n is the number of images. Manifold Ranking, unfortunately, does not scale to support databases with large numbers of images. Our solution, Mogul, is based on two ideas: (1) It efficiently computes ranking scores by sparse matrices, and (2) It skips unnecessary score computations by estimating upper bounding scores. These two ideas reduce the time complexity of Mogul to O(n) from O(n^3) of the inverse matrix approach. Experiments show that Mogul is much faster and gives significantly better retrieval quality than a state-of-the-art approximation approach

    Phosphatidylethanolamine dynamics are required for osteoclast fusion

    Osteoclasts, responsible for bone resorption, are multinucleated cells formed by cell-cell fusion of mononuclear pre-osteoclasts. Although osteoclast fusion is a pivotal step for osteoclastogenesis, little is known about the mechanism involved. To clarify the underlying process, we investigated dynamics of membrane phospholipids during osteoclastogenesis in vitro. We found that the cellular content of phospholipids, phosphatidylethanolamine (PE) in particular, was increased during osteoclast differentiation. Furthermore, PE was greatly increased in the outer leaflet of the plasma membrane bilayer during osteoclastogenesis, being concentrated in filopodia involved in cell-cell fusion. Immobilisation of the cell surface PE blocked osteoclast fusion, revealing the importance of PE abundance and distribution. To identify the molecules responsible for these PE dynamics, we screened a wide array of lipid-related genes by quantitative PCR and shRNA-mediated knockdown. Among them, a PE-biosynthetic enzyme, acyl-CoA:lysophosphatidylethanolamine acyltransferase 2 (LPEAT2), and two ATP-binding cassette (ABC) transporters, ABCB4 and ABCG1, were markedly increased during osteoclastogenesis, and their knockdown in pre-osteoclasts led to reduction in PE exposure on the cell surface and subsequent osteoclast fusion. These findings demonstrate that the PE dynamics play an essential role in osteoclast fusion, in which LPEAT2, ABCB4 and ABCG1 are key players for PE biosynthesis and redistribution

    Hemodynamic Performance of the Biventricular Bypass System Operated in an Independent Variable Rate Mode

    The present study was undertaken to determine whether a biventricular bypass system operated in an independent variable rate (VR) mode can maintain the entire circulation. Two pusher-plate pumps which incorporated the Hall effect position sensors were used to bypass the right and left ventricles in 10 sheep under fibrillation. The flow distributions of the pump output to the carotid and renal arteries were investigated every 6 h using ultrasonic blood flow meters for 24 h in 5 animals, and the controllability of the VR mode was evaluated in 5 long-term experiments. The carotid artery flow ratio to the pump output decreased significantly from 4.7 +/- 0.8% before the bypass to 2.7 +/- 0.9% after 24 h. However, the renal artery flow ratio did not change throughout the experiments. In the long-term experiments, the animals were kept alive from 3 to 48 days (mean 15.6 days). The mean pump output had been maintained at more than 90 ml/min/kg for the first 7 days. After the surgery, the pump driving conditions were not readjusted in any experiment. The results indicate that the biventricular bypass system operated in the independent VR mode automatically maintains the entire circulation at a satisfactory level.</p

    Reactive Hyperemia in Coronary Blood Flow 1

    The reactive hyperemia induced by a temporary occlusion of coronary circulation was investigated in an isolated cat\u27s heart perfused with blood supplied by a donor. The magnitude of hyperemia was quantitatively proportional to the ischemic periods ranging from 6 to 90 seconds. The pretreatment with atropine as well as dinitrophenol did not affect the reactive hyperemia. Diphenhydramine HCl in a dose reducing the effect of 1.0,μg of histamine decreased the magnitude of hyperemia. After pretreatment with cocaine and procaine the reactive hyperemia was inhibited . Serum potassium level of the coronary outflow collected after the occlusion was significantly increased. These findings suggest that the metabolites involved in the reactive hyperemia may not be acetylcholine, but histamine and potassium which probably produce the dilatation of coronary vessels through an axon reflex

    Enhancement of protein production via the strong DIT1 terminator and two RNA-binding proteins in Saccharomyces cerevisiae

    Post-transcriptional upregulation is an effective way to increase the expression of transgenes and thus maximize the yields of target chemicals from metabolically engineered organisms. Refractory elements in the 3′ untranslated region (UTR) that increase mRNA half-life might be available. In Saccharomyces cerevisiae, several terminator regions have shown activity in increasing the production of proteins by upstream coding genes; among these terminators the DIT1 terminator has the highest activity. Here, we found in Saccharomyces cerevisiae that two resident trans-acting RNA-binding proteins (Nab6p and Pap1p) enhance the activity of the DIT1 terminator through the cis element GUUCG/U within the 3′-UTR. These two RNA-binding proteins could upregulate a battery of cell-wall–related genes. Mutagenesis of the DIT1 terminator improved its activity by a maximum of 500% of that of the standard PGK1 terminator. Further understanding and improvement of this system will facilitate inexpensive and stable production of complicated organism-derived drugs worldwide

    Spontaneous rupture of metastatic α-fetoprotein-producing gastric cancer of the liver

    An 80-year-old man was admitted to our hospital because of the rupture of the liver. Laboratory data showed iron-deficiency anemia, although there was no liver dysfunction. A computed tomography scan showed large liver tumor with intraperitoneal hemorrhage, and since a serum level of α-fetoprotein (AFP) was extremely high, we initially suspected a rupture of hepatocellular carcinoma (HCC). Transarterial embolization was performed to stop bleeding from the tumor, followed by an endoscopic examination that revealed advanced gastric cancer. Histological analysis revealed that both the gastric and the hepatic tumors were moderately to poorly differentiated adenocarcinoma, as well as that both tumors were immunohistochemically positive for AFP. Finally, we diagnosed AFP-producing gastric cancer associated with liver metastasis. Rupture of metastatic liver cancer is rare, and accordingly, distinction from HCC is important, particularly for the cases of AFP-producing gastric cancer
