Gravitational-wave research as an emerging field in the Max Planck Society. The long roots of GEO600 and of the Albert Einstein Institute
On the occasion of the 50th anniversary since the beginning of the search for
gravitational waves at the Max Planck Society, and in coincidence with the 25th
anniversary of the foundation of the Albert Einstein Institute, we explore the
interplay between the renaissance of general relativity and the advent of
relativistic astrophysics following the German early involvement in
gravitational-wave research, to the point when gravitational-wave detection
became established by the appearance of full-scale detectors and international
collaborations. On the background of the spectacular astrophysical discoveries
of the 1960s and the growing role of relativistic astrophysics, Ludwig Biermann
and his collaborators at the Max Planck Institute for Astrophysics in Munich
became deeply involved in research related to such new horizons. At the end of
the 1960s, Joseph Weber's announcements claiming detection of gravitational
waves sparked the decisive entry of this group into the field, in parallel with
the appointment of the renowned relativist Juergen Ehlers. The Munich area
group of Max Planck institutes provided the fertile ground for acquiring a
leading position in the 1970s, facilitating the experimental transition from
resonant bars towards laser interferometry and its innovation at increasingly
large scales, eventually moving to a dedicated site in Hannover in the early
1990s. The Hannover group emphasized perfecting experimental systems at pilot
scales, and never developed a full-sized detector, rather joining the LIGO
Scientific Collaboration at the end of the century. In parallel, the Max Planck
Institute for Gravitational Physics (Albert Einstein Institute) had been
founded in Potsdam, and both sites, in Hannover and Potsdam, became a unified
entity in the early 2000s and were central contributors to the first detection

High-level expression of Lactobacillus beta-galactosidases in Lactococcus lactis using the food-grade, nisin-controlled expression system NICE
In this work the overlapping genes (lacL and lacM) encoding heterodimeric beta-galactosidases from Lactobacillus reuteri, Lb. acidophilus, Lb. sakei, and Lb. plantarum were cloned into two different nisin-controlled expression (NICE) vectors and expressed using Lactococcus lactis NZ9000 and NZ3900 as hosts. The lacL gene, encoding the large subunit of the beta-galactosidases, was fused translationally downstream of the nisin-inducible promoter nisA. Chloramphenicol was employed as selection marker for the standard system using L. lactis NZ9000, whereas lactose utilization based on the complementation of the lacF gene was used as a dominant selection marker for the food-grade system employing L. lactis NZ3900. Comparison of the standard and the food-grade expression system, differing only in their selection markers, gave considerable differences in volumetric beta-galactosidase activity, ranging from 1.17 to 14 kU/L of fermentation broth, depending on both the origin of the lacLM genes and the selection marker used. The occurrence of codons less frequently used by L. lactis especially at the beginning of the lacL gene could be an explanation for the significant differences between the expression levels of lacLM from different origins, while plasmid stability might cause the difference obtained when employing the different selection markers
A HPLC-based glycoanalytical protocol allows the use of natural O-glycans derived from glycoproteins as substrates for glycosidase discovery from microbial culture
Many disorders are characterised by changes in O-glycosylation, but analysis of O-glycosylation has been limited by the availability of specific endo- and exo-glycosidases. As a result chemical methods are employed. However, these may give rise to glycan degradation, so therefore novel O-glycosidases are needed. Artificial substrates do not always identify every glycosidase activity present in an extract. To overcome this, an HPLC-based protocol for glycosidase identification from microbial culture was developed using natural O-glycans and O-glycosylated glycoproteins (porcine stomach mucin and fetuin) as substrates. O-glycans were released by ammonia-based β-elimination for use as substrates, and the bacterial culture supernatants were subjected to ultrafiltration to separate the proteins from glycans and low molecular size molecules. Two bacterial cultures, the psychrotroph Arthrobacter C1-1 and a Corynebacterium isolate, were examined as potential sources of novel glycosidases. Arthrobacter C1-1 culture contained a β-galactosidase and N-acetyl-β-glucosaminidase when assayed using 4-methylumbelliferyl substrates, but when defucosylated O-glycans from porcine stomach mucin were used as substrate, the extract did not cleave β-linked galactose or N-acetylglucosamine. Sialidase activity was identified in Corynebacterium culture supernatant, which hydrolysed sialic acid from fetuin glycans. When both culture supernatants were assayed using the glycoproteins as substrate, neither contained endoglycosidase activity. This method may be applied to investigate a microbial or other extract for glycosidase activity, and has potential for scale-up on high-throughput platforms

Functional expression of plant membrane proteins in Lactococcus lactis.
International audienceThe study of most membrane proteins remains challenging due to their hydrophobicity and their low natural abundance in cells. Lactococcus lactis, a Gram-positive lactic bacterium, has been traditionally used in food fermentations and is nowadays widely used in biotechnology for large-scale production of heterologous proteins. This system has been successfully used for the production of prokaryotic and eukaryotic membrane proteins. The purpose of this chapter is to provide detailed protocols for (1) the expression of plant peripheral or intrinsic membrane proteins and then for (2) their solubilization, from Lactococcus membranes, for further purification steps and biochemical characterization
