Efficient, positive, and energy stable schemes for multi-D Poisson-Nernst-Planck systems
In this paper, we design, analyze, and numerically validate positive and
energy-dissipating schemes for solving the time-dependent multi-dimensional
system of Poisson-Nernst-Planck (PNP) equations, which has found much use in
the modeling of biological membrane channels and semiconductor devices. The
semi-implicit time discretization based on a reformulation of the system gives
a well-posed elliptic system, which is shown to preserve solution positivity
for arbitrary time steps. The first order (in time) fully-discrete scheme is
shown to preserve solution positivity and mass conservation unconditionally,
and energy dissipation with only a mild time step restriction. The
scheme is also shown to preserve the steady-state. For the fully second order
(in both time and space) scheme with large time steps, solution positivity is
restored by a local scaling limiter, which is shown to maintain the spatial
accuracy. These schemes are easy to implement. Several three-dimensional
numerical examples verify our theoretical findings and demonstrate the
accuracy, efficiency, and robustness of the proposed schemes, as well as the

CD47 Deficiency Protects Mice from Diet-Induced Obesity and Improves Whole Body Glucose Tolerance and Insulin Sensitivity
CD47 is a transmembrane protein with several functions including self-recognition, immune cell communication, and cell signaling. Although it has been extensively studied in cancer and ischemia, CD47 function in obesity has never been explored. In this study, we utilized CD47 deficient mice in a high-fat diet induced obesity model to study for the first time whether CD47 plays a role in the development of obesity and metabolic complications. Male CD47 deficient and wild type (WT) control mice were fed with either low fat (LF) or high fat (HF) diets for 16 weeks. Interestingly, we found that CD47 deficient mice were protected from HF diet-induced obesity displaying decreased weight gain and reduced adiposity. This led to decreased MCP1/CCR2 dependent macrophage infiltration into adipose tissue and reduced inflammation, resulting in improved glucose tolerance and insulin sensitivity. In addition, CD47 deficiency stimulated the expression of UCP1 and carnitine palmitoyltransferase 1b (CPT1b) levels in brown adipose tissue, leading to increased lipid utilization and heat production. This contributes to the increased energy utilization and reduced adiposity observed in these mice. Taken together, these data revealed a novel role for CD47 in the development of obesity and its related metabolic complications
Leveraging very-high spatial resolution hyperspectral and thermal UAV imageries for characterizing diurnal indicators of grapevine physiology
Efficient and accurate methods to monitor crop physiological responses help growers better understand crop physiology and improve crop productivity. In recent years, developments in unmanned aerial vehicles (UAV) and sensor technology have enabled image acquisition at very-high spectral, spatial, and temporal resolutions. However, potential applications and limitations of very-high-resolution (VHR) hyperspectral and thermal UAV imaging for characterization of plant diurnal physiology remain largely unknown, due to issues related to shadow and canopy heterogeneity. In this study, we propose a canopy zone-weighting (CZW) method to leverage the potential of VHR (≤9 cm) hyperspectral and thermal UAV imageries in estimating physiological indicators, such as stomatal conductance (Gs) and steady-state fluorescence (Fs). Diurnal flights and concurrent in-situ measurements were conducted during grapevine growing seasons in 2017 and 2018 in a vineyard in Missouri, USA. We used neural net classifier and the Canny edge detection method to extract pure vine canopy from the hyperspectral and thermal images, respectively. Then, the vine canopy was segmented into three canopy zones (sunlit, nadir, and shaded) using K-means clustering based on the canopy shadow fraction and canopy temperature. Common reflectance-based spectral indices, sun-induced chlorophyll fluorescence (SIF), and simplified canopy water stress index (siCWSI) were computed as image retrievals. Using the coefficient of determination (R2) established between the image retrievals from three canopy zones and the in-situ measurements as a weight factor, weighted image retrievals were calculated and their correlation with in-situ measurements was explored. The results showed that the most frequent and the highest correlations were found for Gs and Fs, with CZW-based Photochemical reflectance index (PRI), SIF, and siCWSI (PRICZW, SIFCZW, and siCWSICZW), respectively. When all flights combined for the given field campaign date, PRICZW, SIFCZW, and siCWSICZW significantly improved the relationship with Gs and Fs. The proposed approach takes full advantage of VHR hyperspectral and thermal UAV imageries, and suggests that the CZW method is simple yet effective in estimating Gs and Fs
The Effects of Aqueous Extract from Nardostachys chinensis
Aims. The aim of this study was to investigate the effects of the aqueous extract of Nardostachys chinensis Batalin (NCBAE) on blood pressure and cardiac hypertrophy using two-kidney one-clip (2K1C) hypertensive rats. Methods. 2K1C rat models were set up by clipping the left renal artery. Sham-operated rats underwent the same surgical procedure except for renal arterial clipping. 2K1C hypertensive rats were orally given NCBAE at doses of 210, 420, and 630 mg·kg−1·d−1 for 6 weeks. Twelve weeks after surgery, rat SBP and echocardiographic parameters were measured, cardiac histopathology was assessed, serum NO and LDH were detected, and the expression of Bcl-2 and caspase-3 of left ventricular tissue was assessed by western blot. Results. Treatment with NCBAE resulted in a decrease of SBP, LVPWd, LVPWs, IVSd, IVSs, LVW/BW ratio, and cardiomyocyte CSA, an increase of LVEF, and inhibition of 2K1C-induced reduction in serum NO and elevation of LDH compared with 2K1C group. NCBAE intervention also showed a significant increase of Bcl-2 expression and reduction of cleaved caspase-3 level dose-dependently in left ventricular tissue. Conclusion. Our data demonstrate that NCBAE has an antihypertensive property and protective effect on 2K1C-induced cardiac hypertrophy especially at the dose of 630 mg·kg−1·d−1
Increases in Vein Length Compensate for Leaf Area Lost to Lobing in Grapevine
Premise:Leaf lobing and leaf size vary considerably across and within species,including among grapevines (Vitisspp.), some of the best‐studied leaves. Weexamined the relationship between leaf lobing and leaf area across grapevinepopulations that varied in extent of leaf lobing.Methods:We used homologous landmarking techniques to measure 2632 leavesacross 2 years in 476 unique, genetically distinct grapevines fromfive biparentalcrosses that vary primarily in the extent of lobing. We determined to what extent leafarea explained variation in lobing, vein length, and vein to blade ratio.Results:Although lobing was the primary source of variation in shape across theleaves we measured, leaf area varied only slightly as a function of lobing. Rather, leafarea increases as a function of total major vein length, total branching vein length, andvein to blade ratio. These relationships are stronger for more highly lobed leaves, withthe residuals for each model differing as a function of distal lobing.Conclusions:For leaves with different extents of lobing but the same area, the morehighly lobed leaves have longer veins and higher vein to blade ratios, allowing themto maintain similar leaf areas despite increased lobing. Thesefindings show howmore highly lobed leaves may compensate for what would otherwise result in areduced leaf area, allowing for increasedphotosynthetic capacity through similarleaf siz
Development of fermented sea buckthorn (Hippophae rhamnoides L.) juice and investigation of its antioxidant and antimicrobial activity
Sea buckthorn (Hippophae rhamnoides L.) is an edible and medicinal plant species. However, due to its sour taste, it is not readily accepted by consumers. To overcome this, fermentation can be used to change its flavor profile. In this study, we used response surface methodology (RSM) to determine the best process for producing fermented sea buckthorn juice (FSBJ) using probiotics. The biological enzyme activity and total flavonoid content (TFC) of sea buckthorn juice (SBJ) increased after fermentation. When the number of bacteria inoculated was 4.08 × 106 CFU/mL and the inoculation ratio was 30% Z. mobilis, 5% L. casei, 13.75% L. plantarum, 31.25% P. acidilactici, 12.5% L. animalis, and 7.5% P. pentosaceus, the amount of sugar was 2.98% (w/v) after 20 h of fermentation at 37°C, and the superoxide dismutase (SOD) activity reached 725.44 U/mL, and the TFC reached 2.38 mg/mL. FSBJ demonstrated strong antimicrobial activity against Escherichia coli, Staphylococcus aureus and Botrytis cinerea. Then, to investigate the antioxidant capacity of FSBJ, we used H2O2 to induce oxidative stress in C2C12 cells and assessed the protection conferred by FSBJ to damaged cells. It was discovered that after 24 h of treatment with FSBJ, not only was there an increase in the activities of intracellular SOD and glutathione peroxidase (GSH-Px), but also a reduction in reactive oxygen species (ROS) content, catalase (CAT) activity, and malondialdehyde (MDA) content. This research lays the theoretical groundwork and provides reference materials for the improved fermentation of sea buckthorn and demonstrates its resulting antioxidant effect
