    New developments in the methodology available for the assessment of topical corticosteroid-induced skin blanching

    Since the publication of the previous edition of this book there have been considerable developments and controversy in the field of topical corticosteroid bioequivalence assessment. There has been considerable discussion in the literature concerning the use of the Minolta chromameter for the measurement of corticosteroid-induced skin blanching, as it is believed this instrument would produce more objective results than the visual grading procedure. These efforts culminated in the release of a guidance document from the Food and Drug Administration (FDA) detailing the procedures to be followed for the determination of topical corticosteroid bioequivalence using the chromameter. Since the promulgation of this document there have been challenges on the validity and scientific merit of the documented procedures, and recently the FDA itself conceded that it may be necessary to redefine some of the protocol evaluations.     The human skin-blanching (or vasoconstrictor) assay has evolved from initial observations that corticosteroids induce a pallor or whitening of the skin to which they are applied. McKenzie and Stoughton (1962) are generally recognized as having developed the first scientific bioassay for comparing corticosteroid potency. The extensive use of this bioassay to compare drug release from topical delivery systems has demonstrated numerous instances in which the topical bioavailability may vary greatly, dependent on the character of the delivery vehicle. It has become evident that simply incorporating an intrinsically potent drug into a formulation does not necessarily produce a clinically efficacious product

    The human skin blanching assay remains in widespread use as a reliable, qualitative, comparative indicator of topical corticosteroid availability and potency. The experimental refinements promulgated by certain researchers in this field have yielded a versatile bioassay for the accurate assessment of new drugs or delivery vehicles. With the increasing appearance of generic topical corticosteroid formulations which compete with trade-name equivalents, the vital importance of this assay in regulatory affairs and assessing bioequivalence has been re-emphasized. It is stressed that if the blanching assay is to be used in this sphere, then multiple-reading trials must be conducted; important registration or clinical decisions cannot be made with any validity from short-term assessments

    Potentials in Li-Ion Batteries Probed by Operando Ambient Pressure Photoelectron Spectroscopy

    The important electrochemical processes in a battery happen at the solid/liquid interfaces. Operando ambient pressure photoelectron spectroscopy (APPES) is one tool to study these processes with chemical specificity. However, accessing this crucial interface and identifying the interface signal are not trivial. Therefore, we present a measurement setup, together with a suggested model, exemplifying how APPES can be used to probe potential differences over the electrode/electrolyte interface, even without direct access to the interface. Both the change in electron electrochemical potential over the solid/liquid interface, and the change in Li chemical potential of the working electrode (WE) surface at Li-ion equilibrium can be probed. Using a Li4Ti5O12 composite as a WE, our results show that the shifts in kinetic energy of the electrolyte measured by APPES can be correlated to the electrochemical reactions occurring at the WE/electrolyte interface. Different shifts in kinetic energy are seen depending on if a phase transition reaction occurs or if a single phase is lithiated.     Sn-Pb Mixed Perovskites with Fullerene-Derivative Interlayers for Efficient Four-Terminal All-Perovskite Tandem Solar Cells

    Interfacial engineering is the key to high-performance perovskite solar cells (PSCs). While a wide range of fullerene interlayers are investigated for Pb-based counterparts with a bandgap of >1.5 eV, the role of fullerene interlayers is barely investigated in Sn-Pb mixed narrow-bandgap (NBG) PSCs. In this work, two novel solution-processed fullerene derivatives are investigated, namely indene-C60-propionic acid butyl ester and indene-C60-propionic acid hexyl ester (IPH), as the interlayers in NBG PSCs. It is found that the devices with IPH-interlayer show the highest performance with a remarkable short-circuit current density of 30.7 mA cm−2 and a low deficit in open-circuit voltage. The reduction in voltage deficit down to 0.43 V is attributed to reduced non-radiative recombination that the authors attribute to two aspects: 1) a higher conduction band offset of ≈0.2 eV (>0 eV) that hampers charge-carrier-back-transfer recombination; 2) a decrease in trap density at the perovskite/interlayer/C60 interfaces that results in reduced trap-assisted recombination. In addition, incorporating the IPH interlayer enhances charge extraction within the devices that results in considerable enhancement in short-circuit current density. Using a NBG device with an IPH interlayer, a respectable power conversion efficiency of 24.8% is demonstrated in a four-terminal all-perovskite tandem solar cell

    This workshop, `In Vivo Percutaneous Penetration/Absorption' was held in Washington, DC, on May 1-3, 1989. The first workshop in this series, `In Vitro Percutaneous Penetration', took place in November 1986 (the report of this earlier meeting was published in Pharmaceutical Research, 4 (1987) 265-267). The objectives of the workshop were to review the relevant literature and to address in detail: (1) In vivo percutaneous penetration/absorption methodology; (2) The characteristics of dosage forms designed for application to the skin; (3) Critical factors controlling in vivo drug transport into and across the skin; (4) The use of models in the assessment and evaluation of in vivo percutaneous penetration/absorption; and (5) Bioavailability/bioequivalence considerations for topical drug products. Scientific knowledge and technology are rapidly evolving in the topical and transdermal drug products area.     Healthy people with nature in mind

Available from the publisher via the DOI in this record.BACKGROUND: The global disease burden resulting from climate change is likely to be substantial and will put further strain on public health systems that are already struggling to cope with demand. An up- stream solution, that of preventing climate change and associated adverse health effects, is a promising approach, which would create win-win-situations where both the environment and human health benefit. One such solution would be to apply methods of behaviour change to prompt pro-environmentalism, which in turn benefits health and wellbeing. DISCUSSION: Based on evidence from the behavioural sciences, we suggest that, like many social behaviours, pro- environmental behaviour can be automatically induced by internal or external stimuli. A potential trigger for such automatic pro-environmental behaviour would be natural environments themselves. Previous research has demonstrated that natural environments evoke specific psychological and physiological reactions, as demonstrated by self-reports, epidemiological studies, brain imaging techniques, and various biomarkers. This suggests that exposure to natural environments could have automatic behavioural effects, potentially in a pro-environmental direction, mediated by physiological reactions. Providing access and fostering exposure to natural environments could then serve as a public health tool, together with other measures, by mitigating climate change and achieving sustainable health in sustainable ecosystems. However, before such actions are implemented basic research is required to elucidate the mechanisms involved, and applied investigations are needed to explore real world impacts and effect magnitudes. As environmental research is still not sufficiently integrated within medical or public health studies there is an urgent need to promote interdisciplinary methods and investigations in this critical field. Health risks posed by anthropogenic climate change are large, unevenly distributed, and unpredictable. To ameliorate negative impacts, pro-environmental behaviours should be fostered. Potentially this could be achieved automatically through exposure to favourable natural environments, with an opportunity for cost-efficient nature-based solutions that provide benefits for both the environment and public health
