    Purification and characterization of cyclodextrin glucanotransferase from alkalophilic Bacillus sp. G1

    A cyclodextrin glucanotransferase (CGTase) was successively purified by ammonium sulphate precipitation, and affinity chromatography on a-CD (epoxy)-Sepharose 6B column. The specific activity of the CGTase was increased approximately 2200-fold, from 8.43 U/mg protein to 18,866 U/mg protein. SDS-PAGE showed that the purified CGTase was homogeneous and the molecular weight of the purified CGTase was about 75 kDa. The molecular weight of the enzyme that was estimated by gel filtration under native condition was 79 kDa. This has indicated that Bacillus sp. G1 CGTase is a monomeric protein. The isoelectric point (pI) of the enzyme was about 8.8. Characterization of the enzyme exhibited optimum pH and temperature of 6.0 and 60 8C, respectively. The enzyme was stable from pH 7.0 to 9.0 and retained its high activity up to 60 8C. However, in the presence of 20 mM Ca2+, the purified CGTase is able to prolong its thermal stability up to 70 8C. CGTase was strongly inhibited by ZnSO4, CuSO4, CoCl2, FeSO4, FeCl3 and EDTA. Km and Vmax for the purified enzyme were 0.15 mg/ml and 60.39 mg bcyclodextrin/( ml min), respectively, with soluble starch as substrate. In cyclodextrin production, tapioca starch was found to be the best substrate used to produce CDs. The enzyme produced g- and b-CD in the ratio of 0.11:0.89 after 24 h incubation at 60 8C, without the presence of any selective agents

    An examination of the effects of self-regulatory focus on the perception of the media richness: the case of email

    Communication is a key element in organizations’ business success. The media richness theory and the channel expansion theory are two of the most influential theories regarding the selection and use of communication media in organizations; however, literature has focused little on the effects of self-regulation by managers and employees in these theories. To analyze these topics, this study develops an empirical investigation by gathering data from 600 managers and employees using a questionnaire.     Gendered Representations of Male and Female Social Actors in Iranian Educational Materials

    This research investigates the representations of gendered social actors within the subversionary discourse of equal educational opportunities for males and females in Iranian English as a Foreign Language (EFL) books. Using critical discourse analysis (CDA) as the theoretical framework, the authors blend van Leeuwen’s (Texts and practices: Readings in critical discourse analysis, Routledge, London, 2003) ‘Social Actor Network Model’ and Sunderland’s (Gendered discourses, Palgrave Macmillan, Hampshire, 2004) ‘Gendered Discourses Model’ in order to examine the depictions of male and female social actors within this gendered discourse. The gendered discourse of equal opportunities was buttressed by such representations within a tight perspective in proportion to gender ideologies prevailing in Iran. Resorting to CDA, we can claim that resistance against such gendered discourse in Iranian EFL textbooks militates against such gender norms. These representations of male and female social actors in school books are indicative of an all-encompassing education, reinforcing that the discourse of equal opportunities is yet to be realized in the education system of Iran

    The Role of Anti- Cathapsin G among Patients with Rheumatiod Arthritis

    Background: Perinuclear Antineutrophil Cytoplasmic Autoantibodies (pANCA) have been found in patients with rheumatiod arthritis. Cathepsin G was the major target antigen.The present study was to investigate the unknown target antigen of ANCA (Cathapsin G ) in patients with rheumatiod arthritis Objective: This study is to investigate the prevalence of anti-Cathapsin G and rheumatoid factor in Iraqi patients with rheumatiod arthritis. Patienties&Methods: From1st January until 30 June of 2011 fourty five rheumatiod arthritis patients referred to the immunological department in the teaching laboratory of medical city and twenty five apparently healthy individual used as a control group were investeged to rheumatic factor IgG, IgM ,IgA isotypes and anti-Cathapsin G were measured by using enzyme immunoassay technique. Result: This study showed that the age of patients with rheumatiod arthritis ranged between eigthen years and sixty six years with mean age of (36.08) , while the mean level for RF screen, RF IgG, RF IgM, RF IgA isotypes and anti-Cathapsin G were (160.71, 91.7, 136.64, 95.03 and > 9.2 ) respectively. There were 17(37.8%) males and 28(62.2%) females of RA patients. Immunologically, this study revealed that the serum RF screen was found more than 25 U/ml in all rheumatiod arthritis patients (100%, P: 0.0001 HS). On the other hand RF isotypes IgG ,RF IgM and IgA level were elevated more than 20 U/ml and it was detected in 31 (68.9%, P: 0.0001 HS), 31 (68.9%, P : 0.0001 HS) ,and 32 (71.1%, P: 0.0001 HS) patients respectively, while the rheumatiod factors (IgG,IgM, and IgA)were normal in healthy control group . Statistically RF screen has a highly significant association with RF IgG, IgM, and IgA (P= 0.0001). Anti-Cathapsin G was found in 12(26.7%, P value: 0.019) of patients with rheumatic arthritis patients. This study showed a association between the positive result of anti-Cathapsin G with RF IgG and RF IgM (35.5%, P: 0.047) the results were the same for both, while no association found with RF IgA (25%, P: 0.692). Conclusion: A significant correlation was found between anti-Cathapsin G and patients with rheumatiod arthritis and has a significant association with RF IgG and RF IgM while RF IgA had no relation with anti-Cathapsin G

    Multilocational testing of pigeonpea for broad-based resistance to sterility mosaic in India

    During 1978-83, 88 pigeon pea lines resistant to sterility mosaic (SM) from different research centres in India were tested at 10 locations (Badnapur, Bangalore, Dholi, Pantnagar, Faizabad, Kanpur, Ludhiana, Patancheru, Vamban and Varanasi) to identify lines with stable and broad-based resistance. The multilocation evaluation was carried out through the joint Indian Council of Agricultural Research and the ICRISAT Uniform Trial for Pigeon Pea Sterility Mosaic Resistance. SM resistant genotypes were identified at each of the 10 locations. Lines ICP 7867, ICP 10976 and ICP 10977 were resistant or tolerant at all 10 locations. These lines are now being used by breeders at ICRISAT as well as in the Indian national programme for developing SM resistant and high yielding cultivars

    Tau and Charm physics highlights

    In tau physics, we are at the frontier between the completion of the LEP program and the start of analyses from b-factories, which are expected to produce results in the coming years. Nice results from CLEO are steadily delivered in the meantime. For charm, impressive progress have been achieved by fixed target experiments in the search for CP violation and D^0 - \bar D^0 oscillations. First results from b-factories demonstrate the power of these facilities in such areas. The novel measurement of the D* width by CLEO happens to be rather different from current expectations. The absence of a charm factory explains the lack or the very slow progress in the absolute scale determinations for charm decays.Comment: "Typos corrected and references added

    Space-charge limited conduction in epitaxial chromia films grown on elemental and oxide-based metallic substrates

    We study temperature dependent (200 – 400 K) dielectric current leakage in high-quality, epitaxial chromia films, synthesized on various conductive substrates (Pd, Pt and V2O3). We find that trap-assisted space-charge limited conduction is the dominant source of electrical leakage in the films, and that the density and distribution of charge traps within them is strongly dependent upon the choice of the underlying substrate. Pd-based chromia is found to exhibit leakage consistent with the presence of deep, discrete traps, a characteristic that is related to the known properties of twinning defects in the material. The Pt- and V2O3-based films, in contrast, show behavior typical of insulators with shallow, exponentially-distributed traps. The highest resistivity is obtained for chromia fabricated on V2O3substrates, consistent with a lower total trap density in these films. Our studies suggest that chromia thin films formed on V2O3 substrates are a promising candidate for next-generation spintronics

    Punica granatum (Pomegranate) juice provides an HIV-1 entry inhibitor and candidate topical microbicide

    BACKGROUND: For ≈ 24 years the AIDS pandemic has claimed ≈ 30 million lives, causing ≈ 14,000 new HIV-1 infections daily worldwide in 2003. About 80% of infections occur by heterosexual transmission. In the absence of vaccines, topical microbicides, expected to block virus transmission, offer hope for controlling the pandemic. Antiretroviral chemotherapeutics have decreased AIDS mortality in industrialized countries, but only minimally in developing countries. To prevent an analogous dichotomy, microbicides should be: acceptable; accessible; affordable; and accelerative in transition from development to marketing. Already marketed pharmaceutical excipients or foods, with established safety records and adequate anti-HIV-1 activity, may provide this option. METHODS: Fruit juices were screened for inhibitory activity against HIV-1 IIIB using CD4 and CXCR4 as cell receptors. The best juice was tested for inhibition of: (1) infection by HIV-1 BaL, utilizing CCR5 as the cellular coreceptor; and (2) binding of gp120 IIIB and gp120 BaL, respectively, to CXCR4 and CCR5. To remove most colored juice components, the adsorption of the effective ingredient(s) to dispersible excipients and other foods was investigated. A selected complex was assayed for inhibition of infection by primary HIV-1 isolates. RESULTS: HIV-1 entry inhibitors from pomegranate juice adsorb onto corn starch. The resulting complex blocks virus binding to CD4 and CXCR4/CCR5 and inhibits infection by primary virus clades A to G and group O. CONCLUSION: These results suggest the possibility of producing an anti-HIV-1 microbicide from inexpensive, widely available sources, whose safety has been established throughout centuries, provided that its quality is adequately standardized and monitored
