Heterochromatin Dynamics in Response to Environmental Stress in Amazonian Fish
Transcriptionally inactive portions of genomic DNA, condensed with histones and architectural proteins, are known as heterochromatic regions, often positive C band. The advent of epigenetics and new methodological approaches, showed that these regions are extremely dynamic and responsive to different types of environmental stress. The relationship of the constitutive heterochromatin with the transposable elements inactivation, especially from the Rex family, seems to be a frequent condition in fish. In this manuscript we review the existing knowledge of the nature and function of these genomic regions, based on species-based studies, with a focus on species of fish from the Amazon region
Composition and Nature of Heterochromatin in the Electrical Fish (Knifefishes) <em>Gymnotus</em> (Gymnotiformes: Gymnotidae)
Fishes of the genus Gymnotus have been suggested as a good model for biogeographic studies in the South American continent. In relation to heterochromatin, species of this genus have blocks preferably distributed in the centromeric region. The content of these regions has been shown to be variable, with description of transposable elements, pseudogenes of 5S rDNA and satellite sequences. In G. carapo Clade, although geographically separated, species with 2n = 54 chromosomes share the distribution of many 5S rDNA sites, a unique case within the genus. Here, repetitive DNA sequences from G. sylvius (2n = 40) and G. paraguensis (2n = 54) were isolated and mapped to understand their constitution. The chromosome mapping by FISH showed an exclusive association in the centromeres of all chromosomes. However, the cross-FISH did not show positive signs of interspecific hybridization, indicating high levels of heterochromatic sequence specificity. In addition, COI-1 sequences were analyzed in some species of Gymnotus, which revealed a close relationship between species of clade 2n = 54, which have multiple 5S rDNA sites. Possibly, the insertion of retroelements or pseudogenization and dispersion of this sequence occurred before the geographic dispersion of the ancestor of this clade from the Amazon region to the hydrographic systems of Paraná-Paraguay, a synapomorphy for the group
Karyotype variability in neotropical catfishes of the family Pimelodidae (Teleostei: Siluriformes)
Karyotypic data are presented for four species of fish belonging to the Pimelodidae family. These species show a conserved diploid number, 2n = 56 chromosomes, with different karyotypic formulae. The analyzed species showed little amount of heterochromatin located preferentially in the centromeric and telomeric regions of some chromosomes. The nucleolus organizer regions activity (Ag-NORs) and the chromosomal location of ribosomal genes by fluorescent in situ hybridization (FISH), with 18S and 5S probes, showing only one chromosome pair marked bearer of ribosomal genes, the only exception was Pimelodus britskii that presented multiple NORs and syntenic location of the 18S and 5S probes. Non-Robertsonian events, as pericentric inversion and NORs duplication are requested to explain the karyotype diversification in Pseudoplatystoma from the rio Paraguay (MS), Pimelodus from the rio Iguaçu (PR), Sorubim from the rio Paraguay (MS) and Steindachneridion from the rio Paraíba do Sul (SP). The obtained data for the karyotype macrostructure of these species corroborates a conserved pattern observed in Pimelodidae. On the other hand, interspecific variations detected by molecular cytogenetics markers made possible cytotaxonomic inferences and differentiation of the species here analyzed. Dados cariotípicos são apresentados para quatro espécies da família Pimelodidae. Todas apresentaram o mesmo número diploide, 2n = 56 cromossomos, com diferenças nas fórmulas cariotípicas. As espécies aqui analisadas mostraram pouca quantidade de heterocromatina localizada preferencialmente na região centromérica e telomérica de alguns cromossosmos do complemento cariotípico. As regiões organizadoras de nucléolo (Ag-RONs) e a localização dos genes ribossomais pela hibridização in situ fluorescente (FISH), com sondas 18S e 5S, evidenciaram somente um par cromossômico marcado portador de genes ribossomais, à exceção de Pimelodus britskii que apresentou NORs múltiplas e localização sintênica das sondas 18S e 5S. Eventos não-Robertsonianos, como inversão pericêntrica e duplicação das NORs são requeridos para explicar a diversificação cariotípica em Pseudoplatystoma do rio Paraguai (MS), Pimelodus do rio Iguaçu (PR), Sorubim do rio Paraguai (MS) e Steindachneridion do rio Paraíba do Sul (SP). Os dados obtidos para a macroestrutura cariotípica destas espécies corrobora um padrão conservado observado na família Pimelodidae. Por outro lado, evidências de variações interespecíficas pelos marcadores de citogenética molecular empregados possibilitam inferências citotaxonômicas e diferenciação das espécies aqui analisadas
Relação entre o desempenho no vestibular e o desempenho durante o curso de graduação
Para ingressar no ensino superior público ou privado o candidato precisa realizar um exame de ingresso, o chamado exame vestibular. A formação básica, a qual tem a função de desenvolver o educando, dando subsídios para exercer a cidadania e também para progredir no trabalho e em estudos posteriores, tem um papel muito importante para a aprovação no vestibular. Aberto aos concluintes do ensino médio e aprovados no vestibular, o ensino superior deve ser acessível a todas as pessoas no decorrer da vida. O nosso objetivo com o presente trabalho foi o de verificar se os primeiros e os últimos colocados no vestibular para o curso de ciências biológicas mantêm esses resultados durante o curso, e quais os fatores que contribuem para a continuidade ou não desse desempenho. Para coleta de dados foram aplicados questionários direcionados aos cinco primeiros e cinco últimos classificados no vestibular para o curso de ciências biológicas nas duas modalidades licenciatura e bacharelado; também foram analisados os históricos acadêmicos dos indivíduos, a fim de determinar o desempenho nas disciplinas. Para estudo dos resultados foi utilizada uma análise quantiqualitativa. Concluímos que, de maneira geral, os primeiros colocados mantêm um desempenho relativamente melhor que os últimos, e que fatores como a formação básica (ensino fundamental e médio), trabalhar durante o curso e a idade dos alunos são intimamente relacionados com o desempenho durante o mesmo. As duas turmas analisadas mostram características peculiares, e o período em que os cursos são ofertados tem papel relevante nesta diferença, pois abrange públicos muito distintos, e consequentemente reflete dados muito contrastantes. A prova de vestibular de maneira imediata separa quais são os alunos mais bem preparados dos menos preparados para a trajetória acadêmica, no entanto durante o curso outros fatores como família, emprego, formação básica, identificação com o curso determinam o desempenho destes alunos. Outras pesquisas que visem a caracterizar a condição sócioeconômica e também condições de aprendizado de cada aluno parecem ser necessárias para afirmar com exatidão que estes fatores contribuem efetivamente para um desempenho considerado bom ou ruim

Chromosome Painting in Gymnotus carapo "Catalão" (Gymnotiformes, Teleostei): Dynamics of Chromosomal Rearrangements in Cryptic Species.
The genus Gymnotus is a large monophyletic group of freshwater weakly-electric fishes, with wide distribution in Central and South America. It has 46 valid species divided into six subgenera (Gymnotus, Tijax, Tigre, Lamontianus, Tigrinus and Pantherus) with large chromosome plasticity and diploid numbers (2n) ranging from 34 to 54. Within this rich diversity, there is controversy about whether Gymnotus (Gymnotus) carapo species is a single widespread species or a complex of cryptic species. Cytogenetic studies show different diploid numbers for G. carapo species, ranging from 40 to 54 chromosomes with varied karyotypes found even between populations sharing the same 2n. Whole chromosome painting has been used in studies on fish species and recently has been used for tracking the chromosomal evolution of Gymnotus and assisting in its cytotaxonomy. Comparative genomic mapping using chromosome painting has shown more complex rearrangements in Gymnotus carapo than shown in previous studies by classical cytogenetics. These studies demonstrate that multiple chromosome pairs are involved in its chromosomal reorganization, suggesting the presence of a complex of cryptic species due to a post zygotic barrier. In the present study, metaphase chromosomes of G. carapo occidentalis "catalão" (GCC, 2n = 40, 30m/sm+10st/a) from the Catalão Lake, Amazonas, Brazil, were hybridized with whole chromosome probes derived from the chromosomes of G. carapo (GCA, 2n = 42, 30m/sm+12st/a). The results reveal chromosome rearrangements and a high number of repetitive DNA sites. Of the 12 pairs of G. carapo chromosomes that could be individually identified (GCA 1-3, 6, 7, 9, 14, 16 and 18-21), 8 pairs (GCA 1, 2, 6, 7, 9, 14, 20, 21) had homeology conserved in GCC. Of the GCA pairs that are grouped (GCA [4, 8], [5, 17], [10, 11] and [12, 13, 15]), most kept the number of signals in GCC (GCA [5, 17], [10, 11] and [12, 13, 15]). The remaining chromosomes are rearranged in the GCC karyotype. Analysis of both populations of the G. carapo cytotypes shows extensive karyotype reorganization. Along with previous studies, this suggests that the different cytotypes analyzed here may represent different species and supports the hypothesis that G. carapo is not a single widespread species, but a group of cryptic species
