    Exploration of Bis-amino Acid Scaffolds Designed to Self Assemble

    Based on theoretical models scaffold 13 was predicted to self assemble through intermolecular hydrogen bonds.  The synthesis of scaffold 13 was successfully synthesized using standard Fmoc Solid Phase Peptide Synthesis.  Connectivity was confirmed through full structural characterization.  Scaffold 13 was probed for its ability to self-assemble in DMSOd6 in the presence of 0.2 percent TFA.  Our hypothesis for self-assembly of scaffold 13 was not supported under any of the conditions explored.  ROESY correlations failed to indicate self-assembly between interacting scaffolds and a variable temperature NMR study at three separate concentrations also failed to provide evidence of self-assembly

    Estimating Torque Imparted on Spacecraft Using Telemetry

    There have been a number of missions with spacecraft flying by planetary moons with atmospheres; there will be future missions with similar flybys. When a spacecraft such as Cassini flies by a moon with an atmosphere, the spacecraft will experience an atmospheric torque. This torque could be used to determine the density of the atmosphere. This is because the relation between the atmospheric torque vector and the atmosphere density could be established analytically using the mass properties of the spacecraft, known drag coefficient of objects in free-molecular flow, and the spacecraft velocity relative to the moon. The density estimated in this way could be used to check results measured by science instruments. Since the proposed methodology could estimate disturbance torque as small as 0.02 N-m, it could also be used to estimate disturbance torque imparted on the spacecraft during high-altitude flybys

    Peripheral nervous system manifestations in a Sandhoff disease mouse model: nerve conduction, myelin structure, lipid analysis

    <p>Abstract</p> <p>Background</p> <p>Sandhoff disease is an inherited lysosomal storage disease caused by a mutation in the gene for the β-subunit (<it>Hexb </it>gene) of β-hexosaminidase A (αβ) and B (ββ). The β-subunit together with the GM2 activator protein catabolize ganglioside GM2. This enzyme deficiency results in GM2 accumulation primarily in the central nervous system. To investigate how abnormal GM2 catabolism affects the peripheral nervous system in a mouse model of Sandhoff disease (<it>Hexb-/-</it>), we examined the electrophysiology of dissected sciatic nerves, structure of central and peripheral myelin, and lipid composition of the peripheral nervous system.</p> <p>Results</p> <p>We detected no significant difference in signal impulse conduction velocity or any consistent change in the frequency-dependent conduction slowing and failure between freshly dissected sciatic nerves from the <it>Hexb</it>+/- and <it>Hexb</it>-/- mice. The low-angle x-ray diffraction patterns from freshly dissected sciatic and optic nerves of <it>Hexb</it>+/- and <it>Hexb</it>-/- mice showed normal myelin periods; however, <it>Hexb</it>-/- mice displayed a ~10% decrease in the relative amount of compact optic nerve myelin, which is consistent with the previously established reduction in myelin-enriched lipids (cerebrosides and sulfatides) in brains of <it>Hexb-/- </it>mice. Finally, analysis of lipid composition revealed that GM2 content was present in the sciatic nerve of the <it>Hexb</it>-/- mice (undetectable in <it>Hexb</it>+/-).</p> <p>Conclusion</p> <p>Our findings demonstrate the absence of significant functional, structural, or compositional abnormalities in the peripheral nervous system of the murine model for Sandhoff disease, but do show the potential value of integrating multiple techniques to evaluate myelin structure and function in nervous system disorders.</p

    Cholesterol and Lipoprotein Dynamics in a Hibernating Mammal

    Hibernating mammals cease feeding during the winter and rely primarily on stored lipids to fuel alternating periods of torpor and arousal. How hibernators manage large fluxes of lipids and sterols over the annual hibernation cycle is poorly understood. The aim of this study was to investigate lipid and cholesterol transport and storage in ground squirrels studied in spring, summer, and several hibernation states. Cholesterol levels in total plasma, HDL and LDL particles were elevated in hibernators compared with spring or summer squirrels. Hibernation increased plasma apolipoprotein A-I expression and HDL particle size. Expression of cholesterol 7 alpha-hydroxylase was 13-fold lower in hibernators than in active season squirrels. Plasma triglycerides were reduced by fasting in spring but not summer squirrels. In hibernators plasma β-hydroxybutyrate was elevated during torpor whereas triglycerides were low relative to normothermic states. We conclude that the switch to a lipid-based metabolism during winter, coupled with reduced capacity to excrete cholesterol creates a closed system in which efficient use of lipoproteins is essential for survival

    Feasibility Study of Two Candidate Reaction Wheel/thruster Hybrid Control Architecture Designs for the Cassini Spacecraft

    As the first spacecraft to achieve orbit at Saturn in 2004, Cassini has collected science data throughout its four-year prime mission (2004-08), and has since been approved for a first and second extended mission through 2017. Cassini carries a set of three "fixed" reaction wheels and a backup reaction wheel (reaction wheel #4) is mounted on top of an articulable platform. If necessary, this platform could be articulated to orient the backup reaction wheel with the degraded wheel. The reaction wheels are used primarily for attitude control when precise and stable pointing of a science instrument such as the narrow angle camera is required. In 2001-02, reaction wheel #3 exhibited signs of bearing cage instability. As a result, reaction wheel #4 was articulated to align with reaction wheel #3. Beginning in July 2003, Cassini was controlled using wheel #1, #2, and #4. From their first use in the spring of 2000 until today, reaction wheels #1 and #2 have accumulated more than3.5 billions revolutions each. As such, in spite of very carefully management of the wheel spin rates by the mission operation team, there are some observed increases in the drag torque of the wheels' bearings. Hence, the mission operations team must prepare for the contingency scenario in which the reaction wheel #1 (in addition to wheel #3) had degraded. In this hypothetical fault scenario, the two remaining reaction wheels (#2 and #4) will not be able to provide precise and stable three-axis control of the spacecraft. In this study, we evaluate the feasibility of controlling Cassini using the two remaining reaction wheels and four thrusters to meet the science pointing requirements for two key science operational modes: the Optical Remote Sensing and Downlink, Fields, Particles, & Waves operation modes. The performance (e.g., pointing control error, pointing stability, hydrazine consumption rate, etc.) of the hybrid controllers in both operations scenarios will be compared with those achieved using an all-thruster controller design.     Estimation and Modeling of Enceladus Plume Jet Density Using Reaction Wheel Control Data

    The Cassini spacecraft was launched on October 15, 1997 by a Titan 4B launch vehicle. After an interplanetary cruise of almost seven years, it arrived at Saturn on June 30, 2004. In 2005, Cassini completed three flybys of Enceladus, a small, icy satellite of Saturn. Observations made during these flybys confirmed the existence of a water vapor plume in the south polar region of Enceladus. Five additional low-altitude flybys of Enceladus were successfully executed in 2008-9 to better characterize these watery plumes. The first of these flybys was the 50-km Enceladus-3 (E3) flyby executed on March 12, 2008. During the E3 flyby, the spacecraft attitude was controlled by a set of three reaction wheels. During the flyby, multiple plume jets imparted disturbance torque on the spacecraft resulting in small but visible attitude control errors. Using the known and unique transfer function between the disturbance torque and the attitude control error, the collected attitude control error telemetry could be used to estimate the disturbance torque. The effectiveness of this methodology is confirmed using the E3 telemetry data. Given good estimates of spacecraft's projected area, center of pressure location, and spacecraft velocity, the time history of the Enceladus plume density is reconstructed accordingly. The 1 sigma uncertainty of the estimated density is 7.7%. Next, we modeled the density due to each plume jet as a function of both the radial and angular distances of the spacecraft from the plume source. We also conjecture that the total plume density experienced by the spacecraft is the sum of the component plume densities. By comparing the time history of the reconstructed E3 plume density with that predicted by the plume model, values of the plume model parameters are determined. Results obtained are compared with those determined by other Cassini science instruments
