MAGD Trial-Memantine for Agitation in Alzheimerās Dementia
OBJECTIVE: Does memantine improve agitation in dementia
DESIGN: Double blind randomized placebo controlled trial
SETTING: Nursing Homes
PARTICIPANTS: 153 patients with Alzheimerās dementia
INTERVENTION: Memantine vs placebo
MEASUREMENTS: Cohen Mansfield Agitation Inventory
(CMAI), Neuro-Psychiatric Inventory (NPI), Severe Impairment
Battery (SIB), Quality of Life-AD (QOL-AD)
RESULTS:CMAI-there was no statistically significant difference
for mean change- memantine vs placebo p=0.17.
NPI-9 point difference between memantine vs placebo p=0.01
Hallucinations, aberrant motor behaviour and sleep-p=0.03
Agitation, disinhibition and irritability-p=0.001
SIB-Memantine compared to placebo had a strong augmentation
of the percent change from baseline to week 12 p=0.02
QOL-AD-Memantine group are significantly more satisfied
about the contact with friends compared to the patients treated with
placebo p=0.04.
CONCLUSION:
In this study of agitated AD patients benefit was not shown for
agitation on the CMAI with memantine but the NPI reached signficance
and the agitation, disinhibiton and irritability cluster improvement
was significant. Cognition and quality of life improved. We believe
that this study supports the use of Memantine in agitated
patients with Alzheimerās dementi
