    Decay of geometry for a class of cubic polynomials

    In this paper we study a class of bimodal cubic polynomials satisfying two critical points belong to one invariant Cantor set. These maps have generalized Fibonacci combinatorics in terms of generalized renormalization on the twin principal nest. It is proved that such maps possess `decay of geometry' in the sense that the scaling factor of its twin principal nest decreases at least exponentially fast. As an application, we prove that they have no Cantor attractor

    Forsythiaside A inhibits hydrogen peroxide-induced inflammation, oxidative stress, and apoptosis of cardiomyocytes

    Purpose: To investigate the effect of forsythiaside A on heart failure.Methods: An in vitro cell model of myocardial injury was established by incubating H9c2 primary cardiomyocytes with hydrogen peroxide (H2O2). Apoptosis was measured by flow cytometry. Expression of inflammatory factors, including tumor necrosis factor-alpha (TNF-α) and interleukin-6 (IL-6), was determined by quantitative reverse transcription-polymerase chain reaction (qRT-PCR) and enzymelinkedimmunosorbent assay (ELISA). Oxidative stress was evaluated by measuring malondialdehyde (MDA), superoxide dismutase (SOD), and glutathione peroxidase (GSH-Px) levels by ELISA.Results: Incubation with H2O2 increased H9c2 cell apoptosis (p < 0.001). Treatment with forsythiaside A reduced Bax expression and enhanced Bcl-2 expression which suppressed apoptosis of H2O2- induced H9c2 cells. Forsythiaside A also attenuated the H2O2-induced increase in TNF-α and IL-6expressions in H9c2 cells (p < 0.001). The H2O2-induced increase in MDA and decrease in SOD and GSH-Px in H9c2 cells were reversed by treatment with forsythiaside A. IκBα protein expression was downregulated, whereas p65 phosphorylation (p-p65), p-IκBα, nuclear factor erythropoietin-2-related factor 2 (Nrf2), and heme oxygenase 1 (HO-1) were upregulated in H2O2-induced H9c2 cells. Forsythiaside A increased IκBα, Nrf2, and HO-1 expression and decreased p-p65 and p-IκBα expression in H2O2-induced H9c2 cells.Conclusion: Forsythiaside A exerts anti-inflammatory, anti-oxidant, and anti-apoptotic effects against H2O2-induced H9c2 cells through inactivation of NF-κB pathway and activation of Nrf2/HO-1 pathway. These results support the potential clinical application of forsythiaside A for the treatment of heart failure

    Xanthomonas oryzae pv. oryzae TALE proteins recruit OsTFIIAγ1 to compensate for the absence of OsTFIIAγ5 in bacterial blight in rice

    Xanthomonas oryzae pv. oryzae (Xoo), the causal agent of bacterial blight (BB) of rice, uses transcription activator-like effectors (TALEs) to interact with the basal transcription factor gamma subunit OsTFIIAγ5 (Xa5) and activates the transcription of host genes. However, how OsTFIIAγ1, the other OsTFIIAγ protein, functions in the presence of TALEs remains unclear. In this study, we show that OsTFIIAγ1 plays a compensatory role in the absence of Xa5. The expression of OsTFIIAγ1, which is activated by TALE PthXo7, increases the expression of host genes targeted by avirulent and virulent TALEs. Defective OsTFIIAγ1 rice lines show reduced expression of the TALE-targeted susceptibility (S) genes, OsSWEET11 and OsSWEET14, which results in increased BB resistance. Selected TALEs (PthXo1, AvrXa7 and AvrXa27) were evaluated for interactions with OsTFIIAγ1, Xa5 and xa5 (naturally occurring mutant form of Xa5) using biomolecular fluorescence complementation (BiFC) and microscale thermophoresis (MST). BiFC and MST demonstrated that the three TALEs bind Xa5 and OsTFIIAγ1 with a stronger affinity than xa5. These results provide insights into the complex roles of OsTFIIAγ1 and OsTFIIAγ5 in TALE-mediated host gene transcription

    Spousal care intensity, socioeconomic status, and depression among the older caregivers in China: a study on 2011–2018 CHARLS panel data

    Using the Stress Process Model and data from the 2011-2018 China Health and Retirement Longitudinal Study (CHARLS), this study examined the effect of spousal caregiving intensity on the depression level of older caregivers in China. The moderating role that socioeconomic status plays in the relationship between spouses was explored by constructing Multilevel Growth Models (MGM). The care intensity for a spouse was found to have significantly increased depression levels in older caregivers, while the degree of disability of the spouse being cared for (B=0.200, p<0.001) had a greater effect on depression than the duration of care (B=0.007, p<0.01). There was a threshold effect where the provision of more than 10 hours of care per week for a spouse (B=0.931, p<0.001; B=0.970; p<0.01) or caring for a disabled spouse with limited ADLs (B=0.709, p<0.01; B=1.326; p<0.001; B=1.469, p<0.01) increased depression in older caregivers. There were moderating influences including higher professional prestige before retirement (B=-0.006, p<0.01) and higher annual family income (B=-0.037, p<0.05) that increased depression related to the spouse’s degree of disability. It was considered that active familism measures should be formulated for older spousal caregivers, especially those with lower socioeconomic status

    Overexpression of SSBXoc, a Single-Stranded DNA-Binding Protein From Xanthomonas oryzae pv. oryzicola, Enhances Plant Growth and Disease and Salt Stress Tolerance in Transgenic Nicotiana benthamiana

    We previously reported that SSBXoc, a highly conserved single-stranded DNA-binding protein from Xanthomonas spp., was secreted through the type III secretion system (T3SS) and functioned as a harpin-like protein to elicit the hypersensitive response (HR) in the non-host plant, tobacco. In this study, we cloned SsbXoc gene from X. oryzae pv. oryzicola (Xoc), the causal agent of bacterial leaf streak in rice, and transferred it into Nicotiana benthamiana via Agrobacterium-mediated transformation. The expression of SsbXoc in transgenic N. benthamiana enhanced growth of both seedling and adult plants. When inoculated with the harpin Hpa1 or the pathogen Pseudomonas syringae pv. tomato DC3000 (Pst DC3000), the accumulation of reactive oxygen species (ROS) was increased more in SsbXoc transgenic lines than that in wild-type (WT) plants. The expression of pathogenesis-related protein genes (PR1a and SGT1), HR marker genes (HIN1 and HSR203J) and the mitogen-activated protein kinase pathway gene, MPK3, was significantly higher in transgenic lines than in WT after inoculation with Pst DC3000. In addition, SsbXoc transgenic lines showed the enhanced resistance to the pathogenic bacteria P. s. tabaci and the improved tolerance to salt stress, accompanied by the elevated transcription levels of the defense- and stress-related genes. Taken together, these results indicate that overexpression of the SsbXoc gene in N. benthamiana significantly enhanced plant growth and increased tolerance to disease and salt stress via modulating the expression of the related genes, thus providing an alternative approach for development of plants with improved tolerance against biotic and abiotic stresses

    Targeting of Embryonic Stem Cells by Peptide-Conjugated Quantum Dots

    Targeting stem cells holds great potential for studying the embryonic stem cell and development of stem cell-based regenerative medicine. Previous studies demonstrated that nanoparticles can serve as a robust platform for gene delivery, non-invasive cell imaging, and manipulation of stem cell differentiation. However specific targeting of embryonic stem cells by peptide-linked nanoparticles has not been reported.Here, we developed a method for screening peptides that specifically recognize rhesus macaque embryonic stem cells by phage display and used the peptides to facilitate quantum dot targeting of embryonic stem cells. Through a phage display screen, we found phages that displayed an APWHLSSQYSRT peptide showed high affinity and specificity to undifferentiated primate embryonic stem cells in an enzyme-linked immunoabsorbent assay. These results were subsequently confirmed by immunofluorescence microscopy. Additionally, this binding could be completed by the chemically synthesized APWHLSSQYSRT peptide, indicating that the binding capability was specific and conferred by the peptide sequence. Through the ligation of the peptide to CdSe-ZnS core-shell nanocrystals, we were able to, for the first time, target embryonic stem cells through peptide-conjugated quantum dots.These data demonstrate that our established method of screening for embryonic stem cell specific binding peptides by phage display is feasible.
