    The diversity of leptin gene in Iranian native, Holstein and Brown Swiss cattle

    This study describes genetic variability in the leptin in Iranian native, Brown Swiss and Holstein cattle (Bos Indicus and Bos Taurus). This is the first study of genetic polymorphism of the leptin gene in Iranian native cattle. We examined exon 2 of the leptin gene from 587 individuals in six different populations of Iranian native cattles (86 Sarabi, 66 Taleshi, 94 Sistani, 76 Golpayegani, 104 Brown Swiss and 161 Holstein cattle) using PCR-RFLP method. Analysis of the frequencies of the various alleles ineach breed indicated that allele C in Sarabi, Taleshi, Sistani, Golpayegani, Brown Swiss and Holstein cattle with 68, 55, 69, 71, 55 and 57% value were the most frequent alleles. Observed heterozygosities were highest in Golpayegani (57.89%). These new data suggest that allele frequencies of leptin differ between the various Iranian cattle breeds

    Implication of complex vertebral malformation and deficiency of uridine monophosphate synthase on molecular-based testing in the Iranian Holstein bulls population

    The aim of this study was to identify the deficiency of uridine monophosphate synthase (DUMPS) and the complex vertebral malformation (CVM) in Iranian Holstein bulls. A total of 144 blood samples were prepared of Holstein bulls in Abbas Abad Animal Breeding Center and Ferdowsi University of Mashhad's Dairy Farm in Khorasan state of Iran. Genomic PCR-RFLP protocol was performed to amplify the polymorphic region of the bovine uridine monophosphate synthase UMPS gene. Also, genomic PCR-SSCP method was performed for CVM to amplify the polymorphic region of the bovine solutecarrier family 35 member 3 (SLC35A3) genes. The results of this study demonstrated that there was no carrier of DUMPS and CVM in Iranian bulls in these centers

    Total Antioxidant Activity, and Hesperidin, Diosmin, Eriocitrin and Quercetin Contents of Various Lemon Juices

    Purpose: To investigate 38 natural and 62 branded lemon juices for their total antioxidant activities and flavonoid content.Methods: The ferric reducing antioxidant power was applied to evaluation of antioxidant. Additionally quercetin, eriocitrin, diosmin and hesperidin contents were evaluated by high performance liquid chromatography (HPLC).Results: Total antioxidant activity of the juices ranged from 91.38 ± 0.01 to 526.93 ± 0.12 μg/ml. The maximum contents of quercetin, hesperidin and eriocitrin 14.83 ± 2.83, 104.84 ± 12.58 and 10.68 ± 2.41 μg/mL, respectively. The natural juices had diosmin levels as high as 236.18 mL. The limit of detection (LOD) ranged from 0.15 μg mg-1 for hesperidine to 0.53 μg ml-1 for diosmin while the limit of quantification (LOQ) ranged from 0.27 μg mg-1 for hesperidine to 0.93 μg ml-1 for quercetin.Conclusion: The developed method can also be applied to the determination of related extracts of orange cultivars as well as various extracts of medicinal plants.Keywords: Antioxidant, Flavonoid, Hesperidin, Diosmin, Eriocitrin, Quercetin, Lemon juic

    The Predictive Roles of Self-efficacy, Illness Perception, and Social Support in Self-care of Patients With Heart Failure

    Background: As a chronic disease, heart failure (HF) is a growing problem that not only impacts patients but also their family members and friends. Self-care is the main part of HF treatment. Despite the significance of determining variables involved in self-care, the relationships of self-efficacy, illness perception, and social support with self-care and the predictive roles of these variables in the treatment of HF have not been investigated. Objectives: The present study was aimed at exploring the predictive roles of self-efficacy, illness perception, and social support in self-care of patients with heart failure. Methods: In this predictive correlation study, 149 patients with HF were selected through convenience sampling from Tabriz Research Treatment Centre of Heart in 2016. To collect data, a demographic information questionnaire, self-care behaviour scale, general self-efficacy (GSE), brief illness perception questionnaire (Brief-IPQ), and social support scale were used. To analyse data, SPSS software version 16, descriptive statistics, Pearson correlation coefficient, and multiple stepwise regression analyses were employed. Results: Out of 149 subjects, 102 (68%) participants were male, and 47 (32%) of them were female. The mean and standard deviation of samples’ age were (64.40±10.32) ranging from 37 to 88 years. Most of participants were married (82%). The mean scores were as follows: Self-care (40.66±13.16), self-efficacy (45.81±22.03), illness perception (56.05±18.24), and social support (49.09±6.74). Data analysis demonstrated significant correlations between illness perception and self-care (p<0.001, r=0.649), self-efficacy and self-care (p<0.001, r=-0.678), social support and self-care (p<0.001, r=-0.518), and age and self-care (p<0.001, r=-0.506). The standardized coefficient of illness perception was (β=0.274), social support was (β=-0.237), self-efficacy was (β=-0.230), and age was (β=-0.211). In addition, 56% of self-care variance is explained by age, social support, self-efficacy, and illness perception. Conclusion: All of the investigated variables were found to have a predictive role in self-care. Illness perception was recognized as the most effective factor in predicting self-care. Hence, illness perception can be used to explain 27% of self-care ability of patients with HF

    Causes of bimodal melting curve:Asymmetric guaninecytosine (GC) distribution causing two peaks in melting curve and affecting their shapes

    The aim of this study was to present a new situation in which a relatively single short PCR-product might show two separate peaks with sequence specific shapes at the dissociation curve. SYBR-Green I real-time RT-PCR was performed on Lhcgr-gene transcripts in rats. Different programs were used for melting curve simulation and estimating Tm. Statistical tests were performed to determine whether two peaks at the dissociation curve were belonging to a single template. A bimodal melting curve was observed in real-time RT-PCR on a short segment (169 bp) of Lhcgr gene with a single band in gel electrophoresis. Sequencing of the Cloned PCR-product was compatible with template sequence. Realtime PCR using the vector conveying interested sequence, showed again two peaks at dissociation curve. The GC-content of first 100 bases (75%) and last 69 bases (42%) were significantly different. DNA melting simulation programs also confirmed the bimodal pattern, although, their height and wideness were different to actual peaks. Due to the asymmetric GC distribution effect on dissociation curve in short sequences, it is highly recommended to use DNA melting simulation programs to predict the number of peaks in the melting curve when designating primers; however, predicted peak shapes are not always accurate.Key words: Asymmetric GC distribution, bimodal melting curve, DNA melting simulation, SYBR-green I realtime PCR

    Anti-Ulcerative Potential of Punica granatum L (Lythraceae) Hydroalcohol Fruit Peel Extract

    Purpose: To evaluate the antiulcer activities of the methanol extract of pomegranate (Punica granatum L) fruit peel using 80 % ethanol-induced gastric ulcer model in rats.Methods: Male Wistar rats, 175 - 220 g, were fasted for 48 h, but had free access to water. They were randomly divided into nine experimental groups. Ulcer was induced in the rats with 80 % ethanol. The control group animals received the same treatment as those in the test groups except that the extract treatment was replaced by administration of appropriate volume of the dosing vehicle. Histaminereceptor type-2 (H2) blocker, cimetidine (100 mg/kg, i.p.) was used as the reference drug. Oral pretreatment with three different extract doses (25, 50 and 100 mg/kg) was for 15 days, thereafter, ulcer index (UI) and inhibition were calculated.Results: The extract, at 50 mg/kg, of black fruit peel extract produced significant (p &lt; 0.05) protective effect in rats with a preventive index of 65.87 %. Other doses were significantly protective against ethanol-induced gastric ulcer in the rats. On the other hand, north white peel was not effective (50 mg/kg) showed an ulcer index of 49.52 ± 1.99. Histopathological examination of the stomach of the ulcerated animals treated with white peel (50 mg/kg) showed severe erosion of gastric mucosa, submucosal edema and neutrophil infiltration.Conclusion: The study shows indicates the antiulcer properties of the methanol extracts of north white peel, sour summer and black peel (25, 50, 100 mg/kg) of pomegranate.     Profiling of major fatty acids in different raw and roasted sesame seeds cultivars

    The aim of this study was to investigate the fatty acids profile of five sesame cultivars including Branching Naz, Darab, Karaj, Dezful and Black sesame and the effect of time and temperature roasting procedure. The seeds oil content varied from 43±0.28 to 47±0.41% with the average content of 44.4±1.87%. Darab and Black sesame cultivar had the highest and lowest oil content respectively. Oleic and linoleic acids were the two-dominant fatty acids in the sesame seed oil about 80 to 85% of the total amount, whereas palmitic and stearic acids were present at 12 to 15%. Moreover, Dazful and Black sesame had the maximum and minimum content of oxidizability value respectively. The results of the present study showed that the fatty acid contents in studied cultivars were steady during different roasting conditions and fatty acid behavior of samples was good fitted with the high temperatures.Key words: Gas chromatography (GC), fatty acid, roasting procedure, sesame seed, stability

    A stochastic hybrid algorithm for multi-depot and multi-product routing problem with heterogeneous vehicles

    Abstract. A mathematical model and heuristic method for solving multi-depot and multi-product vehicle routing problem (MD-MPVRP) with heterogeneous vehicles have been proposed in this article. Customers can order eclectic products and depots are supposed to deliver customers&apos; orders before the lead time, using vehicles with diverse capacities, costs and velocities. Hence, mathematical model of multi-depot vehicle routing problem has been developed to mirror these conditions. This model is aimed at minimizing the serving distances which culminates in a reduction in prices and also serving time. As the problem is so complex and also solving would be too time-taking, a heuristic method has been offered. The heuristic method, at first, generates an initial solution through a three-step procedure which encompasses grouping, routing and vehicle selection, scheduling and packaging. Then it improves the solution by means of simulated annealing.     Credit card fraud detection using asexual reproduction optimization

    Purpose – The best algorithm that was implemented on this Brazilian dataset was artificial immune system (AIS) algorithm. But the time and cost of this algorithm are high. Using asexual reproduction optimization (ARO) algorithm, the authors achieved better results in less time. So the authors achieved less cost in a shorter time. Their framework addressed the problems such as high costs and training time in credit card fraud detection. This simple and effective approach has achieved better results than the best techniques implemented on our dataset so far. The purpose of this paper is to detect credit card fraud using ARO. Design/methodology/approach – In this paper, the authors used ARO algorithm to classify the bank transactions into fraud and legitimate. ARO is taken from asexual reproduction. Asexual reproduction refers to a kind of production in which one parent produces offspring identical to herself. In ARO algorithm, an individual is shown by a vector of variables. Each variable is considered as a chromosome. A binary string represents a chromosome consisted of genes. It is supposed that every generated answer exists in the environment, and because of limited resources, only the best solution can remain alive. The algorithm starts with a random individual in the answer scope. This parent reproduces the offspring named bud. Either the parent or the offspring can survive. In this competition, the one which outperforms in fitness function remains alive. If the offspring has suitable performance,it will be the next parent, and the current parent becomes obsolete.Otherwise, the offspring perishes, and the present parent survives. The algorithm recurs until the stop condition occurs. Findings – Results showed that ARO had increased the AUC (i.e. area under a receiver operating characteristic (ROC) curve), sensitivity, precision, specificity and accuracy by 13%, 25%, 56%, 3% and 3%, in comparison with AIS, respectively. The authors achieved a high precision value indicating that if ARO detects a record as a fraud, with a high probability, it is a fraud one. Supporting a real-time fraud detection system is another vital issue. ARO outperforms AIS not only in the mentioned criteria, but also decreases the training time by 75% in comparison with the AIS, which is a significant figure. Originality/value – In this paper, the authors implemented the ARO in credit card fraud detection. The authors compared the results with those of the AIS, which was one of the best methods ever implemented on the benchmark dataset. The chief focus of the fraud detection studies is finding the algorithms that can detect legal transactions from the fraudulent ones with high detection accuracy in the shortest time and at a low cost.
