    Size Evolution of Ordered SiGe Islands Grown by Surface Thermal Diffusion on Pit-Patterned Si(100) Surface

    The ordered growth of self-assembled SiGe islands by surface thermal diffusion in ultra high vacuum from a lithographically etched Ge stripe on pit-patterned Si(100) surface has been experimentally investigated. The total surface coverage of Ge strongly depends on the distance from the source stripe, as quantitatively verified by Scanning Auger Microscopy. The size distribution of the islands as a function of the Ge coverage has been studied by coupling atomic force microscopy scans with Auger spectro-microscopy data. Our observations are consistent with a physical scenario where island positioning is essentially driven by energetic factors, which predominate with respect to the local kinetics of diffusion, and the growth evolution mainly depends on the local density of Ge atoms

    Identification and management of chronic pain in primary care:a review

    Chronic pain is a common, complex, and challenging condition, where understanding the biological, social, physical and psychological contexts is vital to successful outcomes in primary care. In managing chronic pain the focus is often on promoting rehabilitation and maximizing quality of life rather than achieving cure. Recent screening tools and brief intervention techniques can be effective in helping clinicians identify, stratify and manage both patients already living with chronic pain and those who are at risk of developing chronic pain from acute pain. Frequent assessment and reassessment are key to ensuring treatment is appropriate and safe, as well as minimizing and addressing side effects. Primary care management should be holistic and evidence-based (where possible) and incorporates both pharmacological and non-pharmacological approaches, including psychology, self-management, physiotherapy, peripheral nervous system stimulation, complementary therapies and comprehensive pain-management programmes. These may either be based wholly in primary care or supported by appropriate specialist referral

    Quantifying the Proteolytic Release of Extracellular Matrix-Sequestered VEGF with a Computational Model

    BACKGROUND: VEGF proteolysis by plasmin or matrix metalloproteinases (MMPs) is believed to play an important role in regulating vascular patterning in vivo by releasing VEGF from the extracellular matrix (ECM). However, a quantitative understanding of the kinetics of VEGF cleavage and the efficiency of cell-mediated VEGF release is currently lacking. To address these uncertainties, we develop a molecular-detailed quantitative model of VEGF proteolysis, used here in the context of an endothelial sprout. METHODOLOGY AND FINDINGS: To study a cell's ability to cleave VEGF, the model captures MMP secretion, VEGF-ECM binding, VEGF proteolysis from VEGF165 to VEGF114 (the expected MMP cleavage product of VEGF165) and VEGF receptor-mediated recapture. Using experimental data, we estimated the effective bimolecular rate constant of VEGF165 cleavage by plasmin to be 328 M(-1) s(-1) at 25 degrees C, which is relatively slow compared to typical MMP-ECM proteolysis reactions. While previous studies have implicated cellular proteolysis in growth factor processing, we show that single cells do not individually have the capacity to cleave VEGF to any appreciable extent (less than 0.1% conversion). In addition, we find that a tip cell's receptor system will not efficiently recapture the cleaved VEGF due to an inability of cleaved VEGF to associate with Neuropilin-1. CONCLUSIONS: Overall, VEGF165 cleavage in vivo is likely to be mediated by the combined effect of numerous cells, instead of behaving in a single-cell-directed, autocrine manner. We show that heparan sulfate proteoglycans (HSPGs) potentiate VEGF cleavage by increasing the VEGF clearance time in tissues. In addition, we find that the VEGF-HSPG complex is more sensitive to proteases than is soluble VEGF, which may imply its potential relevance in receptor signaling. Finally, according to our calculations, experimentally measured soluble protease levels are approximately two orders of magnitude lower than that needed to reconcile levels of VEGF cleavage seen in pathological situations

    Model-based geostatistical mapping of the prevalence of onchocerca volvulus in West Africa.

    Background: The initial endemicity (pre-control prevalence) of onchocerciasis has been shown to be an important determinant of the feasibility of elimination by mass ivermectin distribution. We present the first geostatistical map of microfilarial prevalence in the former Onchocerciasis Control Programme in West Africa (OCP) before commencement of antivectorial and antiparasitic interventions. Methods and Findings: Pre-control microfilarial prevalence data from 737 villages across the 11 constituent countries in the OCP epidemiological database were used as ground-truth data. These 737 data points, plus a set of statistically selected environmental covariates, were used in a Bayesian model-based geostatistical (B-MBG) approach to generate a continuous surface (at pixel resolution of 5 km x 5km) of microfilarial prevalence in West Africa prior to the commencement of the OCP. Uncertainty in model predictions was measured using a suite of validation statistics, performed on bootstrap samples of held-out validation data. The mean Pearson’s correlation between observed and estimated prevalence at validation locations was 0.693; the mean prediction error (average difference between observed and estimated values) was 0.77%, and the mean absolute prediction error (average magnitude of difference between observed and estimated values) was 12.2%. Within OCP boundaries, 17.8 million people were deemed to have been at risk, 7.55 million to have been infected, and mean microfilarial prevalence to have been 45% (range: 2–90%) in 1975. Conclusions and Significance: This is the first map of initial onchocerciasis prevalence in West Africa using B-MBG. Important environmental predictors of infection prevalence were identified and used in a model out-performing those without spatial random effects or environmental covariates. Results may be compared with recent epidemiological mapping efforts to find areas of persisting transmission. These methods may be extended to areas where data are sparse, and may be used to help inform the feasibility of elimination with current and novel tools

    Neglected zoonotic diseases (NZDs) have a significant impact on the livelihoods of the world’s poorest populations, which often lack access to basic services. Water, sanitation and hygiene (WASH) programmes are included among the key strategies for achieving the World Health Organization’s 2020 Roadmap for Implementation for control of Neglected Tropical Diseases (NTDs). There exists a lack of knowledge regarding the effect of animals on the effectiveness of WASH measures. This review looked to identify how animal presence in the household influences the effectiveness of water, hygiene and sanitation measures for zoonotic disease control in low and middle income countries; to identify gaps of knowledge regarding this topic based on the amount and type of studies looking at this particular interaction

    Shell morphological characters of Brachidontes danllinialzus and B. solisianus were investigated in mussels collected in six areas of sympatry in southeast Brazil. Individuals were measured and analyzed for the relation among shell length, height, and width, as well as other systematic characters used to distinguish the two species. All allometric relations varied according to the site sampled, except the height:length ratio, which consistently separated the two species regardless of where they occurred. Values for this ratio change in distinct shell length classes, and 99% confidence intervals are provided for each length class. Internal and external shell characteristics generally used to separate these species varied both during ontogeny and according to environment. Thus, caution is needed when identifying these species, as single characters may not be useful as taxonomic predictors. Combinations of characters are necessary to separate B. darwinianus and B. solisianus with more confidence.42326727

    Dynamics of the acmaeid limpet Collisella subrugosa and vertical distribution of size and abundance along a wave exposure gradient

    The vertical distribution of the acmaeid limpet Collisella subrugosa (Orbigny) was studied on subtropical Brazilian shores. In a semi-sheltered shore at Ubatuba, monthly monitoring indicated that densities were higher in the mid-intertidal, and lower in the high- and low-intertidal. There was evidence of continuous recruitment mainly to mid- and lower levels on the shore, with a peak in the summer. Shell length increased with tidal height, and this pattern was maintained during the study period. To verify whether these results were consistent over a larger spatial scale, the vertical distribution of size, abundance, and shell shape of C. subrugosa was studied along a wave exposure gradient. Densities were not clearly related to wave action, and other sources of variation such as presence of competitors or secondary substrata appear to influence C. subrugosa distribution. Shell length increased towards the upper intertidal on more exposed shores, but no differences, or the reverse pattern was recorded on sheltered shores. These results are consistent with other studies on limpet distribution, and proposed causes include differential recruitment, migration, or microhabitat availability. Shell height increased faster with shell length in lower intertidal levels, and on more exposed sites. Foot surface area also increased faster with shell length on more exposed shores and indicate that wave action is an important factor determining shell shape in C. subrugosa. Additional sampling of upper and lower populations along a gradient of wave exposure in Florianopolis (southern Brazil) resulted in similar patterns of shell size, but the relationships between shell height and length for these populations were similar and isometric. Thus, there is a geographic variation in the vertical distribution of C. subrugosa, and possible causes include distinct physical conditions or differences in phenotypic plasticity of this limpet.681556
