In vitro activities of MCB3681 and 8 comparators against Clostridium difficile isolates with known ribotypes and diverse geographical spread
Treatments for Clostridium difficile infection remain limited, despite the introduction of fidaxomicin, and development of new agents is necessary. We determined the in vitro susceptibilities of 199 prevalent or emerging Clostridium difficile PCR ribotypes to MCB3681, a novel investigational quinolonyl-oxazolidinone, and 8 comparators (metronidazole, vancomycin, fidaxomicin, moxifloxacin, ciprofloxacin, clindamycin, tigecycline and linezolid). MCB3681 showed good activity against C. difficile with no evidence of MCB3681 resistance in isolates showing either or both moxifloxacin and linezolid resistance
Susceptibility of Clostridium difficile isolates of varying antimicrobial resistance phenotypes to SMT19969 and 11 comparators.
We determined the in vitro activity of SMT19969 and 11 comparators, including metronidazole, vancomycin and fidaxomicin, against 107 C. difficile isolates of different antimicrobial resistance phenotypes. Fidaxomicin and SMT19969 were most active. Geometric mean fidaxomicin and SMT19969 MICs were highest in ribotypes known to show multiple resistance. Co-resistance to linezolid and moxifloxacin was evident in ribotypes 001, 017, 027 and 356. High-level ceftriaxone resistance in ribotypes 356 and 018 was location-linked
The Citation Field of Evolutionary Economics
Evolutionary economics has developed into an academic field of its own,
institutionalized around, amongst others, the Journal of Evolutionary Economics
(JEE). This paper analyzes the way and extent to which evolutionary economics
has become an interdisciplinary journal, as its aim was: a journal that is
indispensable in the exchange of expert knowledge on topics and using
approaches that relate naturally with it. Analyzing citation data for the
relevant academic field for the Journal of Evolutionary Economics, we use
insights from scientometrics and social network analysis to find that, indeed,
the JEE is a central player in this interdisciplinary field aiming mostly at
understanding technological and regional dynamics. It does not, however, link
firmly with the natural sciences (including biology) nor to management
sciences, entrepreneurship, and organization studies. Another journal that
could be perceived to have evolutionary acumen, the Journal of Economic Issues,
does relate to heterodox economics journals and is relatively more involved in
discussing issues of firm and industry organization. The JEE seems most keen to
develop theoretical insights
Weatherability and Leach Resistance of Wood Impregnated with Nano-Zinc Oxide
Southern pine specimens vacuum-treated with nano-zinc oxide (nano-ZnO) dispersions were evaluated for leach resistance and UV protection. Virtually, no leaching occurred in any of the nano-ZnO–treated specimens in a laboratory leach test, even at the highest retention of 13 kg/m3. However, specimens treated with high concentrations of nano-ZnO showed 58–65% chemical depletion after 12 months of outdoor exposure. Protection from UV damage after 12 months exposure is visibly obvious on both exposed and unexposed surfaces compared to untreated controls. Graying was markedly diminished, although checking occurred in all specimens. Nano-zinc oxide treatment at a concentration of 2.5% or greater provided substantial resistance to water absorption following 12 months of outdoor exposure compared to untreated and unweathered southern pine. We conclude that nano-zinc oxide can be utilized in new wood preservative formulations to impart resistance to leaching, water absorption and UV damage of wood
Tourette’s disorder and other tic disorders in DSM-5: a comment
Classification of tic disorders will be revised in the forthcoming edition of the Diagnostic and Statistical Manual of Mental Disorders (5th ed.; DSM-5). We do not support the suggestion to move tic disorders to “Anxiety and Obsessive–Compulsive Disorders”, if the section “Disorders Usually First Diagnosed in Infancy, Childhood, or Adolescence” is not retained. Other than that, most proposed changes of the criteria for tic disorders contain a number of welcome improvements, e.g., the more unified definition of tics including the removal of the term “stereotyped” and the better capture of the temporal pattern of tics (e.g., removal of the maximum 3 months criterion for a tic-free period in chronic tic disorders). But, unfortunately there are some inconsistencies in detail, e.g., the unification of diagnostic criteria for tic disorders had not been consistently pursued in transient tic disorder. In sum, the proposed DSM-5 criteria could be seen as an important step forward particularly in clinical routine. However, continued research is needed to justify the existing and proposed classification of tic disorders as well as to better clarify what other changes should be made in the DSM-5 and beyond
Isolation and detection of circulating tumour cells from metastatic melanoma patients using a slanted spiral microfluidic device.
Circulating Tumour Cells (CTCs) are promising cancer biomarkers. Several methods have been developed to isolate CTCs from blood samples. However, the isolation of melanoma CTCs is very challenging as a result of their extraordinary heterogeneity, which has hindered their biological and clinical study. Thus, methods that isolate CTCs based on their physical properties, rather than surface marker expression, such as microfluidic devices, are greatly needed in melanoma. Here, we assessed the ability of the slanted spiral microfluidic device to isolate melanoma CTCs via label-free enrichment. We demonstrated that this device yields recovery rates of spiked melanoma cells of over 80% and 55%, after one or two rounds of enrichment, respectively. Concurrently, a two to three log reduction of white blood cells was achieved with one or two rounds of enrichment, respectively. We characterised the isolated CTCs using multimarker flow cytometry, immunocytochemistry and gene expression. The results demonstrated that CTCs from metastatic melanoma patients were highly heterogeneous and commonly expressed stem-like markers such as PAX3 and ABCB5. The implementation of the slanted microfluidic device for melanoma CTC isolation enables further understanding of the biology of melanoma metastasis for biomarker development and to inform future treatment approaches
Bang-bang control of fullerene qubits using ultra-fast phase gates
Quantum mechanics permits an entity, such as an atom, to exist in a
superposition of multiple states simultaneously. Quantum information processing
(QIP) harnesses this profound phenomenon to manipulate information in radically
new ways. A fundamental challenge in all QIP technologies is the corruption of
superposition in a quantum bit (qubit) through interaction with its
environment. Quantum bang-bang control provides a solution by repeatedly
applying `kicks' to a qubit, thus disrupting an environmental interaction.
However, the speed and precision required for the kick operations has presented
an obstacle to experimental realization. Here we demonstrate a phase gate of
unprecedented speed on a nuclear spin qubit in a fullerene molecule (N@C60),
and use it to bang-bang decouple the qubit from a strong environmental
interaction. We can thus trap the qubit in closed cycles on the Bloch sphere,
or lock it in a given state for an arbitrary period. Our procedure uses
operations on a second qubit, an electron spin, in order to generate an
arbitrary phase on the nuclear qubit. We anticipate the approach will be vital
for QIP technologies, especially at the molecular scale where other strategies,
such as electrode switching, are unfeasible
The ClosER study: results from a three-year pan-European longitudinal surveillance of antibiotic resistance among prevalent Clostridium difficile ribotypes, 2011–2014
Objectives: Until the introduction of fidaxomicin, antimicrobial treatment for Clostridium difficile infection (CDI) was limited to metronidazole and vancomycin. The changing epidemiology of CDI and the emergence of epidemic C. difficile PCR ribotype 027 necessitate continued surveillance to identify shifts in antibiotic susceptibility. ClosER, currently the largest pan-European epidemiological study of C. difficile ribotype distribution and antibiotic susceptibility, aimed to undertake antimicrobial resistance surveillance pre- and post-introduction of fidaxomicin.
Methods: Between July 2011 and July 2014, 39 sites across 22 European countries submitted 2830 C. difficile isolates for ribotyping, toxin testing and susceptibility testing to metronidazole, vancomycin, fidaxomicin, rifampicin, moxifloxacin, clindamycin, imipenem, chloramphenicol and tigecycline.
Results: Ribotypes 027, 014, 001, 078, 020, 002, 126, 015 and 005 were most frequently isolated, and emergent ribotypes 198 and 356 were identified in Hungary and Italy, respectively. All isolates were susceptible to fidaxomicin, with scarce resistance to metronidazole (0.2%, 6/2694), vancomycin (0.1%, 2/2694) and tigecycline (0%). Rifampicin, moxifloxacin and clindamycin resistance was evident in multiple ribotypes. Lack of ribotype diversity correlated with greater antimicrobial resistance. Epidemic ribotypes (027/001) were associated with multiple antimicrobial resistance, and ribotypes 017, 018 and 356 with high-level resistance. Additional factors may also influence local ribotype prevalence.
Conclusions: Fidaxomicin susceptibility was retained post-introduction, and resistance to metronidazole and vancomycin was rare. Continued surveillance is needed, with more accurate classification and clarification of ribotype subtypes to further understand their role in the spread of resistance. Other factors may also influence changes in prevalence of C. difficile ribotypes with reduced antibiotic susceptibility
