    Lack of Phylogeographic Structure in the Freshwater Cyanobacterium Microcystis aeruginosa Suggests Global Dispersal

    Background : Free-living microorganisms have long been assumed to have ubiquitous distributions with little biogeographic signature because they typically exhibit high dispersal potential and large population sizes. However, molecular data provide contrasting results and it is far from clear to what extent dispersal limitation determines geographic structuring of microbial populations. We aimed to determine biogeographical patterns of the bloom-forming freshwater cyanobacterium Microcystis aeruginosa. Being widely distributed on a global scale but patchily on a regional scale, this prokaryote is an ideal model organism to study microbial dispersal and biogeography. Methodology/Principal Findings : The phylogeography of M. aeruginosa was studied based on a dataset of 311 rDNA internal transcribed spacer (ITS) sequences sampled from six continents. Richness of ITS sequences was high (239 ITS types were detected). Genetic divergence among ITS types averaged 4% (maximum pairwise divergence was 13%). Preliminary analyses revealed nearly completely unresolved phylogenetic relationships and a lack of genetic structure among all sequences due to extensive homoplasy at multiple hypervariable sites. After correcting for this, still no clear phylogeographic structure was detected, and no pattern of isolation by distance was found on a global scale. Concomitantly, genetic differentiation among continents was marginal, whereas variation within continents was high and was mostly shared with all other continents. Similarly, no genetic structure across climate zones was detected. Conclusions/Significance : The high overall diversity and wide global distribution of common ITS types in combination with the lack of phylogeographic structure suggest that intercontinental dispersal of M. aeruginosa ITS types is not rare, and that this species might have a truly cosmopolitan distribution

    Use of local cues in the night-time navigation of the wandering desert spider Leucorchestris arenicola (Araneae, Sparassidae)

    Adult male Leucorchestris arenicola can walk round-trips of several tens of meters in search of females. Most excursions end with the spiders returning to their burrow. For small animals homing over distances of several meters is theoretically impossible without the aid of external cues. It was investigated, whether the spiders use local cues or they rely solely on global cues. Individually marked male spiders were captured during their excursions and displaced several meters inside an opaque box. Ten out of twelve displaced spiders returned to their burrows. This shows that the male L. arenicola are using local cues during their homing, as the comparatively small displacement distances could not be detected by means of global, e.g. celestial cues. In order to test whether the spiders could be using olfactory guidance, the burrows were displaced by 2 m while the spiders were out on their journeys. In 12 out of 15 experiments, the spiders did not Wnd their burrows. These results show that the burrows do not function as olfactory beacons for the homing spiders

    Bias in the heritability of preference and its potential impact on the evolution of mate choice

    The evolution of mate choice is a function of the heritability of preference. Estimation in the laboratory is typically made by presenting a female with a limited number of males. We show that such an approach produces a downwardly biased estimate, which we term the heritability of choice. When preference is treated as a threshold trait then less biased estimates are obtained particularly for preferences based on the relative value of the preferred trait. Because females in the wild typically survey on average less than five males we argue that the heritability of choice may be more meaningful than the heritability of preference. The restricted number of males surveyed can lead to a reduction in the phenotypic variance of the preferred trait in the group of males selected by the females if the phenotypic variance in preference is equal to or less than the phenotypic variance in the referred trait. If the phenotypic variance in preference exceeds that of the preferred trait then the opposite occurs. A second effect of the restricted number of males sampled is that females are likely to mate initially with males that are not the most preferred. The failure to find the most preferred male may account for the common observation of multiple matings and extra-pair copulations. We suggest that current explanations for polyandry need to take this failure into account

    The Necessity of α4* Nicotinic Receptors in Nicotine-Driven Behaviors: Dissociation Between Reinforcing and Motor Effects of Nicotine

    Here we utilize a mouse line with a targeted deletion of the α4 subunit (α4−/− mice), to investigate the role of α4* nAChRs in reinforcing and locomotor effects of nicotine. Within a conditioned place preference paradigm, both α4−/− mice and wild-type (WT) littermates showed a similar place preference to nicotine (0.5 mg/kg i.p.) conditioning. When assessed for operant intravenous self-administration of nicotine (0.05 mg/kg/infusion), α4−/− mice did not differ from their WT littermates in self-administration behavior. To further examine a modulatory role for α4* nAChRs in the reinforcing effects of nicotine, a transgenic mouse with a point mutation of the α4 subunit (α4-S248F) that renders increased sensitivity to low dose nicotine, was assessed for nicotine self-administration over a range of doses. At higher doses examined (0.05 and 0.07 mg/kg/infusion) there was no difference in intravenous nicotine self-administration; however, when mice were offered a lower dose of nicotine (0.03 mg/kg/infusion), α4-S248F mice showed greater nicotine intake than controls. Acute administration of 0.5 mg/kg nicotine caused significant locomotor depression in WT mice but α4−/− mice instead showed significant hyperactivity. Following chronic, intermittent administration of this dose of nicotine only WT mice displayed significant tolerance. Analogous experiments utilizing administration of the nicotinic antagonist mecamylamine in WT mice confirmed a dissociation between the putative nicotinic receptor subtypes required for mediating psychomotor and reinforcing effects of nicotine. These data demonstrate a necessary role for α4* nAChRs in the locomotor depressant effect of nicotine but not the reinforcing effects that support ongoing self-administration of nicotine
