Fatores de virulência de Escherichia coli uropatogênicas provenientes de um Hospital Universitário em Ribeirão Preto, São Paulo, Brasil
The aim of the study was to determine the occurrence of virulence genes expressing fimbriae, production of hemolysin, colicin and aerobactin among a hundred Escherichia coli isolates obtained from in-and outpatients of a tertiary-care teaching hospital, between July and August 2000, showing clinical and laboratory signs of urinary tract infection (UTI). The presence of genes (pap, afa, sfa) for fimbriae expression was assayed using specific primers in a polymerase chain reaction. Among the isolates studied, the prevalence of the virulence factors was 96.0%, 76.0%, 24.0%, for hemolysin, aerobactin and colicin, respectively; the prevalence of genes coding for fimbrial adhesive systems was 32.0%, 19.0% and 11.0% for pap, sfa and afa respectively. The strains isolated from the outpatients displayed a greater number of virulence factors compared to those from hospitalized subjects, emphasizing the difference between these two kinds of patients.O objetivo do trabalho foi determinar a ocorrência de fatores de virulência, tais como, a expressão de fímbrias, produção de hemolisina, colicina e aerobactina em 100 cepas de Escherichia coli isoladas de pacientes ambulatoriais e hospitalizados de um hospital universitário de nível de atendimento terciário, entre os meses de julho e agosto de 2000, que apresentavam sinais clínicos e laboratoriais de infecção do trato urinário (ITU). Foram pesquisados os genes pap, afa e sfa responsáveis pela expressão de fímbrias através da técnica de PCR. A freqüência dos fatores de virulência entre as cepas estudadas foi de 96,0%, 76,0% e 24,0% para hemolisina, aerobactina e colicina respectivamente, e a prevalência dos genes para os sistemas de adesinas fimbriais foi de 32,0%, 19,0% e 11,0% para os genes pap, sfa e afa respectivamente. As cepas isoladas dos pacientes ambulatoriais exibiram um número maior de fatores de virulência quando comparadas com aquelas provenientes de indivíduos hospitalizados
Ocorrência de Escherichia coli não-O157 Shigatoxigênica em cachorros com diarréia
Shiga toxigenic Escherichia coli (STEC) and Attaching and effacing E. coli (AEEC) have been associated with diarrhea illness in dogs. From January to December 2006, 92 E. coli isolates from 25 diarrheic dogs were analyzed, by screening for the presence of Shiga toxin-producing (stx 1 and stx 2) and intimin (eae) genes. Twelve isolates were detected by PCR to harbor the Shiga toxin genes (7 the stx 1 (7.6%); 5 the stx 2 (5.4%); and none both of them). Nine (9.8%) of the E. coli isolates studied were eae positive non Shiga toxin-producing. Thirteen (62.0%) isolates, carrying stx or eae gene, also showed a hemolysin production. The strains with virulence genes were also examined for resistance to 12 antimicrobial agents. Resistances to cephalothin (85.7%), streptomycin (81.0%), amoxicillin (71.4%) and gentamicin (71.4%) were predominantly observed.Escherichia coli Shiga toxigênica (STEC) e E. coli Attaching- effacing (AEEC) têm sido associadas à doença diarréica em cachorros. Entre janeiro e dezembro de 2006, 92 cepas de E. coli isoladas de 25 cachorros diarréicos foram examinadas. As cepas foram analisadas para a detecção dos genes produtores de Shiga toxina (stx 1 e stx 2) e da intimina (eae). Por meio de PCR foi observado que sete cepas (7,6%) portavam o gene stx 1, cinco cepas (5,4%) carregavam o gene stx 2 e nenhum cepa apresentou ambos os genes associados. Nove cepas de E. coli (9,8%) apresentaram o gene eae isoladamente. Treze das cepas (62,0%) que apresentaram os genes stx ou eae também apresentaram a produção de a hemolisina. As cepas que apresentaram genes de virulência foram também examinadas em relação à resistência a 12 agentes antimicrobianos. As resistências mais comuns foram para cefalotina (85,7%), estreptomicina (81,0%), amoxicilina (71,4%) e gentamicina (71,4%)
Prevalence of mutans streptococci in 93 members from six Brazilian families
Vários estudos relatam que os estreptococos do grupo mutans (SGM) estão associados à cárie e que existe uma correlação entre o número de lesões de cárie e de SGM na saliva de crianças e adultos. Foi avaliada a presença de SGM na saliva de 93 membros de seis famílias brasileiras com no mínimo três gerações. Amostras de saliva não estimulada foram coletadas e diluídas. Alíquotas de 50 mil de cada suspensão foram gotejadas em ágar SB20 e incubadas em jarras de anaerobiose a 37ºC por 72 horas. As colônias com características de SGM foram contadas e repicadas em caldo tioglicolato e submetidas a biotipagem. Os estreptococos do grupo mutans foram isolados de 80 (86,0 %) membros e as contagens na saliva variaram de 3,0 x 10² (log 2,477) a 1,6 x 10(8) (log 8,204) CFU/ml. Todos os 73 adultos estavam colonizados com SGM, mas a bactéria foi detectada apenas em 7 (35,0%) das 20 crianças. Streptococcus mutans foi identificado em 78 (97,5%) indivíduos, e 51 (63,7%) eram monocolonizados. S. sobrinus foi isolado de 29 (36,3%) membros e 2 (2,5%) estavam monocolonizados. Vinte e sete (33,8%) indivíduos estavam multicolonizados com S. mutans e S. sobrinus. O estudo mostrou uma elevada prevalência (86,0%) de estreptococos do grupo mutans na saliva dos membros das famílias sugerindo o risco de transmissão intrafamilial.Several studies report that mutans streptococci (MS) are closely associated with caries in humans and that there is a correlation between the number of carious lesions and the levels of MS in the saliva of children and adults. The presence of MS in the saliva of 93 members of six Brazilian families with at least 3 generations was investigated. Samples of whole unstimulated saliva were collected and diluted. Aliquots of 50 mul of each suspension were dropped onto SB20 agar and incubated in a candle jar at 37°C for 72h. Colonies resembling MS were counted, collected, seeded in thioglycollate medium and subjected to biochemical typing. Mutans streptococci were isolated from 80 subjects (86.0%) and the counts ranged from 3.0 x 10² (log 2.477) to 1.6 x 10(8) (log 8.204) CFU/ml of saliva. All of the 73 adults were colonized by MS, but the bacteria were detected in only 7 (35.0%) of the 20 children evaluated. Streptococcus mutans occurred in 78 subjects (97.5%), and 51 (63.7%) were monocolonized. S. sobrinus occurred in 29 individuals (36.3%) and 2 (2.5%) were monocolonized. Twenty-seven (33.8%) subjects were multicolonized with S. mutans and S. sobrinus. This study showed a high prevalence (86.0%) of mutans streptococci in the saliva of members of the studied families, which suggests the risk of intrafamilial transmission
Phenotypical characterization and adhesin identification in Escherichia coli strains isolated from dogs with urinary tract infections
Pathogenic strains of Escherichia coli are the most common bacteria associated with urinary tract infections in both humans and companion animals. Standard biochemical tests may be useful in demonstrating detailed phenotypical characteristics of these strains. Thirteen strains of E. coli isolated from dogs with UTIs were submitted to biochemical tests, serotyping for O and H antigens and antimicrobial resistance testing. Furthermore, the presence of papC, sfa, and afa genes was evaluated by PCR, and genetic relationships were established using enterobacterial repetitive intergenic consensus PCR (ERIC-PCR). The antimicrobial that showed the highest resistance rate among the isolates was nalidixic acid (76.9%), followed by cephalotin (69.2%), sulfamethoxazole + trimethoprim (61.5%), tetracycline (61.5%), streptomycin (53.8%), ciprofloxacin (53.8%), ampicillin (46.2%), gentamicin (30.8%) and chloramphenicol (23.1%). No isolate was resistant either to meropenem or nitrofurantoin. Among the five clusters that were identified using ERIC-PCR, one cluster (A) had only one strain, which belonged to a serotype with zoonotic potential (O6:H31) and showed the genes papC+, sfa+, afa-. Strains with the genes papC-, sfa+, afa- were found in two other clusters (C and D), whereas all strains in clusters B and E possessed papC-, sfa-, afa- genes. Sucrose and raffinose phenotypic tests showed some ability in discriminating clusters A, B and C from clusters D and E
Isolation of Shiga toxigenic Escherichia coli from butcheries in Taquaritinga city, State of São Paulo, Brazil
Escherichia coli Shiga toxigênica (STEC) tem sido responsabilizada como o agente etiológico de diversas doenças nos seres humanos. Neste estudo foram analisadas amostras provenientes de 23 açougues (carne moída, moedor de carne e mãos de manipuladores) visando o isolamento de cepas de E. coli utilizando os métodos microbiológicos tradicionais. Um total de 287 cepas de E. coli, isoladas destas amostras, foram submetidas a reação em cadeia da polimerase para a detecção dos genes stx 1, stx 2 e eae. Foram identificadas 4 cepas STEC, 2 provenientes de carne moída e 2 provenientes do moedor de carne, todas as cepas apresentando o gene stx 2 e negativas para a presença do gene eae. Todas as cepas de E. coli, incluindo as 4 cepas STEC, foram examinadas para verificar a resistência antimicrobiana. Foram detectados altos níveis de resistência a diferentes agentes antimicrobianos, sendo as resistências mais elevadas para a tetraciclina (76,6%), amoxicilina (64,1%) e cefalotina (58,8%). Estes altos índices de resistência ressaltam a necessidade de uma utilização mais racional destes agentes no gado bovino.Shiga toxigenic Escherichia coli (STEC) has been implicated as the cause of several human diseases. Samples (ground beef, grinding-machines and human hands) from 23 butcheries were assayed for E. coli using standard microbiological methods, and 287 isolates were submitted to polymerase chain reaction for the detection of stx 1, stx 2 and eae genes. Four STEC isolates were recovered, two from ground beef and two from grinding-machines; all harbored the stx 2 gene and were negative for the eae gene. All E. coli isolates including the four STEC were screened for antibiotic resistance. High levels of resistance against several antimicrobial agents were detected; those most commonly observed were to tetracycline (76.6%), amoxicillin (64.1%) and cephalothin (58.8%).
