    Antimicrobial resistance of isolates of microorganisms identified in the milk of cows with subclinical mastitis in the ural region of Russia

    During 2017 n = 375 milk samples of dairy cows with subclinical mastitis from 18 farms of the Ural Region were examined in order to identify the main pathogenic agents and their antimicrobial resistance. Microbiological research on the milk using real time PCR using the device Rotor Gene-3000 (Australia), with the complex of reagents of the IDS company (Russia) was done, to identify DNA from biological material and determine antimicrobial resistance of microorganisms. The identified microorganisms included: Staphylococcus spp. –77.3%, E. coli – 37.3%, S. aureus – 38.7%, S. agalactiae – 14.7%. Associations of microorganisms included: S. aureus, Staphylococcus spp. – 17.3%; Staphylococcus spp., E. coli – 21.3%; S. agalactiae, E. coli – 2.7%; St. aureus, Staphylococcus spp., E. coli – 8.0%; S. aureus, Staphylococcus spp., S. agalactiae – 2.7%; S. aureus, Staphylococcus spp., S. agalactiae, E. coli – 1.3%. The gene of CTX-M that determines resistance of Enterobacteriaceae to Cephalosporin of the 1st generation was found in 3.6% of samples with E. coli. The gene of blaDHA that determines resistance of Enterobacteriaceae to penicillin and cephalosporin of the 3rd and 4th generations was found in 4.6% samples with E. coli.. 3.6% of the samples with E. coli had two mutational genes CTX-M and blaDHA. The gene MecA that determines resistance of S. aureus to Cephalosporin of the 2nd generation was found in 10.6% samples. Resistance of Staphylococcus spp. and Streptococcus spp. to macrolides of the 1st generation (the gene of ErmB) was identified in 45.3% samples. In conclusion, the research done proved the wide spread of antimicrobial resistance of pathogenic agents causing subclinical mastitis in dairy cows

    Mechanisms of power distribution: principles of kinematic and force analysis

    The article considers the features of performing kinematic and power analysis of a controlled power mechanism for the transmission of wheeled transport and transport-technological machines. Such mechanisms are currently used mainly on cars in order to improve handling, stability, traction and dynamic properties in severe road conditions. Such mechanisms are not produced in Russia, but theoretical studies are currently being conducted, the results of which will allow determining the main parameters of the mechanism and synthesizing its kinematic scheme. The proposed approaches are based on the methods used in the kinematic and force analysis of planetary gears, but it is taken into account that the mechanism of power distribution during operation has two degrees of freedom. It is indicated that there is a structural and functional similarity between the power distribution mechanism of the car and the turning mechanism of the tracked vehicle. A method is proposed for calculating the gear ratio of the power distribution mechanism according to the condition of matching the minimum possible turning radius provided by the steering trapezoid kinematics and the calculated turning radius

    Antiproliferative, proapoptotic, and tumor-suppressing effects of the novel anticancer agent alsevirone in prostate cancer cells and xenografts

    The aim of this study was to explore the mechanisms of action of alsevirone in prostate cancer (PC) in vitro and in vivo: CYP17A1 inhibition, cytotoxic, apoptotic, and antitumor effects in comparison with abiraterone. The CYP17A1-inhibitory activity was investigated in rat testicular microsomes using high-performance liquid chromatography. Testosterone levels were evaluated using enzyme-linked immunoassay. IC50 values were calculated for PC3, DU-145, LNCaP, and 22Rv1 cells using the MTT (3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide) test. The antitumor effect in vivo was studied in DU-145 and 22Rv1 subcutaneous xenografts in Balb/c nude mice. Alsevirone reduced the CYP17A1-inhibitory activity by 98% ± 0.2%. A statistically significant reduction in the testosterone concentration in murine blood was recorded after the 7th administration of 300 mg/kg alsevirone at 0.31 ± 0.03 ng/ml (p <.001) versus 0.98 ± 0.22 ng/ml (p =.392) after abiraterone administration and 1.52 ± 0.49 ng/ml in control animals. Alsevirone was more cytotoxic than abiraterone in DU-145, LNCaP, and 22Rv1 cells, with IC50 values of 23.80 ± 1.18 versus 151.43 ± 23.70 μM, 22.87 ± 0.54 versus 28.80 ± 1.61 μM, and 35.86 ± 5.63 versus 109.87 ± 35.15 μM, respectively. Alsevirone and abiraterone significantly increased annexin V-positive, caspase 3/7-positive, and activated Bcl-2-positive cells. In 22Rv1 xenografts, alsevirone 300 mg/kg × 10/24 h per os inhibited tumor growth: on Day 9 of treatment, tumor growth inhibition = 59% (p =.022). Thus, alsevirone demonstrated significant antitumor activity associated with CYP17A1 inhibition, apoptosis in PC cells, and testosterone reduction. © 2021 Deutsche Pharmazeutische Gesellschaf
