    Fusion of secretory vesicles isolated from rat liver

    Secretory vesicles isolated from rat liver were found to fuse after exposure to Ca2+. Vescle fusion is characterized by the occurrence of twinned vesicles with a continuous cleavage plane between two vesicles in freeze-fracture electron microscopy. The number of fused vesicles increases with increasing Ca2+-concentrations and is half maximal around 10–6 m. Other divalent cations (Ba2+, Sr2+, and Mg2+) were ineffective. Mg2+ inhibits Ca2+-induced fusion. Therefore, the fusion of secretory vesiclesin vitro is Ca2+ specific and exhibits properties similar to the exocytotic process of various secretory cells. Various substances affecting secretionin vivo (microtubular inhibitors, local anethetics, ionophores) were tested for their effect on membrane fusion in our system. The fusion of isolated secretory vesicles from liver was found to differ from that of pure phospholipid membranes in its temperature dependence, in its much lower requirement for Ca2+, and in its Ca2+-specificity. Chemical and enzymatic modifications of the vesicle membrane indicate that glycoproteins may account for these differences

    The generalization of the Gross-Neveu model for noncommutative 3+1 space-time has been analyzed.     The outer halos of elliptical galaxies

    Recent progress is summarized on the determination of the density distributions of stars and dark matter, stellar kinematics, and stellar population properties, in the extended, low surface brightness halo regions of elliptical galaxies. With integral field absorption spectroscopy and with planetary nebulae as tracers, velocity dispersion and rotation profiles have been followed to ~4 and ~5-8 effective radii, respectively, and in M87 to the outer edge at ~150 kpc. The results are generally consistent with the known dichotomy of elliptical galaxy types, but some galaxies show more complex rotation profiles in their halos and there is a higher incidence of misalignments, indicating triaxiality. Dynamical models have shown a range of slopes for the total mass profiles, and that the inner dark matter densities in ellipticals are higher than in spiral galaxies, indicating earlier assembly redshifts. Analysis of the hot X-ray emitting gas in X-ray bright ellipticals and comparison with dynamical mass determinations indicates that non-thermal components to the pressure may be important in the inner ~10 kpc, and that the properties of these systems are closely related to their group environments. First results on the outer halo stellar population properties do not yet give a clear picture. In the halo of one bright galaxy, lower [alpha/Fe] abundances indicate longer star formation histories pointing towards late accretion of the halo. This is consistent with independent evidence for on-going accretion, and suggests a connection to the observed size evolution of elliptical galaxies with redshift.Comment: 8 pages.     Inflation from Warped Space

    A long period of inflation can be triggered when the inflaton is held up on the top of a steep potential by the infrared end of a warped space. We first study the field theory description of such a model. We then embed it in the flux stabilized string compactification. Some special effects in the throat reheating process by relativistic branes are discussed. We put all these ingredients into a multi-throat brane inflationary scenario.     Spontaneous Creation of Inflationary Universes and the Cosmic Landscape

    We study some gravitational instanton solutions that offer a natural realization of the spontaneous creation of inflationary universes in the brane world context in string theory. Decoherence due to couplings of higher (perturbative) modes of the metric as well as matter fields modifies the Hartle-Hawking wavefunction for de Sitter space. Generalizing this new wavefunction to be used in string theory, we propose a principle in string theory that hopefully will lead us to the particular vacuum we live in, thus avoiding the anthropic principle. As an illustration of this idea, we give a phenomenological analysis of the probability of quantum tunneling to various stringy vacua. We find that the preferred tunneling is to an inflationary universe (like our early universe), not to a universe with a very small cosmological constant (i.e., like today's universe) and not to a 10-dimensional uncompactified de Sitter universe. Such preferred solutions are interesting as they offer a cosmological mechanism for the stabilization of extra dimensions during the inflationary epoch.Comment: 52 pages, 7 figures, 1 table.     Cosmic F- and D-strings

    Macroscopic fundamental and Dirichlet strings have several potential instabilities: breakage, tachyon decays, and confinement by axion domain walls. We investigate the conditions under which metastable strings can exist, and we find that such strings are present in many models. There are various possibilities, the most notable being a network of (p,q) strings.     Pretreatment dietary intake is associated with tumor suppressor DNA methylation in head and neck squamous cell carcinomas

    Diet is associated with cancer prognosis, including head and neck cancer (HNC), and has been hypothesized to influence epigenetic state by determining the availability of functional groups involved in the modification of DNA and histone proteins. The goal of this study was to describe the association between pretreatment diet and HNC tumor DNA methylation. Information on usual pretreatment food and nutrient intake was estimated via food frequency questionnaire (FFQ) on 49 HNC cases. Tumor DNA methylation patterns were assessed using the Illumina Goldengate Methylation Cancer Panel. First, a methylation score, the sum of individual hypermethylated tumor suppressor associated CpG sites, was calculated and associated with dietary intake of micronutrients involved in one-carbon metabolism and antioxidant activity, and food groups abundant in these nutrients. Second, gene specific analyses using linear modeling with empirical Bayesian variance estimation were conducted to identify if methylation at individual CpG sites was associated with diet. All models were controlled for age, sex, smoking, alcohol and HPV status. Individuals reporting in the highest quartile of folate, vitamin B12 and vitamin A intake, compared with those in the lowest quartile, showed significantly less tumor suppressor gene methylation, as did patients reporting the highest cruciferous vegetable intake. Gene specific analyses identified differential associations between DNA methylation and vitamin B12 and vitamin A intake when stratifying by HPV status. These preliminary results suggest that intake of folate, vitamin A and vitamin B12 may be associated with the tumor DNA methylation profile in HNC and enhance tumor suppression

    Brane Inflation, Solitons and Cosmological Solutions: I

    In this paper we study various cosmological solutions for a D3/D7 system directly from M-theory with fluxes and M2-branes. In M-theory, these solutions exist only if we incorporate higher derivative corrections from the curvatures as well as G-fluxes. We take these corrections into account and study a number of toy cosmologies, including one with a novel background for the D3/D7 system whose supergravity solution can be completely determined. This new background preserves all the good properties of the original model and opens up avenues to investigate cosmological effects from wrapped branes and brane-antibrane annihilation, to name a few. We also discuss in some detail semilocal defects with higher global symmetries, for example exceptional ones, that could occur in a slightly different regime of our D3/D7 model. We show that the D3/D7 system does have the required ingredients to realise these configurations as non-topological solitons of the theory.     Oralism: a sign of the times? The contest for deaf communication in education provision in late nineteenth-century Scotland

    Disability history is a diverse field. In focussing upon children within deaf education in late nineteenth-century Scotland, this essay reflects some of that diversity. In 1880, the International Congress on the Education of the Deaf in Milan stipulated that speech should have ‘preference’ over signs in the education of deaf children. The mode of achieving this, however, effectively banned sign language. Endeavours to teach deaf children to articulate were not new, but this decision placed pressures on deaf institutions to favour the oral system of deaf communication over other methods. In Scotland efforts were made to adopt oralism, and yet educators were faced with the reality that this was not good educational practice for most pupils. This article will consider responses of Scottish educators of deaf children from the 1870s until the beginning of the twentieth century
