    Cytotoxicity and antitumoral activity of dichloromethane extract and its fractions from Pothomorphe umbellata

    The cytotoxicity of the dichloromethane crude extract (DCE), obtained from the aerial parts of Pothomorphe umbellata (L.) Miq (Piperaceae), was evaluated against nine human cancer cell lines (MCF-7, NCI-ADR/RES, OVCAR-3, PC-3, HT-29, NCI-H460, 786-O, UACC-62, K-562). The DCE presented antiproliferative activity with good potency against all cell lines at low concentrations (between 4.0 and 9.5 µg/mL) and with selectivity (1.55 µg/mL) for the leukemia cell line (K-652). DCE (100, 200, 300 and 400 mg/kg, ip) was also evaluated in the Ehrlich ascites tumor model. Both the survival number and the life span of the animals that died increased by at least 45 and 50%, respectively (8 animals per group), demonstrating P. umbellata extract potential anticancer activity. The results of the in vivo antitumor activity prompted the fractionation of the crude extract. The crude extract was submitted to dry column chromatography with dichloromethane-methanol (99:1). The column effluent fractions were extracted with methanol, dried under vacuum yielding fractions FR1 (less polar), FR2 (medium polarity), and FR3 (polar), which were analyzed for their growth inhibition or cytotoxic properties by a 48-h sulforhodamine B cell viability assay by measuring the total protein content. FR1 demonstrated high potency and cytotoxicity, a result compatible with the high toxicity of oxalic acid; FR2, containing 4-nerolidylcathecol, presented the lowest cytotoxic activity compared to the other two fractions but with selectivity for prostate cancer cell line; FR3, containing a mixture of steroids described in the literature as possessing various biological activities, also presented potent anticancer in vitro activity. These results suggest that P. umbellata DCE in vivo antitumor activity may be a consequence of the activity of different active principles.41141

    Correlação entre caracteres agronômicos, fisiológicos e fitoquímicos em variedades de Artemisia annua L

    The aim of this study was to estimate the correlations among agronomic, physiological and phytochemical traits of two varieties of Artemisia annua and to identify correlations that can be used in selection and breeding processes for this species. The design was completely randomized and the treatments were for Artemisia annua varieties 2/39x5x3M and 2/39x1V, which were subjected to agronomic, physiological and phytochemical evaluations. The relationship among the studied traits was estimated with Pearson's correlation coefficient, and the significance level of correlations was established by the t-test using Genes software. A significant positive correlation was found between the traits canopy volume and essential oil yield for 2/39x5x3M variety, showing that the selection of plants with a larger canopy volume would facilitate indirect selection of the trait essential oil yield. This correlation was not found for the 2/39x1V variety. In both varieties, there was a significant negative correlation between canopy volume and artemisinin content, which suggested that the selection of plants with a large canopy volume and a large number of branches should be avoided if the goal is to increase artemisinin content.O objetivo deste trabalho foi estimar as correlações existentes entre caracteres agronômicos, fisiológicos e fitoquímicos em duas variedades de Artemisia annua e identificar correlações que possam ser utilizadas em processos de seleção e melhoramento da espécie. O delineamento foi inteiramente casualizado e os tratamentos foram às variedades 2/39x5x3M e 2/39x1V de Artemisia annua, submetidas a avaliações agronômicas, fisiológicas e fitoquímicas. A relação existente entre os caracteres estudados foi estimada através do coeficiente de correlação de Pearson e o nível de significância das correlações pelo teste t, realizadas no programa Genes. Encontrou-se correlação positiva e significativa entre os caracteres volume de dossel e rendimento de óleo essencial na variedade 2/39x5x3M, evidenciando que a seleção de plantas com maior volume de dossel possibilitaria a seleção indireta para o cárater rendimento de óleo essencial. Esta correlação não foi encontrada na variedade 2/39x1V.     Synthesis, in vitro antiproliferative and anti-mycobacterium tuberculosis activities of novel β-carboline derivatives

    A series of β-carboline derivatives with amino or guanidinium were synthesized and evaluated in vitro against anti-Mycobacterium tuberculosis and for antiproliferative activities against nine human cancer cell lines. The compounds 1-(4-hydroxyphenyl)-3-carboxamide(ethylamine) β-carboline (24.9 μg mL-1) and 1-(4-methoxyphenyl)-3-carboxamide(ethylamine) β-carboline (26.9 μg mL-1) were the most active against M. Tuberculosis (MTB). Compounds 1-(4-hydroxyphenyl)-3-carboxamide(ethylamine) β-carboline and 1-(4-methoxyphenyl)-3-carboxamide(propylamine) β-carboline, which had the same substituted groups, inhibited the growth of all human tumor cell lines with growth inhibitory activity (GI50) values from 1.37 to 9.20 mmol L-1. Also in this series, compounds 1-(4-hydroxyphenyl)-3-carboxamide(propylamine) β-carboline and 1-(3-nitrophenyl)-3-carboxamide(propylamine) β-carboline demonstrated significant activity against NCI/ADR cells. Among compounds with a terminal guanidine group, compounds 1-(4-hydroxyphenyl)-3-carboxamide(ethyl)guanidine β-carboline (27.8 μg mL-1) and 1-(3-nitrophenyl)-3-carboxamide(ethyl) guanidine β-carboline (37.4 μg mL-1) demonstrated the greatest activity against MTB. Additionally, compounds 1-(4-methoxyphenyl)-3-carboxamide(ethyl)guanidine β-carboline (GI50 = 0.45 mmol L-1) effectively inhibited growth and was highly selective against NCI/ADR.     Docosahexaenoic acid is an essential polyunsaturated fatty acid with important metabolic activities. Its conjugated double bonds make it susceptible to decomposition. Its stability may be improved through fatty acid entrapment with a spray-drying technique; however, the many parameters involved in this technique must be considered to avoid affecting the final product quality. Therefore, this study aimed to evaluate the entrapment conditions and yields of fish oil enriched with docosahexaenoic acid ethyl ester. Microcapsules were obtained from Acacia gum using a spray-drying technique. The experimental samples were analyzed by chromatography and delineated by Statistica software, which found the following optimum entrapment conditions: an inlet temperature of 188 °C; 30% core material; an N 2 flow rate of 55 mm; and a pump flow rate of 12.5 mL/minute. These conditions provided a 66% yield of docosahexaenoic acid ethyl ester in the oil, corresponding to 19.8% of entrapped docosahexaenoic acid ethyl ester (w/w).     In Vitro Activity Of Artemisia Annua L (asteraceae) Extracts Against Rhipicephalus (boophilus) Microplus [atividade In Vitro De Extratos De Artemisia Annua L (asteraceae) Sobre Rhipicephalus (boophilus) Microplus]

    The activity of plant extracts on parasites may indicate groups of substances that are potentially useful for controlling Rhipicephalus (Boophilus) microplus. The aim of the present study was to investigate the in vitro action of Artemisia annua extracts on this tick. The concentrations of the sesquiterpene lactones artemisinin and deoxyartemisinin present in plant extracts were quantified via high-performance liquid chromatography. Four extracts produced from the concentrated crude extract (CCE) were evaluated on larvae using the impregnated paper method, with readings after 24 hours of incubation. The engorged females were immersed in the CCE and in its four derived extracts for five minutes, with incubation for subsequent analysis of biological parameters. The extracts were not effective on the larvae at the concentrations tested (3.1 to 50 mg.mL-1). The CCE showed greater efficacy on engorged females (EC50 of 130.6 mg.mL-1 and EC90 of 302.9 mg.mL-1) than did the derived extracts. These results tend to confirm that the action of artemisinin on engorged females of R. (B.) microplus is conditional to their blood intake. In this case, in vitro methods would be inadequate for effective evaluation of the action of A. annua on R.     Fano resonance in electronic transport through a quantum wire with a side-coupled quantum dot: X-boson treatment

    The transport through a quantum wire with a side coupled quantum dot is studied. We use the X-boson treatment for the Anderson single impurity model in the limit of U=U=\infty . The conductance presents a minimum for values of T=0 in the crossover from mixed-valence to Kondo regime due to a destructive interference between the ballistic channel associated with the quantum wire and the quantum dot channel. We obtain the experimentally studied Fano behavior of the resonance. The conductance as a function of temperature exhibits a logarithmic and universal behavior, that agrees with recent experimental results.Comment: 6 pages, 10 eps figs., revtex

    Synthesis And Evaluation Of New β-carboline-3-(4-benzylidene)- 4h-oxazol-5-one Derivatives As Antitumor Agents

    In the present work, we report the synthesis and in vitro anticancer and antimicrobial activity evaluation of a new series of 1-substituted-β- carboline derivatives bearing a 4-benzylidene-4H-oxazol-5-one unity at C-3. The compound 2-[1-(4- methoxyphenyl)-9H-β-carbolin-3-yl]-4-(benzylidene)-4H- oxazol-5-one (11) was the most active derivative, exhibiting a potent cytotoxic activity against glioma (U251), prostate (PC-3) and ovarian (OVCAR-03) cancer cell lines with IC 50 values of 0.48, 1.50 and 1.07 μM, respectively.
