    Biodistribution PET/CT study of hemoglobin-DFO-89Zr complex in healthy and lung tumor-bearing mice

    Proteins, as a major component of organisms, are considered the preferred biomaterials for drug delivery vehicles. Hemoglobin (Hb) has been recently rediscovered as a potential drug carrier, but its use for biomedical applications still lacks extensive investigation. To further explore the possibility of utilizing Hb as a potential tumor targeting drug carrier, we examined and compared the biodistribution of Hb in healthy and lung tumor-bearing mice, using for the first time89 Zr labelled Hb in a positron emission tomography (PET) measurement. Hb displays a very high conjugation yield in its fast and selective reaction with the maleimide-deferoxamine (DFO) bifunctional chelator. The high-resolution X-ray structure of the Hb-DFO complex demonstrated that cysteine β93 is the sole attachment moiety to the αβ-protomer of Hb. The Hb-DFO complex shows quantitative uptake of89 Zr in solution as determined by radiochromatography. Injection of 0.03 mg of Hb-DFO-89 Zr complex in healthy mice indicates very high radioactivity in liver, followed by spleen and lungs, whereas a threefold increased dosage results in intensification of PET signal in kidneys and decreased signal in liver and spleen. No difference in biodistribution pattern is observed between naïve and tumor-bearing mice. Interestingly, the liver Hb uptake did not decrease upon clodronate-mediated macrophage depletion, indicating that other immune cells contribute to Hb clearance. This finding is of particular interest for rapidly developing clinical immunology and projects aiming to target, label or specifically deliver agents to immune cells

    MicroRNA expression patterns in canine mammary cancer show significant differences between metastatic and non-metastatic tumours

    Background: MicroRNAs may act as oncogenes or tumour suppressor genes, which make these small molecules potential diagnostic/prognostic factors and targets for anticancer therapies. Several common oncogenic microRNAs have been found for canine mammary cancer and human breast cancer. On account of this, large-scale profiling of microRNA expression in canine mammary cancer seems to be important for both dogs and humans. Methods: Expression profiles of 317 microRNAs in 146 canine mammary tumours of different histological type, malignancy grade and clinical history (presence/absence of metastases) and in 25 control samples were evaluated. The profiling was performed using microarrays. Significance Analysis of Microarrays test was applied in the analysis of microarray data (both unsupervised and supervised data analyses were performed). Validation of the obtained results was performed using real-time qPCR. Subsequently, predicted targets for the microRNAs were searched for in miRBase. Results: Results of the unsupervised analysis indicate that the primary factor separating the samples is the metastasis status. Predicted targets for microRNAs differentially expressed in the metastatic vs. non-metastatic group are mostly engaged in cell cycle regulation, cell differentiation and DNA-damage repair. On the other hand, the supervised analysis reveals clusters of differentially expressed microRNAs unique for the tumour type, malignancy grade and metastasis factor. Conclusions: The most significant difference in microRNA expression was observed between the metastatic and non-metastatic group, which suggests a more important role of microRNAs in the metastasis process than in the malignant transformation. Moreover, the differentially expressed microRNAs constitute potential metastasis markers. However, validation of cfa-miR-144, cfa-miR-32 and cfa-miR-374a levels in blood samples did not follow changes observed in the non-metastatic and metastatic tumours. © 2017 The Author(s)

    Effect of the COVID-19 pandemic on surgery for indeterminate thyroid nodules (THYCOVID): a retrospective, international, multicentre, cross-sectional study

    Background Since its outbreak in early 2020, the COVID-19 pandemic has diverted resources from non-urgent and elective procedures, leading to diagnosis and treatment delays, with an increased number of neoplasms at advanced stages worldwide. The aims of this study were to quantify the reduction in surgical activity for indeterminate thyroid nodules during the COVID-19 pandemic; and to evaluate whether delays in surgery led to an increased occurrence of aggressive tumours.Methods In this retrospective, international, cross-sectional study, centres were invited to participate in June 22, 2022; each centre joining the study was asked to provide data from medical records on all surgical thyroidectomies consecutively performed from Jan 1, 2019, to Dec 31, 2021. Patients with indeterminate thyroid nodules were divided into three groups according to when they underwent surgery: from Jan 1, 2019, to Feb 29, 2020 (global prepandemic phase), from March 1, 2020, to May 31, 2021 (pandemic escalation phase), and from June 1 to Dec 31, 2021 (pandemic decrease phase). The main outcomes were, for each phase, the number of surgeries for indeterminate thyroid nodules, and in patients with a postoperative diagnosis of thyroid cancers, the occurrence of tumours larger than 10 mm, extrathyroidal extension, lymph node metastases, vascular invasion, distant metastases, and tumours at high risk of structural disease recurrence. Univariate analysis was used to compare the probability of aggressive thyroid features between the first and third study phases. The study was registered on ClinicalTrials.gov, NCT05178186.Findings Data from 157 centres (n=49 countries) on 87 467 patients who underwent surgery for benign and malignant thyroid disease were collected, of whom 22 974 patients (18 052 [78 center dot 6%] female patients and 4922 [21 center dot 4%] male patients) received surgery for indeterminate thyroid nodules. We observed a significant reduction in surgery for indeterminate thyroid nodules during the pandemic escalation phase (median monthly surgeries per centre, 1 center dot 4 [IQR 0 center dot 6-3 center dot 4]) compared with the prepandemic phase (2 center dot 0 [0 center dot 9-3 center dot 7]; p<0 center dot 0001) and pandemic decrease phase (2 center dot 3 [1 center dot 0-5 center dot 0]; p<0 center dot 0001). Compared with the prepandemic phase, in the pandemic decrease phase we observed an increased occurrence of thyroid tumours larger than 10 mm (2554 [69 center dot 0%] of 3704 vs 1515 [71 center dot 5%] of 2119; OR 1 center dot 1 [95% CI 1 center dot 0-1 center dot 3]; p=0 center dot 042), lymph node metastases (343 [9 center dot 3%] vs 264 [12 center dot 5%]; OR 1 center dot 4 [1 center dot 2-1 center dot 7]; p=0 center dot 0001), and tumours at high risk of structural disease recurrence (203 [5 center dot 7%] of 3584 vs 155 [7 center dot 7%] of 2006; OR 1 center dot 4 [1 center dot 1-1 center dot 7]; p=0 center dot 0039).Interpretation Our study suggests that the reduction in surgical activity for indeterminate thyroid nodules during the COVID-19 pandemic period could have led to an increased occurrence of aggressive thyroid tumours. However, other compelling hypotheses, including increased selection of patients with aggressive malignancies during this period, should be considered. We suggest that surgery for indeterminate thyroid nodules should no longer be postponed even in future instances of pandemic escalation.Funding None.Copyright (c) 2023 Published by Elsevier Ltd.     Obiektem badań były gleby Gospodarstwa Rolnego Karolew, które od ponad 10 lat uprawiane są w płodozmianie w systemie uprawy uproszczonej oraz przyległe do nich gleby gospodarstw indywidualnych, na których jest również stosowany płodozmian ale system uprawy tradycyjnej. Zasadniczym celem prowadzonych badań była wstępna ocena podstawowych właściwości fizycznych i parametrów fizykomechanicznych gleb i agregatów glebowych w wymienionych dwóch systemach uprawy. Badania przeprowadzono na modelowanych agregatach glebowych, zagęszczanych i formowanych w ściśle kontrolowanych warunkach wilgotności, temperatury i zagęszczenia. Przeprowadzone badania wykazały różnice we właściwościach fizykomechanicznych struktury agregatowej tych samych gleb w różnych systemach uprawy. Agregaty modelowane z materiału glebowego, który podlegał uproszczeniom uprawowym, cechowały się nieco mniejszą wodoodpornością, ale lepszą agregacją wtórną zarówno po statycznym, jak i dynamicznym działaniu wody. Zróżnicowane były również pojemności wodne agregatów; agregaty w systemie uprawy klasycznej wykazywały mniejszą o około 1-4% kapilarną pojemność wodną, pomimo znacznie dłuższego czasu podsiąku kapilarnego. Zdolność do pęcznienia agregatów w glebie będącej w uproszczeniu uprawowym była większa o około 2-9% od agregatów w systemie uprawy tradycyjnej.The object of the research were soils in the Karolew Farm, which have been cultivated in crop rotation and tillage simplification system for over 10 years, as well as the neighbouring soils in individual farms, also cultivated in crop rotation, yet traditional tillage system. The main aim of the research was the initial assessment of physical properties and physical and mechanical parameters of soils and soil aggregates in the two tillage systems. The study was conducted on moulded soil aggregates, which had been thickened and formed in strictly controlled conditions of humidity, temperature and compaction. The study proved differences in physical and mechanical properties in soil aggregates of the same soil but under different tillage systems. Aggregates moulded from the soil material which had undergone tillage simplifications showed slightly lower water resistance but better secondary aggregation after both static and dynamic water actions. There were also differences in the water capacity of aggregates: the capillary water capacity of aggregates under a traditional tillage system was lower by about 1-4 % despite longer capillary rise. The ability to swelling of aggregates under simplified tillage system was higher by about 2-9% than the one of aggregates under a traditional tillage system

    Differentially expressed proteins in the blood serum of piglets in response to a diet supplemented with inulin

    In the present study we introduced a two-dimensional electrophoresis and matrix-assisted laser desorption/ionisation time of flight mass spectrometry-based proteomic workflow to identify proteins that show altered expression as a result of the addition of 2% of water extract of inulin-type fructans to the diet of growing piglets. This analysis allowed us to detect an average of 240 spots per gel with a mass range from 10 to 250 kDa and a pH ranging from 3 to 10. Twenty protein spots were found to show statistically significant differences in their expression. Of these, 7 protein spots were up-regulated, whereas 13 showed down-regulation in response to the experimental diet. In total, 13 spots were identified, representing 8 distinct gene products. The experimental diet caused a significant change in proteins directly or indirectly involved in hemostasis and the innate immune response. Increased levels of fibrinogen along with decreased plasminogen expression may indicate that a fructan-rich diet favours the deposits of fibrin and promotes blood clotting. We also found increased expression of vitronectin and the alpha subunit of the complement component C8 which may protect the host organism against excessive cytolitic activity of the activated complement. The piglets from the experimental group had slightly increased values of IgG and IgA, whereas the IgM level tended to be decreased. The fructan-rich diet did not have any influence on plasma total cholesterol, HDL and LDL cholesterol and triglyceride levels

    Identification of TRPM6 and TRPM7 expression changes in response to a diet supplemented with inulin in porcine kidney

    Magnesium is the fourth most abundant mineral element in vertebrates and the second most common intracellular cation. Recently identified Mg<sup>2+</sup>-specific channels – TRPM6 and TRPM7 – have been shown to be essential for whole-body and cellular Mg<sup>2+</sup> homeostasis. The aim of the study was to determine the effect of inulin on the expression of TRPM6 and TRPM7 in the renal cortex and medulla of growing pigs. The study was carried out on 16 Danbred  ×  Duroc castrated male piglets fed a cereal-based diet without inulin or with 2 % addition of inulin from chicory root from the 10th day of life. In pigs fed a diet with inulin, TRPM6 expression was greater in both the renal cortex and medulla compared to the control group. The expression of TRPM7 in both the renal cortex and medulla in the control group and in piglets fed a diet enriched with inulin was relatively stable. To our knowledge, this is the first study aimed at the identification of TRPM6 and TRPM7 in the kidneys of pig. It is proposed that inulin addition to fodder resulted not only in a magnesium absorption increase, but also, due to prolonged low plasma Mg concentration of examined piglets, renal magnesium retention. Therefore, higher magnesium reabsorption via increased TRPM6 expression in the kidney was probably observed in order to supplement deficiencies of this element. Diet-unresponsive expression of TRPM7 supports the concept that this channel is not involved in the extracellular magnesium homeostasis
